In vitro and in vivo cardiac toxicity of flavored electronic nicotine delivery systems.

The usage of flavored electronic nicotine delivery systems (ENDS) is popular, specifically in the teen and young adult age-groups. The possible cardiac toxicity of the flavoring aspect of ENDS is largely unknown. Vaping, a form of electronic nicotine delivery, uses "e-liquid" to generate "e-vapor," an aerosolized mixture of nicotine and/or flavors. We report our investigation into the cardiotoxic effects of flavored e-liquids. E-vapors containing flavoring aldehydes such as vanillin and cinnamaldehyde, as indicated by mass spectrometry, were more toxic in HL-1 cardiomyocytes than fruit-flavore…

Is Maintenance or Switching Between Freebase and Nicotine Salt Liquid Associated with Electronic Nicotine Delivery Systems Dependence?

BACKGROUND: Electronic nicotine delivery systems (ENDS) use one of two formulations of nicotine-freebase or nicotine salt. This study examines whether maintenance or switching between nicotine formulations is associated with ENDS dependence using longitudinal survey data. METHODS: 543 U.S. adults (21+) using ENDS frequently (5+ days/week) self-reported and uploaded photos of their most used ENDS liquids in wave 3-5 online surveys from September 2021 to April 2023. Nicotine formulation from photo data was used if available; otherwise, self-reported data were used. ENDS dependence was measured …

Toxicity of humectants propylene glycol and vegetable glycerin in electronic nicotine delivery systems.

BACKGROUND: E-cigarettes and other electronic nicotine delivery systems (ENDs) remain a significant public health risk. Although deployed as tobacco smoking cessation tools, e-cigarettes have gained greater popularity among non-smokers, specifically adolescents and young adults. Previous research has focused primarily on the toxicities associated with nicotine, flavorings, and other chemicals generated from e-cigarette liquid aerosolization; however, little attention has been given to the two primary and most abundant chemicals found in most e-cigarette liquids - propylene glycol (PG) and vege…

Electronic Nicotine Delivery Systems (ENDS) use Among Members of a Community Engagement Program.

Electronic nicotine delivery systems (ENDS) are relatively new and ENDS use data from community engagement programs may help us understand usage patterns and facilitate targeted longitudinal studies. Community members in Florida, USA, were asked about ENDS use, tobacco use, and health history/concerns by Community Health Workers. Among 7253 members recruited during 2014 to 2021 into our HealthStreet program, 1177 had ever used ENDS; the proportion increased from 12 to 27% from 2014 to 2021 (adjusted odds ratio (aOR) 2.5; 95% CI 1.7-3.5; Ever versus never used ENDS). Ever tobacco use was strong…

Electronic nicotine delivery systems use predicts transitions in cigarette smoking among young adults.

BACKGROUND: Although cigarette smoking during young adulthood is characterized by volatility, few studies examine if use of electronic nicotine delivery systems (ENDS) impacts transitions in cigarette use behaviors across this developmental period. The purpose of this longitudinal study was to examine the role of ENDS use on three transitions in cigarette smoking among young adults; initiation, desistance, and re-uptake. METHODS: Participants were 5029 18-29-year-olds (64.2% female) enrolled in one of 24 Texas colleges at baseline and involved in an eight-wave, 4.5-year study. A multi-state, …

Use of Electronic Nicotine Delivery Systems among Adults with Mental Health Conditions, 2015.

Adults with mental health conditions (MHC) are especially likely to smoke and experience tobacco-related health disparities. Individuals with MHC may also use electronic nicotine delivery devices (ENDS) at disproportionately high rates. However, there is a relative dearth of knowledge regarding ENDS use among individuals with MHC. In a large representative sample of U.S. adults (n = 6051), associations between self-reported MHC diagnoses and ENDS use and susceptibility were examined, stratified by smoking status. Participants with MHC were approximately 1.5 times more likely to have used ENDS

Assessment of nicotine pharmacokinetics and abuse liability in randomized, crossover studies of Vuse Alto electronic nicotine delivery systems.

INTRODUCTION: Understanding the neuropharmacological effects and abuse liability (AL) of electronic nicotine delivery systems (ENDS) is essential for evaluating their potential role in tobacco harm reduction (THR). Nicotine delivered via ENDS engages central nervous system (CNS) pathways involved in reinforcement, craving, and dependence. This research characterized nicotine pharmacokinetics (PK) and AL of Vuse Alto ENDS across multiple nicotine concentrations and flavor and contextualized these findings relative to combustible cigarettes and nicotine replacement therapy (NRT) gum. METHODS: T…

Uncovering the Dual Effect of Menthol and Nicotine Levels in Electronic Nicotine Delivery Systems on Pulmonary Surfactant Function.

Electronic nicotine delivery systems (ENDS) are now increasingly used, with commercial electronic cigarettes frequently containing high levels of nicotine and menthol, which is a popular flavoring agent. This has raised multiple concerns about the health risks associated with menthol-flavored ENDS. Although menthol and nicotine are known for their individual effects on respiratory health, their combined impact on pulmonary surfactants remains poorly understood. Therefore, this study aimed at understanding the interactions between the primary components of all ENDS liquids (PG and VG), nicotine…

Modeling drug exposure in rodents using e-cigarettes and other electronic nicotine delivery systems.

Smoking tobacco products is the leading cause of preventable death worldwide. Coordinated efforts have successfully reduced tobacco cigarette smoking in the United States; however, electronic cigarettes (e-cigarette) and other electronic nicotine delivery systems (ENDS) recently have replaced traditional cigarettes for many users. While the clinical risks associated with long-term ENDS use remain unclear, advancements in preclinical rodent models will enhance our understanding of their overall health effects. This review examines the peripheral and central effects of ENDS-mediated exposure to …

Does a 10-puff bout reflect natural puffing patterns for electronic nicotine delivery systems?

INTRODUCTION: An important research gap is how users of electronic nicotine delivery systems (ENDS) group puffs together, and whether that pattern accurately reflects the standardized 10-puff procedure often employed in laboratory-based work. Previously defined "puff cluster" parameters were applied to data from our own work and then the patterns compared to those published elsewhere. METHODS: For all studies (N=8), the same definition of a puff cluster was used (number of puffs separated by <60 seconds), as was the same cluster categorizations: 1 puff (single), 2-5 puffs (short), 6-10 puffs …

Negative Affect and Cigarette Cessation in Dual Users of Cigarettes and Electronic Nicotine Delivery Systems.

Objectives: Dual use of cigarettes and electronic nicotine delivery systems (ENDS) is increasingly common in adult smokers, who often report using ENDS to quit smoking. Elevated negative affect is an established predictor of increased difficulty quitting smoking combustible cigarettes but has not yet been examined in the context of cigarette cessation for dual users. Method: This study examined whether mood-related factors predict cigarette smoking cessation among dual users (N = 364) over 12-months. Self-reported cigarette smoking at 12 months, with abstinence defined as no smoking for the pa…

Tobacco Use, Electronic Nicotine Delivery Systems, and Vulnerable Populations: Current Landscape and Opportunities for Improvement.

The tobacco epidemic has claimed countless lives, caused significant morbidity, and cost billions of dollars in direct costs and lost productivity. Despite its acute vascular effects, nicotine alone has not been definitively linked to cardiovascular events. Rather, additives found in cigarettes and other tobacco products likely play a bigger role in tobacco's link to cardiovascular events. The emergence of electronic nicotine delivery systems introduces a new challenge, particularly among certain groups. Understanding the groups vulnerable to tobacco product use, identifying factors that influ…
