D-Cycloserine in Neuropsychiatric Diseases: A Systematic Review.

D-Cycloserine, known from tuberculosis therapy, has been widely introduced to neuropsychiatric studies, since its central active mechanism as a partial NMDA-agonist has been found. In this review, we evaluate its therapeutic potential in neuropsychological disorders and discuss its pitfalls in terms of dosing and application frequency as well as its safety in low-dose therapy. Therefore, we identified 91 clinical trials by performing a Medline search. We demonstrate in part preliminary but increasing evidence that D-cycloserine may be effective in various psychiatric diseases, including schizo…

d-Cycloserine augmentation of cognitive remediation in schizophrenia.

d-Cycloserine (DCS) has been shown to enhance memory and, in a previous trial, once-weekly DCS improved negative symptoms in schizophrenia subjects. We hypothesized that DCS combined with a cognitive remediation (CR) program would improve memory of a practiced auditory discrimination task and that gains would generalize to performance on unpracticed cognitive tasks. Stable, medicated adult schizophrenia outpatients participated in the Brain Fitness CR program 3-5 times per week for 8weeks. Subjects were randomly assigned to once-weekly adjunctive treatment with DCS (50mg) or placebo administer…

D-cycloserine enhances generalization of fear extinction in children.

BACKGROUND: For exposure therapy to be successful, it is essential that fear extinction learning extends beyond the treatment setting. D-cycloserine (DCS) may facilitate treatment gains by increasing generalization of extinction learning, however, its effects have not been tested in children. We examined whether DCS enhanced generalization of fear extinction learning across different stimuli and contexts among children with specific phobias. METHODS: The study was a double-blind placebo-controlled randomized controlled trial among dog or spider phobic children aged 6-14. Participants ingested…

[Comparative clinical electroencephalographic study of cycloserine and terizidon tolerance].

The aim of the study was comparative investigation of cycloserin and terizidone tolerance by patients with the lung tuberculosis. The investigation procedure included parallel clinical observations and electroencephalographic studies (EEG) using light and sonic stimulation. Sixty six and 26 patients treated with cycloserin and terizidone respectively were observed. The results of the studies showed that by the mechanism of their action on the central nervous system terizidone and cycloserine were similar and increased the general reactivity of the brain. However, terizidone promoted an increas…

Cycloserine resistance among drug-resistant tuberculosis cases in Taiwan.

Cycloserine (CS) is a widely used drug for drug-resistant tuberculosis (DR-TB) treatment. The World Health Organization (WHO) recently suggested a critical concentration (CC) for CS using the MGIT 960 system (MGIT). To strengthen our DR-TB management program, we performed CS resistance analyses to provide comprehensive drug susceptibility testing (DST). This retrospective study included Mycobacterium tuberculosis complex (MTBC) isolates obtained from 114 rifampin-resistant (RR) and multidrug-resistant (MDR) TB cases. We compared the results of phenotypic DST (pDST) and genotypic DST (gDST) and…

Augmentation of exposure therapy with post-session administration of D-cycloserine.

BACKGROUND: Pre-session administration of d-cycloserine (DCS) has been found to augment exposure therapy outcomes in a variety of anxiety disorders. To be able to enhance learning only for successful exposure sessions, it would be beneficial to have the option of administering DCS after rather than before the session, a strategy encouraged by pre-clinical work. We believe the present study is the first published report on the efficacy of post-session administration of DCS in humans. METHOD: Adults (N = 29) with a DSM-IV diagnosis of acrophobia were randomized to receive two sessions of virtua…
