Cytokines in Cytokine Storm Syndrome.

As the eponymous mediators of the cytokine storm syndrome, cytokines are a pleomorphic and diverse set of soluble molecules that activate or suppress immune functions in a wide variety of ways. The relevant cytokines for each CSS are likely a result of differing combinations of environmental triggers and host susceptibilities. Because cytokines or their receptors may be specifically targeted by biologic therapeutics, understanding which cytokines are relevant for disease initiation and propagation for each unique CSS is of major clinical importance. This chapter will review what is known about…

Cytokines and drugs targeting cytokines in Graves' disease and thyroid eye disease.

Graves' disease (GD) is an autoimmune disorder characterized by the production of stimulatory autoantibodies against the thyrotropin receptor, leading to hyperthyroidism and diffuse goiter. The most frequent extrathyroidal manifestation is Graves' ophthalmopathy (GO), also known as thyroid eye disease (TED), which is responsible for significant morbidity due to orbital inflammation, tissue remodeling, and visual impairment. In recent years, increasing evidence has demonstrated that cytokines play a central role in both GD and TED, orchestrating immune cell activation, autoantibody production, …

The Cytokines of Asthma.

Asthma is a chronic inflammatory airway disease associated with type 2 cytokines interleukin-4 (IL-4), IL-5, and IL-13, which promote airway eosinophilia, mucus overproduction, bronchial hyperresponsiveness (BHR), and immunogloubulin E (IgE) synthesis. However, only half of asthma patients exhibit signs of an exacerbated Type 2 response. "Type 2-low" asthma has different immune features: airway neutrophilia, obesity-related systemic inflammation, or in some cases, few signs of immune activation. Here, we review the cytokine networks driving asthma, placing these in cellular context and incorpo…

Cytokines in aortic dissection.

Aortic dissection (AD) is one of the most dangerous forms of vascular disease, characterized by endometrial rupture and intramural hematoma formation. Generally, the pathological process is complicated and closely related to the infiltration of inflammatory cells into the aortic wall and apoptosis of vascular smooth muscle cells. Currently, multiple cytokines, including interleukins, interferon, the tumor necrosis factor superfamily, colony stimulating factor, chemotactic factor, growth factor and so on, have all been demonstrated to play a critical role in AD. Additionally, studies of the lin…

Cytokines in human leptospirosis.

BACKGROUND: Leptospirosis is a zoonotic disease with increased public health concern. Cytokines produced in response to the infection with pathogenic leptospires have been proposed to be involved in the pathogenesis of the disease. The aim of the study was to measure and evaluate the levels of 27 cytokines in patients with acute leptospirosis. METHODS: The levels of 27 cytokines were measured from 42 acute leptospirosis cases; 47 samples were obtained from severe cases. Statistical analysis was performed using SPSS. RESULTS: IL-6, IL-8, GM-CSF, IP-10, MCP-1, and VEGF levels differed signific…

Cytokines in acute chikungunya.

INTRODUCTION: Acute chikungunya (CHIKV) is predominantly an acute onset of excruciatingly painful, self-limiting musculoskeletal (MSK) arbovirus illness and this was further reported by us during the 2006 Indian epidemic [Chopra et al. Epidemiol Infect 2012]. Selected serum cytokines profile in subjects within one month of onset of illness is being presented. METHODS: Out of 509 clinical CHIKV cases (43% population) identified during a rural population survey, 225 subjects consented blood investigations. 132 examined within 30 days of febrile onset are the study cohort. Anti-CHIKV IgM and IgG…

Cytokines and Behcet's disease.

Behcet's Disease (BD) is a systemic vasculitis of unknown etiology. Increasing studies find that a sophisticated interlacing cytokine network is closely implicated in the onset, evolution and even organ damages of the disease. Cytokines involved can be categorized as Th1 type, Th2 type, Th17 type, chemokines and other proinflammatory cytokines, etc. The vicious cycle of cytokine network plays a substantial role in the disease pathogenesis and even in organ lesions, and might be disorganized by blocking one of the key links of the cytokines, which in turn may provide essential clues to outlook …

[Chronic prostatitis and cytokines].

Chronic prostatitis (CP) is a common disease in men, and its pathogenesis remains to be clarified. The precise regulation of cytokines is involved in all types and all stages of CP. The interaction of inflammation, inflammatory cells and cytokines leads to the development and progression of CP. With further understanding of the immunological and molecular biological mechanisms of the disease and more inflammatory cytokines used in its detection, it is of special significance to apply cytokines in the classification and treatment of CP. This review outlines the roles of cytokines in the pathoge…

Cytokines and cardiovascular disease.

The role of cytokines in the pathogenesis of cardiovascular disease is increasingly evident since the identification of immune/inflammatory mechanisms in atherosclerosis and heart failure. In this review, we describe how innate and adaptive immune cascades trigger the release of cytokines and chemokines, resulting in the initiation and progression of atherosclerosis. We discuss how cytokines have direct and indirect effects on myocardial function. These include myocardial depressant effects of nitric oxide (NO) synthase-generated NO, as well as the biochemical effects of cytokine-stimulated ar…

Cytokines in recurrent miscarriage.

Cytokines act at all stages of pregnancy from implantation to parturition. This review examines their relevance in recurrent miscarriage. However, recurrent miscarriage may be due to an inherently abnormal embryo (e.g., chromosomal abberations) or maternal factors (e.g., uterine anomalies or antiphospholipid antibodies). In the former, cytokines are not causative, but may be part of the mechanism of abortion. In the antiphospholipid syndrome, cytokines such as TNFalpha and IL-6 may be responsible for the associated thrombosis. Hence, an appropriate cytokine milieu could be responsible for whet…

Brain cytokines and disease.

Cytokines (e.g. various interleukins and subfamily members, tumor necrosis factors, interferons, chemokines and growth factors) act in the brain as immunoregulators and neuromodulators. Over a decade ago, the integrative article 'Immunoregulators in the Nervous System' (Neurosci Biobehav Rev 1991; 15: 185-215) provided a comprehensive framework of pivotal issues on cytokines and the nervous system that recently have been extensively studied. Cytokine profiles in the brain, including cytokine generation and action, have been studied in multiple models associated with neuropathophysiological con…

Illness, cytokines, and depression.

Various medical conditions that involve activation of the immune system are associated with psychological and neuroendocrine changes that re … release, and the expression of splenic TNF-alpha mRNA. Taken together, these findings suggest that cytokines are involved in the etiology and symptomatology of illness-associated depression.

Cytokines in Gaucher's disease.

Gaucher's disease (GD) is characterized by hepatosplenomegaly, bone marrow infiltration, osteonecrosis, which may all be associated with the presence of pathological macrophages that contain undegraded glycosphingolipids. Levels of serum cytokines, which are soluble products of mononuclear phagocytes (MNP), were evaluated in 24 GD patients. Levels of interleukin-1beta (IL-1beta), interleukin-1 receptor antagonist (IL-1Ra), interleukin-6 (IL-6), tumor necrosis factor-alpha (TNF-alpha), and soluble interleukin-2 receptor (sIL-2R) in GD patients were significantly higher than in normal controls. …

Serum cytokines in thyrotoxicosis.

Overproduction of thyroid hormones promotes bone resorption in vivo and in vitro, and we have evaluated whether mediators of such effects could include the osteotropic cytokines. Previous studies have demonstrated raised serum interleukin (IL)-6 in thyrotoxic patients, but differentiating the contribution of the elevated thyroid hormones from that of the autoimmune inflammation present in Graves' disease (GD) has been difficult. We undertook a longitudinal study of 34 patients (19-45 yr old) with GD, toxic nodular goiter (TNG), or a history of thyroid carcinoma but no evidence of disease recur…

Blocking cytokines with genes.

The pathophysiology of rheumatoid arthritis (RA) is primarily driven by proinflammatory cytokines such as interleukin-1 (IL-1) and tumor necrosis factor alpha. Several biological agents have been identified that effectively block the activity of these cellular messengers, and administration of these agents to animal models of RA and human patients has been found to have therapeutic benefit. The application of gene therapy for the treatment of RA and other articular diseases is being explored to overcome current limitations with delivery of therapeutics to joint tissues. To date, laboratory res…

[Cytokines and skin diseases].

Many skin diseases such as eczema and psoriasis are characterised by a chronic inflammatory skin condition. In this respect they resemble other chronic diseases such as rheumatoid arthritis, bronchial asthma, ulcerous colitis and Crohn's disease. A persistent accumulation, predominantly of T-lymphocytes constitutes the central pathophysiological feature of such diseases. The past 15-20 years have witnessed the characterisation of an extensive series of peptides known as cytokines. These are soluble, relatively low molecular weight peptides which at low concentrations mediate regulation of cell…

[Therapeutic use of cytokines].

Cytokines represent a growing number of biologically highly active polypeptides, which exert important functions in haematopoiesis and immune response. Cytokines are predominantly released by activated lymphocytes and macrophages. Increasing insight into the role of these factors was accompanied by large scale availability made possible by genetic engineering. Several cytokines represent accepted therapeutical vehicles, others are still under investigation. To date the following indications are established part of therapy: Correction of renal anaemia by erythropoietin as well as interferon for…
