The effect of clofibrate on hyperbilirubinemia of term neonates.

Clofibrate is a glucuronosyl transferase inducer that has been proposed to increase the elimination of bilirubin in neonates with hyperbilirubinemia. This study was conducted to determine the therapeutic effect of clofibrate in term neonates with non-hemolytic jaundice. This study was conducted on 52 newborns with pathologic unconjugated jaundice in Qazvin children hospital. Newborns divided randomly in two groups. Case group treated with clofibrate and intensive phototherapy, while control group treated only with intensive phototherapy. Serum bilirubin level was measured before and 6, 12, 24

Copper deficiency in rats: the effect of clofibrate.

This study was undertaken to determine whether hepatic lipogenesis plays a role in the exacerbation of copper (Cu) deficiency. Forty-eight male rats were fed from weaning a Cu-deficient or adequate diet containing 62% carbohydrate as either starch or fructose with or without clofibrate for 5 weeks. Clofibrate was fed since it had been shown to possess hypolipidemic properties. Administration of clofibrate reduced the activity of the lipogenic enzyme glucose-6-phosphate dehydrogenase. Total hepatic lipid, however, was not reduced. Clofibrate did not affect hepatic lipid concentration and the pa

[Clofibrate therapy--current status from a gerontological viewpoint].

Hyperlipoproteinemias are the most frequent metabolic disorders occurring in old age. Because of their capacity to initiate, increase, or accelerate the process of atherogenesis, it is necessary that such disturbances of metabolism be properly treated. One of the most effective drugs which is finding wide application in the medical treatment of hyperlipoproteinemia is clofibrate of which the active substance is clofibrinic acid (Regadrin). The results of an extensive long-time study of clofibrate by World Health Organization workers had led to a world wide discussion of the effects and side ef

Myopathy induced by clofibrate treatment in normolipaemic patients.

On the basis of the occurrence of myopathy in a clofibric acid treated child with diabetes insipidus, three other children with the same diseases were examined in order to find indications for a myopathic side-effect of the drug. When the children were found to have increased creatine phosphokinase activity and EMG changes, two of the authors took clofibric acid themselves. In both test persons subclinical myopathy was produced. After stopping the drug, transitional hypertriglyceridaemia occurred. These side-effects should serve as a warning of an uncritical application of clofibric acid and i

Hypolipemic activity of clofibrate-related compounds. 2nd Communication.

Clofibrate-related compounds were synthesized from substituted phenoxyacetic acids by esterification with salicylic acid derivatives, by amidation of thiazolidines or by reaction with cinnarizine. Lipid lowering effect was investigated in normolipemic and hyperlipemic rats. The long-term hyperlipemic model used permits differentiation of hypolipemic efficacy, i.e., from clofibrate or compounds of similar activity.

One-year trials with halofenate, clofibrate, and placebo.

The hypolipidemic as well as other laboratory and clinical effects of halofenate, clofibrate, and placebo were compared in 29 patients with type IV hyperlipoproteinemia in a double-blind, controlled, therapeutic trial of 1 yr duration. Plasma drug levels were obtained to monitor compliance. Clofibrate and halofenate lowered serum triglycerides to a similar extent. The hypotriglyceridemic effect of halofenate was significant only when data from noncompliant patients were discarded. Only clofibrate lowered baseline levels of plasma cholesterol. Very low density lipoproteins were decreased and lo

Evaluation of the carcinogenic potential of clofibrate in the neonatal mouse.

This study was conducted in support of the International Life Sciences Institute (ILSI) alternative carcinogenicity models initiative to evaluate the carcinogenic potential of clofibrate, a nongenotoxic peroxisome proliferator-activated receptor (PPAR) alpha agonist, following oral administration to neonatal mice. Male and female neonatal CD-1 mice were dosed with clofibrate at doses of 100, 250, and 500 mg/kg or with the positive control, diethylnitrosamine (DEN), at 2 mg/kg by oral gavage on days 9 and 16 post birth and observed for approximately 1 year for the development of tumors. Clofibrate-induced myopathy in patients with diabetes insipidus.

Clofibrate has been considered to be a relatively safe antidiuretic in the treatment of diabetes insipidus. However, we have recently had four cases of clofibrate-induced myopathy in patients with diabetes insipidus due to hypothalamic lesions. Physicians should therefore be aware of its occurrence and carefully monitor serum levels of CPK, GOT and GPT during the treatment of diabetes insipidus with clofibrate, especially in patients with associated hypothyroidism, latent or overt, which possibly favors the development of myopathy.

The Effects of Clofibrate on Neonatal Jaundice: A Systematic Review.

BACKGROUND: Neonatal jaundice is a prevalent disease that causes many complications, including kernicterus and even death. Previous studies have shown that clofibrate as an aryloxy isobutyric acid derivate can be effectively applied for the treatment of neonatal jaundice. Thus, this review was carried out to investigate the effects and mechanism of action of clofibrate on neonatal jaundice. METHODS: The keywords such as "Clofibrate" in combination with "Neonatal jaundice" or "Neonatal hyperbilirubinemia" or "Newborn Jaundice" were used to search for relevant publications indexed in the Instit

Clofibrate in combination with phototherapy for unconjugated neonatal hyperbilirubinaemia.

BACKGROUND: There are many pathological conditions leading to an elevated unconjugated bilirubin level (hyperbilirubinaemia) in neonates. Currently the standard therapies for unconjugated hyperbilirubinaemia include phototherapy and exchange transfusion. In addition to phototherapy, clofibrate has been studied as a treatment for hyperbilirubinaemia in several countries. OBJECTIVES: To determine the efficacy and safety of clofibrate in combination with phototherapy versus phototherapy alone in unconjugated neonatal hyperbilirubinaemia. SEARCH METHODS: Randomised controlled trials were identif

Evaluation of the carcinogenic potential of clofibrate in the rasH2 mouse.

The purpose of the study was to support of the International Life Sciences Institute (ILSI) alternative carcinogenicity models initiative to evaluate the carcinogenic potential of the nongenotoxic carcinogen, clofibrate, a peroxisome proliferator-activated receptor (PPAR) alpha agonist, following oral administration to rasH2 mice. Peroxisome proliferators are one of the most widely studied of the nongenotoxic carcinogens and have diverse industrial and therapeutic uses (Gonzalez et al. J. Nat. Cancer Inst. 90: 1702-1709, 1998); however, the nongenotoxic mechanism of carcinogenicity is currentl

[Pharmacokinetics of clofibrate in jaundiced newborn infants at term].

BACKGROUND: Clofibrate (CFB) has been proposed to increase elimination of bilirubin in neonates with hyperbilirubinemia. Nevertheless, its disposition, at this age, remains unknown. The aim of this work was to characterize pharmacokinetics of an oil formulation of CFB in neonates at term with jaundice. PATIENTS AND METHODS: Two groups (G1 and G2) of eight neonates, presenting with jaundice, entered an open, non randomized and comparative study. Five blood samples were collected over 50 hours following a single oral administration of 100 mg/kg or 50 mg/kg CFB, respectively, in G1 and G2. Serum

Effect of clofibrate and hydrocortisone on intestinal trichinellosis in mice.

These investigations show the immunosuppressive effects of Clofibrate (CPIB) and hydrocortisone on the intestinal cycle of Trichinella nelsoni. They demonstrate an effect of CPIB similar to that of hydrocortisone in the establishment of the muscular and intestinal phase of this parasite. A higher retention rate of intestinal adult worms was observed in hydrocortisone-treated mice. These facts are consistent with the idea that CPIB can act on diverse immune cells, involving both thymus-dependent and thymus-independent antigens.

Influence of clofibrate on blood viscosity in primary hyperlipoproteinemia.

In patients with primary hyperlipoproteinemia (23 patients type II a and II b, 13 patients type IV), changes in lipoprotein concentrations (lipid fraction), fibrinogen concentrations, in kinematic serum and plasma viscosity (determined by means of a capillary viscosimeter) and changes in relative apparent viscosity at shear rate 46 s-1 and 115 s-1 (determined by means of a plate-cone-viscosimeter) were measured before and after a clofibrate therapy (2 X 1 g/day) lasting for 7 to 9 weeks. Serum and plasma viscosity was lowered significantly in both types II and IV. The apparent viscosity decrea
