Protocol for the isolation, culturing, and evaluation of primary Müller glial cells from pig retina as cellular models in health and disease.

Müller glial cells maintain retinal homeostasis and provide structural and metabolic support. The pig eye closely resembles the human eye in morphology and physiology, offering strong translational potential. Here, we present a protocol for isolating and culturing primary Müller glial cells from pig retina to study their function in health and disease. We describe steps for eyeball dissection, retinal isolation, photoreceptor cell depletion, and retinal digestion. We then detail procedures for the selection, culture, and characterization of Müller glial cells.

Inhibition of Foxp4 Disrupts Cadherin-based Adhesion of Radial Glial Cells, Leading to Abnormal Differentiation and Migration of Cortical Neurons in Mice.

Heterozygous loss-of-function variants of FOXP4 are associated with neurodevelopmental disorders (NDDs) that exhibit delayed speech development, intellectual disability, and congenital abnormalities. The etiology of NDDs is unclear. Here we found that FOXP4 and N-cadherin are expressed in the nuclei and apical end-feet of radial glial cells (RGCs), respectively, in the mouse neocortex during early gestation. Knockdown or dominant-negative inhibition of Foxp4 abolishes the apical condensation of N-cadherin in RGCs and the integrity of neuroepithelium in the ventricular zone (VZ). Inhibition of

Characterization of Retinal Development in 13-Lined Ground Squirrels.

PURPOSE: The cone-dominant, 13-lined ground squirrel (13-LGS) retina mimics the human central retina, but a thorough examination of retinal development in this species has not been reported. Here, the embryonic and postnatal development of the 13-LGS retina was studied to further characterize 13-LGS as a practical alternative animal model for investigating cone-based vision in health and disease. METHODS: The spatiotemporal expression of key progenitor and cell type markers was examined in retinas from defined embryonic and postnatal stages using immunohistochemistry. Postnatal gene expressio

Müller Glia maintain their regenerative potential despite degeneration in the aged zebrafish retina.

Ageing is a significant risk factor for degeneration of the retina. Müller glia cells (MG) are key for neuronal regeneration, so harnessing the regenerative capacity of MG in the retina offers great promise for the treatment of age-associated blinding conditions. Yet, the impact of ageing on MG regenerative capacity is unclear. Here, we show that the zebrafish retina undergoes telomerase-independent, age-related neurodegeneration but that this is insufficient to stimulate MG proliferation and regeneration. Instead, age-related neurodegeneration is accompanied by MG morphological aberrations an

Crosstalk Between Microglia and Müller Glia in the Age-Related Macular Degeneration: Role and Therapeutic Value of Neuroinflammation.

Age-related macular degeneration (AMD) is a progressive neurodegeneration disease that causes photoreceptor demise and vision impairments. In AMD pathogenesis, the primary death of retinal neurons always leads to the activation of resident microglia. The migration of activated microglia to the ongoing retinal lesion and their morphological transformation from branching to ameboid-like are recognized as hallmarks of AMD pathogenesis. Activated microglia send signals to Müller cells and promote them to react correspondingly to damaging stimulus. Müller cells are a type of neuroglia cells that ma

Photoprotective Mechanism of Fucoxanthin in Ultraviolet B Irradiation-Induced Retinal Müller Cells Based on Lipidomics Analysis.

Long-term exposure to sunlight and/or blue light causes vision damage to people of all ages. Dietary pigments and polyphenols have been shown to have photoprotective potential for eyes; however, many unknowns regarding the protective mechanism remain. In this study, we used ultraviolet B (UVB) irradiation-induced retinal Müller cells (RMCs) to screen for dietary polyphenols and pigment compounds with effective photoprotective activity. Fucoxanthin (FX) was shown to have the best therapeutic effect, and the mechanism was evaluated via lipidomics analysis. Both intra- and extracellular ROS, mito

Tmem216 deficiency induces hydrocephalus via impaired ciliogenesis and disrupted ependymal planar polarity in mice.

Defects in multiciliated ependymal cells (ECs), which line the cerebral ventricular walls and generate unidirectional cerebrospinal fluid (CSF) flow through coordinated ciliary beating, can lead to CSF accumulation and hydrocephalus. TMEM216, a ciliopathy-associated gene mutated in Joubert, Meckel, and related syndromes, encodes an essential component of the protein complexes that form the transition zone (TZ) at the base of primary cilia. Here, we demonstrate that central nervous system-specific inactivation of Tmem216 results in severe postnatal hydrocephalus. TMEM216 localizes to the TZ of

[The role of Müller cells in the formation and development of macular hole].

Müller cells are important glial cells in the retina, which play important roles in maintaining the stability of the retina by mechanical support, homeostasis, and physiological metabolism, as well as protecting photoreceptor cells and retinal pigment epithelial cells. The degeneration and destruction of Müller cells are often accompanied by various retinal diseases, and the function of Müller cells is changed under pathological conditions. Based on the summary of the morphology, distribution and function of Müller cells, this article analyzes the different manifestations and changes of Müller

Emerging roles of brain tanycytes in regulating blood-hypothalamus barrier plasticity and energy homeostasis.

Seasonal changes in food intake and adiposity in many animal species are triggered by changes in the photoperiod. These latter changes are faithfully transduced into a biochemical signal by melatonin secreted by the pineal gland. Seasonal variations, encoded by melatonin, are integrated by third ventricular tanycytes of the mediobasal hypothalamus through the detection of the thyroid-stimulating hormone (TSH) released from the pars tuberalis. The mediobasal hypothalamus is a critical brain region that maintains energy homeostasis by acting as an interface between the neural networks of the cen

The Properties and Functions of Glial Cell Types of the Hypothalamic Median Eminence.

The median eminence (ME) is part of the neuroendocrine system (NES) that functions as a crucial interface between the hypothalamus and pituitary gland. The ME contains many non-neuronal cell types, including oligodendrocytes, oligodendrocyte precursor cells (OPCs), tanycytes, astrocytes, pericytes, microglia and other immune cells, which may be involved in the regulation of NES function. For example, in mice, ablation of tanycytes (a special class of ependymal glia with stem cell-like functions) results in weight gain, feeding, insulin insensitivity and increased visceral adipose, consistent w

Transcytosis of tanycytes in the circumventricular organs of adult mouse brain.

Tanycytes are specialized ependymal cells lining the ventricular spaces of the adult brain and thereby provide an interface between the cerebrospinal fluid (CSF) and brain parenchyma. They act as energy homeostasis, neuroendocrine regulation, and CSF-brain barrier; however, their functional significance in CSF-brain communication currently remains unknown. In the present study, we investigated the presence of tanycytic transcytosis using fluorescent tracers; a GM1 ligand, cholera toxin B (CTB), and a mannose-6-phosphate/insulin-like growth factor-Ⅱ receptor ligand, wheat germ agglutinin (WGA).

REM phase: An ingenious mechanism to enhance clearance of metabolic waste from the retina.

The Rapid Eye Movement (REM) phase of sleep, also known as "active sleep" because of physiological similarities to waking state, is characterized by intense cerebral electrical activity, propensity to dream vividly and suppression of skeletal muscle activity (atonia) except for the extraocular muscles which give rise to the so-called REM. In 1998 David Maurice, an ophthalmologist, proposed that REM sleep was associated with an eye function: it would be required to stir the anterior chamber and bath it with aqueous humor to prevent corneal anoxia during sleep. However, potential metabolic probl

Müller Cell Metabolic Signatures: Evolutionary Conservation and Disruption in Disease.

Müller cells are glia that play important regulatory roles in retinal metabolism. These roles have been evolutionarily conserved across at least 300 million years. Müller cells have a tightly locked metabolic signature in the healthy retina, which rapidly degrades in response to insult and disease. This variation in metabolic signature occurs in a chaotic fashion, involving some central metabolic pathways. The cause of this divergence of Müller cells, from a single class with a unique metabolic signature to numerous separable metabolic classes, is currently unknown and illuminates potential al

cxcl18b-defined transitional state-specific nitric oxide drives injury-induced Müller glia cell-cycle re-entry in the zebrafish retina.

In lower vertebrates, retinal Müller glia (MG) exhibit a life-long capacity of cell-cycle re-entry to regenerate neurons following the retinal injury. However, the mechanism driving such injury-induced MG cell-cycle re-entry remains incompletely understood. Combining single-cell transcriptomic analysis and in vivo clonal analysis, we identified previously undescribed cxcl18b-defined MG transitional states as essential routes toward MG proliferation following green/red cone (G/R cone) ablation. Inflammation blockage abolished the triggering of these transitional states, which expressed the gene

Müller glia-microglia cross talk reprograms the Müller glia transcriptome for cell division-related processes during retina regeneration.

In the zebrafish retina, Müller glia (MG) respond to retinal injury by dividing and producing a multipotent progenitor for retinal repair. This cell division is regulated by microglia; however, the underlying mechanism remains unknown. Here, we report that MG-derived Il34 attracts microglia to sites of retinal injury where they stimulate MG proliferation via the release of cytokines, like M17, Spp1, Tnfa, and Tnfb. Remarkably, RNA sequencing analysis of MG's regeneration-associated transcriptome with and without microglia depletion suggests microglia stimulate MG proliferation by preferentiall

Enhanced cortical neural stem cell identity through short SMAD and WNT inhibition in human cerebral organoids facilitates emergence of outer radial glial cells.

Cerebral organoids exhibit broad regional heterogeneity accompanied by limited cortical cellular diversity despite the tremendous upsurge in derivation methods, suggesting inadequate patterning of early neural stem cells (NSCs). Here we show that a short and early Dual SMAD and WNT inhibition course is necessary and sufficient to establish robust and lasting cortical organoid NSC identity, efficiently suppressing non-cortical NSC fates, while other widely used methods are inconsistent in their cortical NSC-specification capacity. Accordingly, this method selectively enriches for outer radial g

Fate bias during neural regeneration adjusts dynamically without recapitulating developmental fate progression.

BACKGROUND: Regeneration of neurons in the central nervous system is poor in humans. In other vertebrates neural regeneration does occur efficiently and involves reactivation of developmental processes. Within the neural retina of zebrafish, Müller glia are the main stem cell source and are capable of generating progenitors to replace lost neurons after injury. However, it remains largely unknown to what extent Müller glia and neuron differentiation mirror development. METHODS: Following neural ablation in the zebrafish retina, dividing cells were tracked using a prolonged labelling technique

Müller glia in short-term dark adaptation of the Austrolebias charrua retina: Cell proliferation and cytoarchitecture.

Fish with unique life cycles offer valuable insights into retinal plasticity, revealing mechanisms of environmental adaptation, cell proliferation, and thus, potentially regeneration. The variability of the environmental factors to which Austrolebias annual fishes are exposed has acted as a strong selective pressure shaping traits such as nervous system plasticity. This has contributed to adaptation to their extreme conditions including the decreased luminosity as ponds dry out. In particular, the retina of A. charrua has been shown to respond to 30 days of decreased luminosity by exacerbating

m6A epitranscriptomic modification regulates neural progenitor-to-glial cell transition in the retina.

N 6-methyladenosine (m6A) is the most prevalent mRNA internal modification and has been shown to regulate the development, physiology, and pathology of various tissues. However, the functions of the m6A epitranscriptome in the visual system remain unclear. In this study, using a retina-specific conditional knockout mouse model, we show that retinas deficient in Mettl3, the core component of the m6A methyltransferase complex, exhibit structural and functional abnormalities beginning at the end of retinogenesis. Immunohistological and single-cell RNA sequencing (scRNA-seq) analyses of retinogene

Cellular components of the idiopathic epiretinal membrane.

Idiopathic epiretinal membrane (iERM) is a fibrocellular proliferation on the inner surface of the retina, which leads to decreased visual acuity and even central visual loss. As iERM is associated to advanced age and posterior vitreous detachment, a higher prevalence is expected with increasing life expectancy and aging of the global population. Although various cell types of retinal and extra-retinal origin have been described in iERMs (Müller glial cells, astrocytes, hyalocytes, retinal pigment epithelium cells, myofibroblasts, and fibroblasts), myofibroblasts have a central role in collage
