Myophosphorylase deficiency (glycogen storage disease Type V) in a herd of Charolais cattle in New Zealand: confirmation by PCR-RFLP testing.

AIM: To describe a disease of muscle in Charolais calves and confirm the putative diagnosis of inherited myophosphorylase deficiency. METHODS: Variously stained paraffin sections of muscle prepared from affected calves were used to describe the lesions. A polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) test was developed and applied to affected calves, their sires, dams and other individuals. RESULTS: The lesions were those of rhabdomyolysis of skeletal muscles and sub-sarcolemmal spaces in normal fibres. The PCRRFLP test confirmed the expected mutation for phos

McArdle disease (Glycogen Storage Disease Type V) from an exercise physiology and biochemistry perspective: Important considerations for exercise and physical activity testing and data interpretation.

BACKGROUND: Traditionally, the management and research of inborn errors of metabolism have been primarily overseen by genetic specialists, whose expertise has guided both therapeutic strategies and efforts to identify potential cures. In more recent times, exercise scientists and exercise physiologists have been shown to provide important insights into these conditions from new perspectives. Because exercise and physical activity testing is often used to determine the main outcome variables for interventions designed to improve the lives of those with inborn errors of metabolism, it is logical

New perspectives for the treatment and follow-up of glycogen storage disease type V: DL-3-hydroxybutyric acid with modified Atkins diet and quadriceps femoris shear wave elastography.

OBJECTIVES: Glycogen storage disease type V is caused by the mutations in muscle glycogen phosphorylase gene. This is the first report which DL-3-hydroxybutyric acid was used in combination with modified Atkins diet for the treatment of a patient with glycogen storage disease type V and quadriceps femoris shear wave elastography was performed to evaluate the treatment efficacy. CASE PRESENTATION: A 13-year-old girl was referred with fatigue and muscle cramps with exercise and there were no pathological findings in physical examination. Creatine kinase levels with 442 U/L. No phosphorylase enz…

[Clinical and biochemical correlations in certain metabolic myopathies].

Muscular glycogenosis is a disease resulting from genetical abnormalities altering an enzyme which is involved in glycogen metabolism. In addition to disorders of glycogenosis and glycolysis, there are other pathological processes such as alpha-glycosidase deficiency and diseases associated with abnormal polysaccharide structure. A short review of the various diseases with their particular features is reported.

[Differential diagnosis of glycogenoses].

Different types of hepatic, muscular, and generalized forms of glycogen stogare disease, a hereditary disease caused by glycogen metabolism disorders, are analysed. The clinical and biochemical features of these diseases are described. The data on enzyme defects and methods for their detection in different types of glycogenoses as well as on the prevalence of the disease in different countries are presented.

Data from the European registry for patients with McArdle disease and other muscle glycogenoses (EUROMAC).

BACKGROUND: The European registry for patients with McArdle disease and other muscle glycogenoses (EUROMAC) was launched to register rare muscle glycogenoses in Europe, to facilitate recruitment for research trials and to learn about the phenotypes and disseminate knowledge about the diseases through workshops and websites. A network of twenty full and collaborating partners from eight European countries and the US contributed data on rare muscle glycogenosis in the EUROMAC registry. After approximately 3 years of data collection, the data in the registry was analysed. RESULTS: Of 282 patient
