Protein C activator from Agkistrodon halys halys venom has been isolated and purified by ion-exchange and gel-filtration chromatography. The purified enzyme consists of the single peptide-chain with molecular weight of 34 kDa. Its pH-optimum was in 7.5-8.0 range. The enzyme was inhibited by DFP, benzamidine, PMSF, EGTA. Protein C activator was as effective as "Protac" (Pentapharm AG, Switzerland).

A mild, metal-free annulative coupling of azirine-2H-carbaldehydes with benzamidine hydrochlorides is reported for the efficient synthesis of substituted 5-amino pyrimidines. This transformation proceeds via a cascade of condensation, intramolecular cyclisation, strain-release-driven azirine ring-opening, and aromatisation. This step-economical strategy highlights the untapped potential of strained azirines as versatile synthons for the synthesis of biologically potent 2,4-arylpyrimidin-5-amines.

A serine protease from Bothrops alternatus snake venom was isolated using DEAE-Sephacel, Sephadex G-75 and Benzamidine-Sepharose column chromatography. Reaction of the secondary alkynyl(Mes*)PH phosphane 2 with (Fmes)BH2 ⋅SMe2 gives the geminal PH/BH frustrated Lewis pair (FLP) 3. The PH and the BH functions are jointly used in the reduction of carbon monoxide under mild reaction conditions to give the [P]-CH2 -O-[B] product. A subsequent cycloaddition sequence results in the liberation of formaldehyde. The FLP 3 reacts with benzonitrile to give a P-benzamidine, and it couples two isonitriles at the FLP framework.

Renin is produced from an inactive precursor, prorenin, through endoproteolytic cleavage at paired basic amino-acid residues. Using (35S)methionine-labeled prorenin, that was synthesized with Xenopus oocyte expression system, as a substrate, we have determined the tissue distribution and the nature of prorenin-converting activity in mouse. The highest activity was found in the submandibular gland of male ICR mouse. The activity of the enzyme seemed to be parallel to that of renin. This enzyme activity, with an optimal pH 8.0-8.5, was inhibited by leupeptin, antipain and benzamidine.

A peripheral membrane protease was purified from mitochondria of rat submaxillary gland. On non-denaturing PAGE the purified enzyme showed a single protein band with the enzyme activity. BACKGROUND: Midgut enzymatic activity is one of the obstacles that Leishmania must surpass to succeed in establishing infection. Trypsins are abundant digestive enzymes in most insects. We have previously described two trypsin cDNAs of L. longipalpis: one (Lltryp1) with a bloodmeal induced transcription pattern, the other (Lltryp2) with a constitutive transcription pattern.
