Development of tolerance to chronic barbital treatment in the cerebellar cyclic guanosine monophosphate system and its response to subsequent barbital abstinence.

Female Sprague-Dawley rats were placed on a 6-week barbital feeding regimen, previously documented to result in drug tolerance and dependence. Groups of animals were sacrificed on the 2nd and 5th day of each week, up to week 5, or at various time points following drug withdrawal. Cerebellae and cerebral cortices were collected for cyclic GMP (cGMP) measurements. Initial suppressions of cGMP were seen in both the cerebellum and cerebral cortex during early weeks of the feeding regimen. These gradually returned essentially to control levels by the end of 5 weeks, a finding consistent with the de…

A barbital derivative as an atypical antidepressant drug in mice.

An isopropyl derivative of barbital (5,5-diethyl-2-(isopropyloxy)pyrimidine-4,6-dione, O2IB) was administered intraperitoneally 30 min before tests in mice. Former experimental investigations have shown that O2IB has an antidepressant psychopharmacological spectrum. It increases toxicity of yohimbine in mice at 175 mg/kg, antagonises from 50 mg/kg on hypothermia induced by a high dose of apomorphine and is active on the behavioural despair test at 125 mg/kg. These effects are those observed with classical antidepressants. Since phenytoin has an antidepressant profile in mice, carbamazepine is …

Demonstration of neurochemical tolerance following chronic barbital treatment.

Control rats (C) and rats rendered barbital dependent (BD) by the consumption of barbital chronically in their food for 6 weeks were treated with saline or 1 of 3 acute dosages of barbital 16 minutes before killing. Cyclic guanosine monophosphate (cGMP) levels and circulating levels of barbital were measured. Significantly higher circulating levels of barbital were required to produce a comparable suppression of cerebellar cGMP in the BD as opposed to the C rats. These data provide evidence for the development of tolerance to barbital in the cerebellar cGMP system during chronic barbital treat…

Tolerance to ethanol and cross-tolerance to pentobarbital and barbital.

A chronic regimen of ethanol by intubation, which produced clear tolerance to ethanol-induced hypothermia, ataxia and narcosis, produced only a marginal degree of cross-tolerance to these effects of pentobarbital. The lack of appreciable cross-tolerance to pentobarbital-induced hypothermia and ataxia was also observed over a wide range of test doses. However, cross-tolerance to barbital was observed after chronic treatment with ethanol. Increased rate of drug biotransformation did not contribute significantly to the observed tolerance and cross-tolerance. The difference in the extent of cross-…

[Sex differences in the physical dependence on barbital in rats].

Sex differences in physical dependence on barbital (BAR), which is not readily metabolized in rats, were studied by the drug-admixed food method. The concentration of BAR in the food was gradually increased from 1 and 2 up to 6 and 8 mg/g food concurrently available to each rat over a period of 36 days in both male and female Sprague-Dawley rats. Sedation and muscle relaxation were observed in males at the 4 and 6 mg/g condition and to a greater degree at 6 and 8 mg/g of food. This effect was seen only at the highest drug concentration, 6 and 8 mg/g food, in females. Physical dependence was as…

Effect of p-chlorophenylalanine on the acquisition of tolerance to barbital.

The effect of p-chlorophenylalanine (p-CPA) pretreatment on barbital tolerance in the rat as measured by motor impairment on the moving belt test was examined in two separate studies. The first used a 2 X 2 design with doses of p-CPA (125 mg/kg) or water, and sodium barbital (300 mg/kg) or water. The treatments continued for 28 days with tests every 7 days. The p-CPA dose used was previously shown to produce and maintain greater than 95% depletion of brain serotonin (5-HT). Tolerance developed to the test doses, and even greater tolerance to the chronic treatment doses. In both cases the p-CPA…

Specific action of tranylcypromine to precipitate barbital withdrawal convulsions.

This research was designed to determine whether the convulsion-eliciting action of tranylcypromine (TCP) during bartibal (B) withdrawal was specific to physical dependence on B, and to compare the findings with the action of pentetrazol (PTZ). Challenge with 15-20 mg/kg IP TCP at 48 or B withdrawal resulted in the elicitation of clonic-tonic convulsion (CTC) in all rats (n = 6) within 10 min. Another challenge with 5-20 mg/kg TCP led to the dose-related precipitation and intensification of CTC. The CTC-inducing action of TCP was relatively reduced as the B withdrawal signs were gradually mitig

Increased susceptibility of audiogenic rats to barbital withdrawal convulsions.

Non-responsive progeny from Sprague-Dawley derived rats genetically susceptible to sound-induced (audiogenic) convulsions (AGS-negative) and non-responsive progeny from Sprague-Dawley derived rats not gentically susceptible to audiogenic convulsions (SD-negative) were subjected to a seven-day treatment regimen of sodium barbital. 125 mg/kg, every 12 hours. This represents a lower dose and shorter treatment period than that normally used in this laboratory to induce barbiturate dependence in rats. Animals were subjected to a 115 dB sound stimulus 38 hours following the last dose of sodium barbi…

Pharmacological characterization of alcohol and barbital physical dependence in mice.

To confirm our previous finding that the underlying mechanism for the development of physical dependence on alcohol and barbital are fundamentally different from each other, further studies on the effect of drugs that modify neurotransmission on the alcohol and barbital withdrawal signs were carried out in alcohol and barbital dependent mice. Phentolamine, propranolol and methysergide exacerbated the alcohol withdrawal signs but suppressed the barbital withdrawal signs indicating the qualitative difference of the underlying mechanism. Some quantitative differences were also found in the effect…

Characteristic biotransformation of aminopyrine in rat when administered with barbital.

After the administration of aminopyrine with or without barbital to rats, aminopyrine and its main metabolites were detected in plasma and the brain by means of gas chromatography-mass spectrometry. It was clarified that a marked increase of 4-monomethylaminoantipyrine was observed in the case of coadministration of aminopyrine and barbital, while the plasma level of aminopyrine decreased significantly compared with single administration.

Atropine in the abstinence after chronic barbital treatment in the rat.

Since cholinergic mechanisms seem to be involved in the changes induced by chronic barbital treatments in male rats, various treatments with atropine were given during the abstinence after 32--33 weeks of exposure to barbital (200 mg/kg/day). The effects of the atropine treatment were recorded as a tolerance towards a hexobarbital anaesthesia threshold. In Experiment 1, 1.5 mg/kg/day of atropine was given on Days 23--29 after the end of the barbital treatment. Two weeks after the end of the atropine treatment a significant tolerance (+ 16%, Fig. 1A) was seen in barbital-treated animals given t…
