Twenty-three guinea pigs, presensitized with horse serum, were injected intracardially with 1 ml of 1.5 X 10(-4) M cytochalasin B in a 2.5% dimethyl sulfoxide solution, 1.5 h before being challenged with the serum. Eighteen of the guinea pigs (80%) survived. A single intracardiac injection of cytochalasin B to the control group of guinea pigs at 1.5 h before administration of the allergen did not prevent their sensitization and death following challenge.

Mouse eggs and spermatozoa were treated in various ways with 5 or 10 mug cytochalasin B/ml. The fertilization rate in vitro was reduced by treatment with the drug but 80-90% of the eggs fertilized were triploid. Many of the experimental eggs were penetrated by one or more spermatozoa but remained unfertilized (75% compared with 9% in control eggs). It is suggested that cytochalasin B weakens the zona reaction and interferes with fusion of gametes but does not prevent the block to polyspermy.

Adult toxicity, embryo lethality, and teratogenicity of cytochalasins D and E (CD and CE) were determined for A/J, C57BL/6JH, aeae, and OELN mouse strains. The adult LD50 for CD (1.9-2.6 mg/kg) and CE (3.1-3.3) were similar for the strains. Swiss albino and C57BL/10 eggs from induced ovulations, and spontaneously ovulated A eggs, were activated in vitro by a heat shock of 44 degrees C for 5 to 7-5 min and cultured in the presence of 10 mug/ml of Cytochalasin B (CB) for 5-8 h. An intramolecular Diels-Alder (IMDA) reaction efficiently accelerated by Schreiner's thiourea is reported, to build a functionalized cytochalasin scaffold (periconiasin series) for biological purposes. DFT calculation highlighted a unique multidentate cooperative hydrogen bonding in this catalysis. The deprotection end game afforded a collection of diverse structures and showed the peculiar reactivity of the Diels-Alder cycloadducts upon functionalization. Biological studies revealed strong cytotoxicity of a few compounds on breast cancer cell lines while actin polymerization is preserved.

Intercellular communication in the tumor microenvironment has a major impact on therapeutic outcomes. In particular, highly aggressive cancers such as glioblastoma benefit from efficient cellular communication by developing resistance to therapy or by infiltrating surrounding tissue to escape radiotherapy. Tiny membrane tunnels, such as tunneling nanotubes (TNTs), can serve as an effective communication tool for tumor cells. With these structures, the cells can build a large network that allows them to react and adapt to their environment as quickly as possible, thereby combating the therapy.
