Phylogenetic analysis of Erwinia species based on 16S rRNA gene sequences.

The phylogenetic relationships of the type strains of 16 Erwinia species were investigated by performing a comparative analysis of the sequences of the 16S rRNA genes of these organisms. The sequence data were analyzed by the neighbor-joining method, and each branch was supported by moderate bootstrap values. The phylogenetic tree and sequence analyses confirmed that the genus Erwinia is composed of species that exhibit considerable heterogeneity and form four clades that are intermixed with members of other genera, such as Escherichia coli, Klebsiella pneumoniae, and Serratia marcescens. Clus…

Transcriptome Analysis Reveals Cross-Kingdom Virulence Factors in Erwinia persicina Cp2.

Erwinia persicina is a well-documented plant pathogenic bacterium, causing soft rot in various plant hosts. There are rare previous reports that it is associated with animal diseases. In this study, we used an alfalfa root infection model and a mouse model via gavage administration of E. persicina strain Cp2. Strain Cp2 was pathogenic on both alfalfa and mice. On alfalfa, Cp2 primarily caused leaf spot and etiolation. Mice exhibited moderate lesions in their liver, stomach and kidney tissues, with inflammation in the liver and kidney after gavage. Furthermore, viable Cp2 cells were successfull…

The Erwinia chrysanthemi type III secretion system is required for multicellular behavior.

Enterobacterial animal pathogens exhibit aggregative multicellular behavior, which is manifested as pellicles on the culture surface and biofilms at the surface-liquid-air interface. Pellicle formation behavior requires production of extracellular polysaccharide, cellulose, and protein filaments, known as curli. Protein filaments analogous to curli are formed by many protein secretion systems, including the type III secretion system (TTSS). Here, we demonstrate that Erwinia chrysanthemi, which does not carry curli genes, requires the TTSS for pellicle formation. These data support a model wher…

Genomic diversity of Erwinia carotovora subsp. carotovora and its correlation with virulence.

We used genetic and biochemical methods to examine the genomic diversity of the enterobacterial plant pathogen Erwinia carotovora subsp. carotovora. The results obtained with each method showed that E. carotovora subsp. carotovora strains isolated from one ecological niche, potato plants, are surprisingly diverse compared to related pathogens. A comparison of 23 partial mdh sequences revealed a maximum pairwise difference of 10.49% and an average pairwise difference of 2.13%, values which are much greater than the maximum variation (1.81%) and average variation (0.75%) previously reported for …

[PEGylated recombinant L-asparaginase from Erwinia carotovora: Production, properties, and potential applications].

N-hydroxysuccinimide ester of monomethoxy polyethylene glycol hemisuccinate was synthesized. It acylated amino groups in a molecule of recombinant L-asparaginase from Erwinia carotovora. A method of L-asparaginase modification by the obtained activated polyethylene glycol derivative was developed. The best results were produced by modification of the enzyme with a 25-fold excess of reagent relative to the enzyme tetramer. The modified L-asparaginase was isolated from the reaction mixture by gel filtration on Sepharose CL-6B. The purified bioconjugate did not contain PEG unbound to the protein,…

Identification of a chromosome-borne class C beta-lactamase from Erwinia rhapontici.

OBJECTIVES: To characterize the beta-lactamase gene content of Erwinia rhapontici. METHODS: The beta-lactamase gene was cloned, sequenced and expressed in Escherichia coli. RESULTS: The cloned gene conferred a resistance pattern of an Ambler class C beta-lactamase in E. coli. The AmpC-type enzyme had a pI value of 8.6 and shared 62% amino acid sequence identity with that of Escherichia fergusonii. The ampC gene was associated with a regulatory ampR gene and beta-lactamase production was inducible. CONCLUSIONS: This work provides further evidence of the molecular heterogeneity of beta-lactam…

Structural and functional role of Gly281 in L-asparaginase from Erwinia carotovora.

L-asparaginases (E.C.3.5.1.1, L- ASNases) have been widely used in clinical practice as chemotherapeutic drugs of acute lymphoblastic leukaemia (ALL). In order to evaluate the structural and functional role of selected residues in ASNases we report the screening of a library of L-ASNase mutants aiming to find detrimental mutations that significantly affect catalysis and substrate specificity. The library of mutants was created using the staggered extension process (StEp) and the genes of L- ASNases from Erwinia chrysanthemi (ErL-ASNase) and Erwinia carotovora (EcaL-ASNase). A mutant that displ…

Characterization of ferrioxamine E as the principal siderophore of Erwinia herbicola (Enterobacter agglomerans).

Several strains of the enterobacterial group Erwinia herbicola (Enterobacter agglomerans) were screened for siderophore production. After 3 days of growth in a low-iron medium, all strains studied produced hydroxamate siderophores. The retention values of the main siderophore during thin-layer chromatography on silica gel plates and on HPLC reversed-phase columns were identical with those of an authentic sample of ferrioxamine E (norcardamine). Gas-chromatographic analysis of the HI hydrolyzate yielded succinic acid and 1,5-diaminopentane in equimolar amounts; fast-atom-bombardment (FAB) mass …

Assessment of PEG-Asparaginase and Erwinia Asparaginase Activity Under Variable Storage Conditions.

Background: PEG-Asparaginase and Erwinia asparaginase are enzyme-based anticancer therapies used in the treatment of acute lymphoblastic leukaemia (ALL) and lymphoblastic lymphoma (LBL), where adequate plasma enzyme activity is required for therapeutic efficacy. In many study groups, therapeutic drug monitoring is routinely applied due to pharmacokinetic variability and the risk of hypersensitivity reactions followed by increased clearance and insufficient treatment. In clinical practice, samples may be exposed to prolonged transport and variable pre-analytical conditions. Knowledge on pre-ana…

Peptidylarginine deiminase inhibitor treatment is associated with increased phage susceptibility in Erwinia amylovora.

Understanding the phage resistance mechanisms in Erwinia amylovora is crucial for improving phage therapy applications targeting this plant pathogen, which is responsible for fire blight disease in apple and pear orchards. This study investigated E. amylovora phage resistance in the presence of peptidylarginine deiminase (PAD) inhibitors, which have been found in other studies to prevent the conversion of arginine to citrulline as a post-translational modification that may initiate outer membrane vesical (OMV) formation. Infection of the E. amylovora Ea6-4 strain by ϕEa21-4 phages, in the pres…

Erwinia amylovora bacteriophages as biological agents for targeted and sustainable fire blight management.

Fire blight, caused by the Gram-negative bacterium Erwinia amylovora, is one of the most destructive diseases of pome-fruit crops worldwide. Despite decades of research, its management remains challenging because efficient and sustainable control options are limited, and many commercially important apple and pear cultivars remain highly susceptible. Conventional treatments for bacterial disease suppression are often hindered by inconsistent efficacy, phytotoxic effect, environmental concerns and regulatory restrictions, necessitating new sustainable alternative options. Virulent bacteriophages…

Successful treatment with Erwinia L-asparaginase for recurrent natural killer/T cell lymphoma.

We describe a patient with natural killer (NK)/T cell lymphoma who relapsed after autologous peripheral blood stem cell transplantation (auto-PBSCT) and was successfully treated with Escherichia coli (E. coli) and Erwinia L-asparaginase. A 38-year-old male patient with ulcerated tumor at the left thigh was diagnosed as having nasal type NK/T cell lymphoma on the basis of histopathological and flowcytometric findings of tumor, revealing diffuse infiltration of atypical lymphoid cells into blood vessels and expression of CD7 and CD56 antigens, but not CD3. He had tumor infiltration in the bone m…

Identification of a global repressor gene, rsmA, of Erwinia carotovora subsp. carotovora that controls extracellular enzymes, N-(3-oxohexanoyl)-L-homoserine lactone, and pathogenicity in soft-rotting Erwinia spp.

The production of extracellular enzymes such as pectate lyase (Pel), polygalacturonase (Peh), cellulase (Cel), and protease (Prt) is activated by the cell density (quorum)-sensing signal, N-(3-oxohexanoyl)-L-homoserine lactone (HSL); plant signals; and aep genes during postexponential growth of Erwinia carotovora subsp. carotovora 71. Studies with mutants of E. carotovora subsp. carotovora 71 derepressed in exoenzyme production led to the identification of a negative regulator gene, rsmA (rsm, repressor of secondary metabolites). Nucleotide sequencing, transcript assays, and protein analysis e…

A comprehensive strategy to address shortage of Erwinia asparaginase in pediatric acute lymphoblastic leukemia.

BACKGROUND: Pegylated form of E. coli derived asparaginase (PEG) is a crucial component of pediatric ALL therapy. Patients who develop a hypersensitivity (HSR) reaction with PEG receive an alternative form - Erwinia asparaginase (EA). However, an international shortage in 2017 had made it challenging to treat these patients. We have developed a comprehensive strategy to address this need. PATIENTS AND METHODS: This is a single center, retrospective analysis. All patients receiving PEG were premedicated to reduce infusion reactions. Patients who developed HSR underwent PEG desensitization. Pat…

Application of phytosynthesized silver nanoparticles (SNPs) against Erwinia amylovora causing fire blight disease.

The bacterium Erwinia amylovora is responsible for the destructive disease known as fire blight in pear trees. This highly detrimental condition poses a significant threat to the health and vitality of these trees. The existing strategies for managing fire blight disease involve the regular use of copper compounds and streptomycin, particularly during periods when environmental factors are conducive to the spread of the infection. Silver nanoparticles, also known as SNPs, are tiny specks of silver ranging in size from 10 to 100 nm. These particles are created through various chemical and biolo…

Exopolysaccharides favor the survival of Erwinia amylovora under copper stress through different strategies.

Erwinia amylovora causes fire blight, a destructive disease of rosaceous plants very difficult to control. We demonstrated that copper, employed to control plant diseases, induces the "viable-but-nonculturable" (VBNC) state in E. amylovora. Moreover, it was previously reported that copper increases production of its main exopolysaccharide (EPS), amylovoran. In this work, the copper-complexing ability of amylovoran and levan, other major EPS of E. amylovora, was demonstrated. Following this, EPS-deficient mutants were used to determine the role of these EPSs in survival of this bacterium in AB …

Design, Synthesis, Antibacterial Evaluation, and Mechanistic Insights of Garlic-Derived Disulfides against Erwinia amylovora.

Erwinia amylovora, the causal agent of fire blight, poses a severe and ongoing threat to the global production of Rosaceous crops. We synthesized 54 garlic-derived disulfides and identified compound b as the most potent candidate (EC50 = 2.85 μg/mL), which significantly surpassed allicin and zhongshengmycin (ZSM). In planta assays on Cuiguan pear demonstrated that b provided exceptional curative and protective efficacy, effectively suppressing fire blight infections in flowers, leaves, shoots, and fruits. Mechanistically, b triggered lethal ROS accumulation by inhibiting catalase (CAT) and sup…

Inhibitory activity and mechanism of aryl furan thioesters containing 1,3,4-thiadiazole against Erwinia amylovora.

BACKGROUND: The Gram-negative bacterium Erwinia amylovora induces fire blight disease in Rosaceae hosts, severely damaging pome fruit trees including pear, apple, and hawthorn cultivars. Traditional control methods, such as antibiotics and copper-based agents, are associated with plant damage and the emergence of resistant bacterial strains, and present significant challenges. This study aims to design and synthesize a novel eco-friendly bactericide. RESULTS: In this study, we designed and synthesized 30 small-molecule compounds by combining 1,3,4-thiadiazole, a key moiety of zinc thiazole, w…

Lignin bioconversion based on genome mining for ligninolytic genes in Erwinia billingiae QL-Z3.

BACKGROUND: Bioconversion of plant biomass into biofuels and bio-products produces large amounts of lignin. The aromatic biopolymers need to be degraded before being converted into value-added bio-products. Microbes can be environment-friendly and efficiently degrade lignin. Compared to fungi, bacteria have some advantages in lignin degradation, including broad tolerance to pH, temperature, and oxygen and the toolkit for genetic manipulation. RESULTS: Our previous study isolated a novel ligninolytic bacterial strain Erwinia billingiae QL-Z3. Under optimized conditions, its rate of lignin degr…
