Toxicity and biochemical action of the antibiotic fungicide tetramycin on Colletotrichum scovillei.

Tetramycin, a novel polyene macrolide antibiotic, has strong activity against a broad spectrum of fungi and may have potential uses in future agricultural applications. Thus, the antifungal activity and biochemical action of tetramycin on Colletotrichum scovillei were investigated in this study. The experimental results indicated that tetramycin had strong inhibitory activity against the mycelial growth, spore germination and germ tube elongation of C. scovillei. The baseline sensitivity curves were unimodal, with mean EC50 values of 1.98 ± 0.078 μg/mL and 0.003 ± 0.005 μg/mL for mycelial grow…

Mating and Pathogenicity of the Dominant Colletotrichum Species Associated with Anthracnose Disease of Mango.

Anthracnose is one of the main diseases of mango, which seriously affects the yield and quality. Previous studies found that mango anthracnose in China involves at least 13 species of Colletotrichum. From mango with anthracnose symptoms samples, we previously obtained 134 strains from 13 species, and 26.0% were C. fructicola, while 27.6% were C. siamense, and 31.3% were C. asianum, with a few specimens for each of the remaining species. These three main species were used in intraspecific mating tests to assess sexual reproduction. The intraspecific mating tests revealed that C. fructicola and …

CgFre2 contributes to ferric reductase activity, growth and pathogenicity in Colletotrichum gloeosporioides.

Ferric reductases are oxidoreductases that catalyze the reduction of environmental Fe³ ⁺ to bioavailable Fe² ⁺ and mediate cellular iron uptake and metabolism. However, the specific biological functions of ferric reductases in Colletotrichum gloeosporioides have not been fully characterized. In this study, 15 ferric reductase-like proteins were screened from the genome annotation of C. gloeosporioides, and CgFre2 was selected for further functional characterization. Targeted deletion of CgFre2 resulted in reduced mycelial growth and a sharp decrease in conidiation. Further observation of conid…

Resistance to thiophanate-methyl and azoxystrobin in Colletotrichum species from Brazilian grapes.

BACKGROUND: Grape ripe rot (GRR) caused by Colletotrichum species can cause yield loss in vineyards in Southern and Northeast Brazil. Thiophanate-methyl (FRAC 1) and azoxystrobin (FRAC 11) fungicides are frequently used to control GRR, but resistance to these fungicides in Colletotrichum species may impact their efficacy. RESULTS: Sensitivity to thiophanate-methyl was assessed for 89 isolates of the C. gloeosporioides species complex using mycelial growth inhibition. Isolates were classified as sensitive (n = 2), low-resistant (n = 70) or highly resistant (n = 17). Highly resistant isolates h…

The Role of Fatty Acids from Plant Surfaces in the Infectivity of Colletotrichum fioriniae.

Aqueous extracts derived from flowers stimulate germination, secondary conidiation, and appressorial formation of various latent fruit rotting fungi. Even raindrops passing over flowers accumulate sufficient activity to influence the infectivity of fruit rotting fungi. Using a spore germination bioassay, high levels of bioactivity were found in chloroform extracts from plant tissues, implicating the nonpolar components of the cuticle. The fatty acid (FA) and fatty acid methyl ester (FAME) composition (C9-C20) of blueberry and cranberry tissues as well as aqueous flower extracts were characteri

Antifungal Activity of Alpha-Mangostin against Colletotrichum gloeosporioides In Vitro and In Vivo.

We investigated alpha-mangostin (α-mangostin, α-MG), a xanthone natural product extracted from the pericarp of mangosteen (Garcinia mangostana), for its antifungal activities and possible mechanism against Colletotrichum gloeosporioides, which causes mango anthracnose. The results demonstrated that α-MG had a relatively high in vitro inhibitory activity against C. gloeosporioides among 20 plant pathogenic fungi. The median effective concentration (EC50) values of α-MG against mycelial growth were nearly 10 times higher than those of spore germination inhibition for both strains of C. gloeospor…

The MdERF1B-miR482c-MdCCR4-MdTLP1b module regulates apple resistance to Colletotrichum gloeosporioides.

Glomerella leaf spot (GLS), incited by Colletotrichum gloeosporioides, poses a significant threat to apple (Malus domestica) production. microRNAs (miRNAs) are critical regulators of plant-pathogen crosstalk. However, the underlying mechanism through which miR482 regulates apple resistance to GLS remains unclear. Here, we show that overexpression of miR482c compromises resistance against C. gloeosporioides, while silencing miR482c confers resistance to C. gloeosporioides. 5'RACE assays in apple and co-expression assays in Nicotiana benthamiana confirmed that miR482c targets and cleaves the MdC…

An oxidized ergosterol from Pleurotus cystidiosus active against anthracnose causing Colletotrichum gloeosporioides.

This study was undertaken to study the antifungal activity of Pleurotus cystidiosus against Colletotrichum gloeosporioides. This was achieved by fractionating the mushroom, P. cystidiosus initially to acetone (A), dichloromethane (D), and hexane (H) and studying the antifungal activity using the standard poisoned food technique. All the test solutions used were in the concentration of 20,000 ppm. The percentage inhibition of extracts A, D, and H was 12, 7, and 0.4%, respectively. Antifungal assay guided fractionation of the most active extract A resulted in four fractions; A1, A2, A3, and A4 h…

Systematic Identification and Functional Characterisation of Colletotrichum fructicola Effectors During Camellia oleifera Colonisation.

Camellia oleifera is an important woody oil crop in southern China, but its production is severely threatened by anthracnose caused by Colletotrichum fructicola. C. fructicola deploys secreted effector proteins to establish infection. However, systematic identification and functional characterisation of C. fructicola effector genes upregulated during infection remains largely unexplored. Here, we integrated genome-wide secretome prediction with RNA-seq data from C. oleifera leaves inoculated with C. fructicola to identify candidate effectors induced during infection, followed by functional scr…

Novel Design of Citral-Thiourea Derivatives for Enhancing Antifungal Potential against Colletotrichum gloeosporioides.

Although much progress has been made in developing botanical fungicides to combat fungal diseases in crops, there remains a great need to improve the efficiency and long-term safety of these fungicides. This study proposes a novel strategy for designing citral-thiourea derivatives that feature such desirable properties. The motivation of the work herein was to enhance the antifungal activity of citral against C. gloeosprioides by exploiting the synergistic effect that arises from combining citral and thiourea compounds, thereby producing citral-thiourea derivatives that exhibit good long-term
