Induction of chromosome changes by metal compounds in cultured CHO cells.

The effect of 4 metal salts on the induction of chromosome aberrations and sister chromatid exchange (SCE) in cultured Chinese hamster ovary (CHO) cells was investigated. It was observed that CdCl2, NiCl2, CrO3 and HgCl2 were effective in causing various types of chromosome aberrations. The percentages of abnormalities increased with increasing doses of the metals used. These metal compounds also raised the SCE rates compared to 5-bromo-2'-deoxyuridine controls. The importance of rapid screening of potentially hazardous metals is described.

Co-expression enhances cloning efficiency and protein production in CHO cells.

Chinese hamster ovary (CHO) cell lines are widely used in the biopharmaceutical industry, particularly for the production of monoclonal antibodies and recombinant protein drugs. Recombinant cell lines are typically derived from single-cell clones to ensure product consistency and stability. However, the difficult-to-express recombinant proteins may impair single-cell proliferation, thereby significantly reducing cloning efficiency. In this study, co-expression of insulin-like growth factor-1 (IGF-1) was used to improve the single-cell cloning efficiency. Co-expression of IGF-1 significantly im

Identification of ICR170-induced XPD mutations in UV-sensitive CHO cells.

We highlight selected contributions of Dr. Richard Setlow that contributed to our earlier understanding of excision repair processes and set the stage for dissecting nucleotide excision repair (NER) in mammalian cells through molecular genetics. More than 20 years ago, large-scale screens for UV-sensitive mutants of hamster CHO cells isolated approximately 200 mutants, many of which were assigned to the XPD/ERCC2 complementation group, but the nature of the mutations was not determined. The XPD protein performs not only an essential viability function as a structural component of transcription

Mechanisms for recombination between stably integrated vector sequences in CHO cells.

Possible mechanisms for homologous recombination in CHO cells have been investigated using a stably integrated vector, pIII-14gpt. The vector contains 2 inactive neo gene fragments in tandem arrangement. Functional neo gene activity can be restored by recombination between homologous regions in the 2 fragments. Cells in which this event has taken place become resistant to the antibiotic G418. Possible mechanisms for neo gene reactivation in this system are unequal exchange between chromatids, intrachromatidal deletion and gene conversion. DNA from a total of 74 G418-resistant cell clones have

Pioneer Factor FOXA1 Boosts CHO Cell Productivity.

Industrial production of biologics commonly involves the integration of transgenes into the genomes of host cells, such as Chinese hamster ovary (CHO) cells. A major determinant of productivity is the epigenetic control of the transgene promoter, accessibility of which can decrease during production due to the spread of heterochromatin. Pioneer factors such as forkhead box A1 (FOXA1) can bind heterochromatin, increase its accessibility and facilitate transcription of target genes. We show that FOXA1 can bind the EF1α and CMV promoters, which are widely used in industry. Overexpressing FOXA1 in

Nicotinamide Reverses the Warburg Effect in CHO Cell Culture.

The Warburg effect, the preferential conversion of glucose-derived pyruvat to lactate despite available oxygen, is a key feature of Chinese hamster ovary (CHO) cell culture. Lactate accumulation in recombinant protein-producing cell culture is an inefficient usage of glucose, as well as being deleterious to cells. Lactate accumulation lowers culture pH, requiring base addition to maintain bioreactor pH setpoint, which subsequently leads to hyperosmolarity, adversely impacting cell growth, productivity and product quality. A key driver for the Warburg effect, and hence lactate accumulation, is

Application of aurintricarboxylic acid to boost CHO cell performance.

PURPOSE: Currently, a robust process using healthy Chinese hamster ovary (CHO) cells, the dominating host for biopharmaceutical protein therapeutics, is crucial for high productivity and successful scaling-up. However, issues which are originated from cell performance and detrimental to protein products such as dramatic viability drop and accumulation of toxic metabolites still exist. Aurintricarboxylic acid (ATA), reported as an antioxidant chelator to prevent apoptosis and boost cell growth, was applied as an additive in this study to address the above problems. METHODS: Two CHO cells were

Evaluation of gene editing in CHO cells using the Cas-CLOVER system.

Recent advances in gene editing technologies have transformed the genetic engineering of Chinese hamster ovary (CHO) hosts, enabling the development of cell lines with improved stability and productivity. In this study, we employed the programmable nuclease (PN) Cas-CLOVER to precisely target the Glutamine synthetase (GS) locus in CHO cells. A total of 30 unique serum-free, suspension-adapted CHO-K1 candidate host cell lines were subjected to Cas-CLOVER-mediated gene editing, generating over one hundred potential GS knockout (GSKO) clones. A subset of the GSKO clones was subsequently validated

Sequence optimization targeting mRNA stability enhances monoclonal antibody titers in CHO cells.

This study presents a DNA sequence optimization approach that integrates mRNA stability as a tunable design parameter to enhance monoclonal antibody expression in Chinese hamster ovary (CHO) cells. A comprehensive combinatorial library of synonymous coding-sequence variants of an IgG1 light chain was integrated as single copies at a defined genomic locus in CHO cells with identical regulatory elements. Steady-state mRNA abundance, quantified by deep sequencing of gDNA and mRNA, served as a proxy for mRNA stability. These data were used to train a machine learning model that predicts mRNA abund

SETDB1 knockdown boosts recombinant protein in CHO cells via epigenetic transcriptional silencing.

Chinese hamster ovary (CHO) cells serve as the predominant mammalian expression system for recombinant protein production. However, clonal heterogeneity and instability in recombinant protein expression remain significant challenges. SET domain bifurcated histone lysine methyltransferase 1 (SETDB1) catalyzes histone 3 lysine 9 trimethylation (H3K9me3), a critical epigenetic modification regulating gene expression. Despite its functional importance, the dynamics of SETDB1 expression and its regulatory impacts in CHO cells remain poorly characterised. Through transcriptomic analysis of high- and

[Recombinant porcine interferon-gamma expressed in CHO cells and its antiviral activity].

The aim of this study was to produce recombinant porcine interferon gamma (rPoIFN-γ) by Chinese hamster ovarian (CHO) cells expression system and to analyze its antiviral activity. Firstly, we constructed the recombinant eukaryotic expression plasmid pcDNA3.1-PoIFN-γ and transfected into suspension cultured CHO cells for secretory expression of rPoIFN-γ. The rPoIFN-γ was purified by affinity chromatography and identified with SDS-PAGE and Western blotting. Subsequently, the cytotoxicity of rPoIFN-γ was analyzed by CCK-8 test, and the antiviral activity of rPoIFN-γ was evaluated using standard

Physiological alterations of GS-CHO cells in response to adenosine monophosphate treatment.

Growth-arrested strategies (e.g. hypothermia and hyperosmolarity) have been widely employed to enhance cell-specific productivity (qP) in mammalian cell culture bioprocess. In addition to enhanced qP, alterations in cell physiology, such as cell size and cell cycle phase, have also attracted extensive attention under growth-arrested conditions. However, to date, very few reports on associations between physiological changes in growth-inhibiting approaches have been published. In this study, we explored associations between the physiological changes of GS-CHO cells in response to adenosine mono

Mechanism of UV-induced deoxynucleoside triphosphate pool imbalance in CHO-K1 cells.

Several laboratories have reported that exposure of cells to UV radiation results in a significant imbalance in deoxynucleoside triphosphate pool concentrations. In our CHO-K1 cells, a rapid drop in dCTP is accompanied by a rapid increase in dTTP. Examination of enzyme activities associated with synthesis/degradation of these molecules suggests that UV transiently enhances a putative dCTPase, dCMP deaminase and CdR kinase activities. This results in accumulation of excess dUMP which is probably converted to dTMP, then to dTTP. The absence of dCMP deaminase in V79 cells prohibits this rapid res

A screening method for isolating DNA repair-deficient mutants of CHO cells.

A simple procedure for isolating mutagen-sensitive clones of CHO cells was developed and applied in mutant hunts in which colonies were screened for hypersensitivity to killing by ultraviolet radiation (UV, ethyl methanesulfonate (EMS), or mitomycin C (MMC). Each of two UV-sensitive clones studied in detail had a D37 dose of 1.0 J/m2 compared to 7.0 J/m2 for the wild-type cells, and each was shown to have no detectable repair replication following exposure to UV doses of up to 26 J/m2. Although these mutants resemble xeroderma pigmentosum human mutants with respect to their repair defect and c

Improving recombinant protein productivity in CHO cells via multi-omics data integration.

Chinese hamster ovary (CHO) cells represent the dominant host system for the production of recombinant therapeutic proteins. In recent decades, extensive research has focused on process/media optimization and cell line engineering to improve both the productivity and quality of biopharmaceutical proteins produced in CHO cells. Nevertheless, the inherent complexity of biological pathways and the heterogeneous cellular responses to different environmental conditions have posed substantial challenges to traditional methodologies. Recent advances in omics technologies have enabled comprehensive ch

Using endoplasmic reticulum engineering to improve recombinant protein production in CHO cells.

Chinese hamster ovary (CHO) cells are commonly used to produce recombinant therapeutic proteins (RTPs). While recent strategies have significantly improved the expression levels of RTPs in CHO cells, insufficient secretion and endoplasmic reticulum (ER) stress remain major bottlenecks. Therefore, further understanding of the mechanism of the ER stress response, optimization of ER-related folding and degradation pathways, and development of more efficient ER engineering tools are expected to overcome this issue and maximize RTP production. In this review, we summarize the role of ER in recombin

Genetic Factors Affecting the Quality of Recombinant Therapeutic Proteins in CHO Cells.

Recombinant therapeutic proteins (RTPs) are widely used to treat various major diseases. Chinese hamster ovary (CHO) cells are the preferred mammalian cell expression system for the production of RTPs. However, maintaining high productivity while ensuring good-quality RTPs is still challenging. Glycosylation, aggregation, charge variants, and degradation are the main quality attributes of RTPs and can impact their safety, biological activity, stability, and half-life. Modifications of associated genetic factors have been performed to improve the quality of the RTPs. For example, knocking out t

Oxidative stress and endoreduplication induced by blue light exposure to CHO cells.

Blue light is commonly used for the treatment of Neonatal Jaundice and as a photodynamic therapy for cancer. In comparison to ultraviolet light, blue light has a lower toxicity due to the differences in photon energies. However, blue light can still be mutagenic to cells. The proposed mechanism suggests blue light exposure induces reactive oxygen species inducing oxidative stress. In this study, we examined how blue light exposure caused genotoxic effects utilizing Chinese hamster ovary (CHO) cells and UV135 cells when exposed to fluorescent blue light. Cytotoxic effects of blue light exposure

Mannose metabolism in recombinant CHO cells and its effect on IgG glycosylation.

Understanding the causes of high-mannose (HM) glycosylation of recombinant IgG in CHO cells would facilitate the production of therapeutics. CHO cells grown with mannose as the major carbon source demonstrated a dramatic increase in total HM glycosylation in recombinant IgG, with no effect on cell growth, viability, or titer. Quantitative metabolomics and (13) C flux analysis were used to explore the mechanism for increased HM glycosylation and understand the metabolism of mannose in CHO cells. It was demonstrated that mannose was a good carbon source for CHO cell growth and IgG production, re
