A physiologically based pharmacokinetic model, which is an extension of the Bischoff-Dedrick multiorgan model, was developed to described the kinetics of barbiturates (hexobarbital, phenobarbital, and thiopental) in the rat. The model is composed of 11 organ or tissue compartments. The brain compartment was featured as a nonflow-limited organ for some low lipid soluble barbiturates. Michaelis-Menten constants for drug metabolism (Km, Vmax) were determined from in vitro experiments using liver microsomes. Ethanol, hexobarbital, chlorpromazine and haloperidol had no effect on the composition and total amount of gangliosides in rat and mouse brains. The gangliosides seemed to be resistant to ethanol and the above mentioned drugs. None of them either in single or repeated doses had a major effect on the sialic acid-containing glycosphingolipids. The results of this study are at variance with those reported by Klemm & Foster (Life Sci., 1986, 39, 897-902) who described a sharp decrease of the total level of brain gangliosides after injection of a single dose of ethanol.

Studies were conducted in an effort to determine if a circadian rhythm in response of mice to hexobarbital could be evaluated employing dose-response curves. Results showed a circadian change in both the position and slope of the dose-response curves when evaluated at 0800, 1400, 2000 and 0200 hours. Glucocorticoids (GCs) are used to treat numerous diseases, but their use in limited by adverse side effects. One such effect is occasional increased anxiety. Since the intensity of hepatic microsomal oxidation has been shown to alter responses to GC, we examined the possibility that rats with lower rates of hepatic GC metabolism would have increased anxiety. The effects of lithium and disulfiram on the 5-hydroxytryptamine (5-HT) system and hexobarbital (HX)-induced hypnosis were studied. Treatment with lithium significantly prolonged HX hypnosis. Disulfiram, a potent inhibitor of brain aldehyde dehydrogenase, also produced a prolongation of HX hypnosis.
