Ultrastructural features in the structure of blood capillaries of the Haversian canal.

Electron microscopy was used to study osteon capillaries of the compact tissue of human growing long bones. The capillaries were found to be of the continuous type. On the lumenal surface of endothelial cells of the arterial capillaries there were a great number of micropinocytotic vesicles as well as a lot of branching cytoplasmic processes forming a characteristic spongioid layer. Vesicles and filopodia were considerably fewer on the basal surface of the cells. The endothelial cells of the venous capillaries had a small number of micropinocytotic vesicles and single cytoplasmic projections o

Relapsing polychondritis: an ultrastructural study.

Ear cartilage from a typical case of relapsing polychondritis was examined with the electron microscope. A large number of dense granules and vesicles, which were compatible with matrix vesicles or lysosomes, surrounded the affected chondrocytes. In less severely damaged chondrocytes, these granules and vesicles appeared to be formed by pinching off of the cytoplasmic processes or by budding from the processes. Calcification of the granules was minimal. In severely damaged chondrocytes, an admixture of these granules and cytoplasmic organelles occurred. It is speculated that many of these dens

Entamoeba histolytica: ultrastructure of trophozoites recovered from experimental liver lesions.

Ultrastructural studies on Entamoeba histolytica have been carried out mostly with trophozoites cultured for many years. Under these conditi … sed in amebas obtained from liver lesions 15 min and 6h after inoculation. On the other hand, large cytoplasmic vesicles with a microfibrillar content appeared in trophozoites cultured from 72 or 96 h hepatic lesions. With fluorescence microscopy, a chitin-like material was identified in these vesicles by reactivity with calcofluor M2R. Ultrastructurally, these cytopl …

Dilemma-Solving PIC Vesicles: Ligand-Driven Assembly of Stable yet Degradable Nanocarriers for Cytosolic Protein Delivery.

Polyion complex (PIC) vesicles represent a promising platform for protein therapeutics delivery, yet balancing extracellular stability with intracellular degradability remains challenging. Herein, we present a ligand-engineering strategy to fabricate dual-functional PIC vesicles. The design centers on a disulfide bridged bis-dipicolinic acid ligand, L2(S-S), which chelates with Gd3+ at a stoichiometric ratio of 1:3 to form anionic branched coordination polymers. These polymers coassemble with cationic PEG113-b-PMETAC73 diblock copolymers, creating uniform vesicles capable of in situ protein lo

Mesothelial intercellular junctions and pathways.

Freeze-etch preparations of mesothelial cells taken from the peritoneum of mouse reveal the presence of vesicles invaginating the apical and the basal cell surfaces. These vesicles are scarcely seen within the cytoplasm. Long tortuous tubular profiles extend for considerable distance within the cytoplasm and are frequently associated with the vesicles. The possible nature and role of the vesicles and the tubules in transport phenomena across the mesothelial barrier, are discussed in relation to the pore theory advanced by physiologists and the "stomata" concept observed by early German and con

Light, fluorescence and electron microscopic studies of rabbit subclavian glomera.

The subclavian glomera (aortic bodies) of young New Zealand white rabbits were studied with the light, fluorescence, and electron microscopes. Two cell types were identified: type I, granule-containing (chief) cells, and type II, agranular (sustentacular) cells. The type I cells possessed large nuclei, the normal complement of cytoplasmic organelles and numerous electron-opaque cytoplasmic granules. The type II cells were agranular with attenuated cytoplasmic processes which partially or completely ensheathed the type I cells. The glomera were well vascularized. Capillary endothelial cells con

Redox-sensitive stimulation of type-1 ryanodine receptors by the scorpion toxin maurocalcine.

The scorpion toxin maurocalcine acts as a high affinity agonist of the type-1 ryanodine receptor expressed in skeletal muscle. Here, we investigated the effects of the reducing agent dithiothreitol or the oxidizing reagent thimerosal on type-1 ryanodine receptor stimulation by maurocalcine. Maurocalcine addition to sarcoplasmic reticulum vesicles actively loaded with calcium elicited Ca²⁺ release from native vesicles and from vesicles pre-incubated with dithiothreitol; thimerosal addition to native vesicles after Ca²⁺ uptake completion prevented this response. Maurocalcine enhanced equilibrium

Perturbation of lymphatic endothelial cells in experimental Brugia malayi infections.

Infection with filarial nematodes induces lymphatic dysfunction in many individuals in endemic areas. The present morphometric study describes changes in lymphatic endothelial cells of Brugia-infected cats. Transmission electron microscopy revealed a decrease in the number of cytoplasmic vesicles in endothelial cells from lymphatic vessels harboring filarial nematodes (4.9 vesicles per micron 2 cytoplasm) when compared with similar cells from the contralateral uninfected control vessels (9.0 vesicles per micron 2). The size of the vesicles and their location within the cytoplasm was not change…

Single capillaries in the frog mes … s the endothelial cells, i.e. the percentage of lumenal vesicles labelled was greater than that for cytoplasmic vesicles which in turn was greater than that for vesicles at the ablumenal surface. 7. Whereas the labelling of lumenal vesicles from zero time up to 16-20 sec, the main increase in labelling of cytoplasmic vesicles occurred between 10 and 20 sec. 8. It is c …

SYPL1 defines a vesicular pathway essential for sperm cytoplasmic droplet formation and male fertility.

The cytoplasmic droplet is a conserved dilated area of cytoplasm situated at the neck of the sperm flagellum. Viewed as residual cytoplasm inherited from late spermatids, the cytoplasmic droplet contains numerous saccular elements as its key content. However, the origin of these saccules and the function of the cytoplasmic droplet have long been speculative. Here, we identify the molecular origin of these cytoplasmic droplet components by uncovering a vesicle pathway essential for formation and sequestration of saccules within the cytoplasmic droplet. This process is governed by a transmembran

Actin nucleator formins regulate the tension-buffering function of caveolin-1.

Both the mechanosensitive actin cytoskeleton and caveolae contribute to active processes such as cell migration, morphogenesis, and vesicular trafficking. Although distinct actin components are well studied, how they contribute to cytoplasmic caveolae, especially in the context of mechano-stress, has remained elusive. Here, we identify two actin-associated mobility stereotypes of caveolin-1 (CAV-1)-marked intracellular vesicles, which are characterized as 'dwelling' and 'go and dwelling'. In order to exploit the reason for their distinct dynamics, elongated actin-associated formin functions ar

Electron microscopic localization of lactate dehydrogenase in osteoclasts of chick embryo tibia.

Lactate dehydrogenase (LDH) was localized in osteoclasts of fixed and unfixed 19-day chick embryo tibias using a copper ferrocyanide capture … t they contained material from the bone surface. These results suggest that the LDH associated with cytoplasmic vesicles of the osteoclasts may be important in processing of material resorbed from the bone surface and that osteoclasts mitochondria may utilize lactate from the bone fluid for energy production.

Astroglial vesicular network: evolutionary trends, physiology and pathophysiology.

Intracellular organelles, including secretory vesicles, emerged when eukaryotic cells evolved some 3 billion years ago. The primordial organelles that evolved in Archaea were similar to endolysosomes, which developed, arguably, for specific metabolic tasks, including uptake, metabolic processing, storage and disposal of molecules. In comparison with prokaryotes, cell volume of eukaryotes increased by several orders of magnitude and vesicle traffic emerged to allow for communication between distant intracellular locations. Lysosomes, first described in 1955, a prominent intermediate of endo- an

Early ultrastructural changes of myocardial endothelial cells in traumatic haemorrhagic shock in rats.

The aim of this study was to assess early traumatic haemorrhagic shock (THS)-induced changes in myocardial endothelial cells (mECs) both mor … ytoplasmic processes, and interendothelial junctions--and determined the number and distribution of cytoplasmic vesicles in every mEC. THS induced a significantly increased formation of cytoplasmic processes and a redistribution of cytoplasmic vesicles towards the cell centre, with the changes peaking between 15 and 60 min. There was no evidence of an increase in mEC …

Identification and proteomic profiling of exosomes in human urine.

Urine provides an alternative to blood plasma as a potential source of disease biomarkers. One urinary biomarker already exploited in clinical studies is aquaporin-2. However, it remains a mystery how aquaporin-2 (an integral membrane protein) and other apical transporters are delivered to the urine. Here we address the hypothesis that these proteins reach the urine through the secretion of exosomes [membrane vesicles that originate as internal vesicles of multivesicular bodies (MVBs)]. Low-density urinary membrane vesicles from normal human subjects were isolated by differential centrifugatio

Quantitative studies of rat carotid body type I cells.

The general structure and results of quantitative studies of rat carotid body type I cells are described. In contrast to previous reports, there was no change in mitochondrial V/v% on stimulating the carotid body with 10% oxygen. The volume of cytoplasm occupied by electron-dense cored vesicles was significantly increased, whilst their density per square micrometre of cytoplasm was decreased during hypoxia. Thus, the size of vesicles is increased by hypoxic stimulation. On the basis of vesicle diameter and density we were unable to find evidence of more than one variety of type I cell.

Differentiation of Entamoeba histolytica: a possible role for enolase.

The study of the encystation process of Entamoeba histolytica has been hampered by the lack of experimental means of inducing mature cysts in vitro. Previously we have found that cytoplasmic vesicles similar to the encystation vesicles of Entamoeba invadens are present in E. histolytica trophozoites only in amebas recovered from experimental amebic liver abscesses. Here we report that a monoclonal antibody (B4F2) that recognizes the cyst wall of E. invadens also identifies a 48 kDa protein in vesicles of E. histolytica trophozoites recovered from hepatic lesions. This protein is less expressed…
