Cyanide insensitive respiration in mammalian cells: an artifact of mycoplasmal contamination.

Oxygen consumption by mammalian cells occurs primarily in the mitochondria although alternate oxidation pathways are also present. The resistance (often high) of these alternate pathways to metabolic inhibitors like CN- determine the residual oxygen consumption in the presence of the inhibitor. In many plant cells and micro-organisms the cyanide insensitive pathway can account for an appreciable fraction of the total oxygen consumption. We were studying oxidation by electron-affinic drugs in nonmitochondrial pathways and were surprised to find some mammalian cell lines having 10% - 90% residua

In vitro measurement of respiration of choroid plexus cells in communicating hydrocephalus.

Communicating hydrocephalus occurs as a result of inappropriate CSF production in the circumstance of diminished absorption. Ventricular enlargement does not progress as rapidly as the measured normal production of CSF would suggest. Thus, compensatory mechanisms must exist to respond in some manner to the altered pathophysiology. In this experiment the metabolic activity (O2 consumption) of the choroid plexus cells in animals with communicating hydrocephalus was compared with that of normals. The modified Cartesian diver technique of Zeuthen13 was employed. These preliminary measurements show

Coupling of glycine and alanine transport to respiration in cells of Escherichia coli.

Energy coupling for uptake of glycine and alanine in glycerol grown cells of Escherichia coli differs from that of the aromatic amino acids. Respiration and uptake of glycine and alanine show similar inactivations in cells exposed to high intensity violet light or to various concentrations of cyanide. In contrast, uptake of phenylalanine, tyrosine, and tryptophan is resistant to effects of light or cyanide. Anoxia largely inhibits uptake of glycine and alanine while that of the aromatic amino acids is only partially affected. Furthermore, ferricyanide (but not ferrocyanide) completely restores

RETRACTED: T cell metabolism. The protein LEM promotes CD8⁺ T cell immunity through effects on mitochondrial respiration.

Источник пометил публикацию как отозванную

Protective CD8(+) T cell-mediated immunity requires a massive expansion in cell number and the development of long-lived memory cells. Using forward genetics in mice, we identified an orphan protein named lymphocyte expansion molecule (LEM) that promoted antigen-dependent CD8(+) T cell proliferation, effector function, and memory cell generation in response to infection with lymphocytic choriomeningitis virus. Generation of LEM-deficient mice confirmed these results. Through interaction with CR6 interacting factor (CRIF1), LEM controlled the levels of oxidative phosphorylation (OXPHOS) complex

UDP-glucose regulates dendritic cell mitochondrial respiration via a nitric oxide-dependent mechanism.

Bone marrow-derived dendritic cell (BMDC) activation is associated with rewiring of cellular metabolism and concurrent large-scale changes in gene expression promoting a proinflammatory program characterized by expression of inducible nitric oxide synthase and the production of nitric oxide (NO). NO inhibits vital cellular activities including mitochondrial respiration. Mitochondrial respiration inhibition via NO occurs at discrete levels of activating stimulus, termed the mitochondrial respiration threshold, and regulation of this threshold is not fully understood. In this work, we characteri

Mitochondrial respiration restricts Listeria monocytogenes infection by slowing down host cell receptor recycling.

Mutations in mitochondrial genes impairing energy production cause mitochondrial diseases (MDs), and clinical studies have shown that MD patients are prone to bacterial infections. However, the relationship between mitochondrial (dys)function and infection remains largely unexplored, especially in epithelial cells, the first barrier to many pathogens. Here, we generate an epithelial cell model for one of the most common mitochondrial diseases, Leigh syndrome, by deleting surfeit locus protein 1 (SURF1), an assembly factor for respiratory chain complex IV. We use this genetic model and a comple

Dapagliflozin, An SGLT2 Inhibitor, Improves Endothelial Cell Energy Metabolism Through Enhanced Mitochondrial Respiration.

BACKGROUND/AIMS: Flozins (sodium-glucose cotransporter 2 inhibitors, SGLT2i) are a new class of antidiabetic drugs that reduce cardiovascular mortality and hospitalization rates in heart failure, regardless of type 2 diabetes status. Besides lowering glycemia by inhibiting renal glucose reabsorption, SGLT2 inhibitors may exert sodium-dependent hemodynamic effects and improve cardiomyocyte energy metabolism, substrate preference, and mitochondrial function. However, their impact on endothelial cells remains largely unknown. This study aimed to analyse the effects and mechanisms of SGLT2i on end

Teriflunomide Inhibits Human FOXP3+ Regulatory T Cell Function by Interference With Mitochondrial Respiration.

Regulatory FOXP3+ T cells (Tregs) have been characterized with unique metabolic demands, preferentially relying on fatty acid β-oxidation (FAO) and oxidative phosphorylation (OXPHOS). Several studies have indicated that Treg mitochondrial fitness is crucial for maintaining their stability and suppressive activity with an emphasis on complex-III of the electron transport chain (ETC). Dysfunctional Tregs isolated from patients with autoimmunity like multiple sclerosis (MS) show diminished mitochondrial respiration and the induction of a T helper (Th)1-like phenotype, characterized by increased p

Reduced mitochondrial respiration in peripheral T cells after paediatric heamatopoietic stem cell transplantation.

BACKGROUND: Recovery and functional differentiation of T-cell subsets are central for the development of immune function and complications after allogeneic hematopoietic stem cell transplantation (HSCT), but little is known about the cellular respiration and factors influencing T-cell metabolic fitness during immune maturation after HSCT. METHOD: We included 20 HSCT patients and analysed mitochondrial oxidative phosphorylation and mitochondrial fitness in peripheral blood mononuclear cell samples collected at days +90 and +180 after HSCT. RESULTS: Phenotypic analysis revealed lower overall T

Dimethyl Fumarate Enhances Venetoclax-Induced Cell Death by Inhibiting Mitochondrial Respiration and Cell Cycle Control in Colorectal Cancer.

Colorectal cancer (CRC) is a leading cause of cancer death, with resistance and apoptosis evasion-often via Bcl-2-representing major challenges. The redox-modulating drug dimethyl fumarate (DMF) has demonstrated efficacy in hematologic malignancies; however, its potential in solid tumors remains unclear. Here, we show that DMF, especially in combination with the Bcl-2 inhibitor venetoclax (ABT-199), induces apoptosis in HCT-116 CRC cells. DMF impairs mitochondrial respiration, causing membrane hyperpolarization, ATP depletion, autophagy, and cell cycle arrest. Combined treatment increases meta

Enhanced glucose transport in response to inhibition of respiration in Clone 9 cells.

An acceleration of ATP synthesis by anaerobic glycolysis provides important compensation for interference with respiration in a variety of cells. Effective compensation for an inhibition of respiration, however, can occur in cells in which glucose entry is rate limiting only if sufficient glucose becomes available through an enhancement of transport. We present here a detailed study of the effects of inhibition of respiration in Clone 9 cells, a continuous cell line characterized by low internal glucose concentrations (less than 10% that of the external medium) and minimal stores of glycogen.

[Respiration and oxidative phosphorylation in isolated liver cells under freezing conditions].

Respiration and oxidative phosphorylation of hepatocytes were studied as affected by low temperatures and cryoprotectors. Fast freezing of these cells down to -196 degrees C with the presence of mentioned substances causes considerable destruction of their structures. The intensity of the oxygen endogenic uptake lowers significantly, respiration and oxidative phosphorylation become uncoupled. Two-stage freezing down to -196 degrees C also causes a complete uncoupling of the oxidation and phosphorylation processes in the hepatocyte mitochondria. When freezing hepatocytes down to -196 degrees C

Sexual Dimorphism in Red Blood Cell Mitochondrial Respiration during Breeding Fasts in King Penguins.

AbstractBecause of their extended fasting period on land during breeding, male king penguins have been extensively studied in order to unravel the physiological adaptations that enable them to fast while having to find a partner, defend their territory, or brood their offspring. While the different phases of fasting and the nature of the metabolic fuels used are well characterized in male king penguins, few studies have focused on the efficiency of the conversion of the metabolic resources into energy at a cellular level through mitochondrial respiration. Furthermore, little information is ava

Disruption of mitochondrial respiration inhibits volume-regulated anion channels and provokes neuronal cell swelling.

Hypoxia and inhibitors of mitochondrial respiration impair the regulatory volume decrease (RVD) of cerebellar granule neurons after hypotonic swelling. RVD is linked to the opening of volume-regulated anion channels (VRACs). VRACs are outwardly rectifying, inactivate slowly during maintained depolarization, and are permeable to the cellular organic osmolyte taurine. Channel activation requires nonhydrolytic ATP binding and is not modulated by intracellular ADP. VRAC opening is reversibly depressed by hypoxia and by mitochondrial inhibitors such as oligomycin, rotenone, and antimycin A. These r

Fatty acid oxidation fuels mitochondrial respiration to drive epidermal stem cell fate and differentiation.

Skin barrier function relies on the epidermis, whose integrity is maintained by basal stem cells that continuously renew and differentiate into a multilayered architecture. Disrupted epidermal differentiation underlies numerous hyperproliferative and inflammatory skin disorders. While transcriptional and epigenetic mechanisms are known to regulate late differentiation, the molecular events driving early commitment remain elusive. Here, we reveal that early mitochondrial reprogramming, characterized by the activation of oxidative phosphorylation, is a determinant of differentiation initiation.

Optimization of Extracellular Flux Assay to Measure Respiration of Anchorage-independent Tumor Cell Spheroids.

Three-dimensional (3D) cell culture models are widely used in tumor studies to more accurately reflect cell-cell interactions and tumor growth conditions in vivo. 3D anchorage-independent spheroids derived by culturing cells in ultra-low attachment (ULA) conditions is particularly relevant to ovarian cancer, as such cell clusters are often observed in malignant ascites of late-stage ovarian cancer patients. We and others have found that cells derived from anchorage-independent spheroids vary widely in gene expression profiles, proliferative state, and metabolism compared to cells maintained un

Improvement of Platelet Respiration by Cell-Permeable Succinate in Diabetic Patients Treated with Statins.

Diabetes mellitus (DM) is the most severe metabolic disease that reached the level of a global pandemic and is associated with high cardiovascular morbidity. Statins are the first-line lipid-lowering therapy in diabetic patients with or without a history of atherosclerotic disease. Although well tolerated, chronic treatment may result in side effects that lead to treatment interruption. Mitochondrial dysfunction has emerged as a central pathomechanism in DM- and statin-induced side effects. Assessment of mitochondrial respiration in peripheral platelets has been increasingly used as a mirror o
