Histamine release and therapy of severe dermatographism.

A case of unusually severe dermatographism is reported in which baseline plasma histamine levels were repeatedly elevated and histamine release into the circulation was demonstrable upon challenge by scratching the skin. Passive transfer was positive when the patient was challenged after a 48-hr interval, and the responsible factor was heat labile consistent with an IgE-dependent mechanism. Five additional patients with severe dermatographism had elevated baseline plasma histamine levels, but an augmented response upon stroking the skin was not evident. The response of this latter group to ant…

Histamine release in dogs by Emulphor EL620.

A vehicle containing ethanol and Emulphor EL620 lowers blood pressure and increases heart rate in morphine-chloralose anesthetized dogs. These effects are associated with histamine release caused by Emulphor EL620.

Effect of adenosine and histamine receptor stimulation on canine histamine release to pentagastrin.

The effects of adenosine and histamine 2 and histamine 3 receptor agonists on the regulation of gastric histamine release were examined in anesthetized mixed-breed dogs. All compounds were infused directly into the gastrosplenic artery to avoid perturbations in systemic hemodynamics, and the gastric histamine release was stimulated with pentagastrin. The histamine concentration in plasma samples was measured utilizing gas chromatography-negative-ion chemical ionization mass spectroscopy. Pentagastrin consistently stimulated gastric histamine release with the peak stimulation occurring at 5 min…

Histamine release by morphine and diamorphine in man.

Intravenous morphine and diamorphine are routinely used for postoperative analgesia but the relative histamine releasing abilities of these … pain scores were recorded before and after analgesic administration, and a series of eight basophil histamine release studies was also performed. Significant histamine release (plasma concentration > 2 ng.ml-1 or rise of > 700% baseline) occurred in 23.5% of the morphine group and 21.1% of the diamorphine group. Histamine was released earlier in those receiving dia …

The inhibition of histamine release by antiallergic drugs.

Three new antiallergic drugs, Doxantrazole, PRD-92 and N5', as well as disodium cromoglycate, inhibited the IgE-mediated PCA reaction in the rat triggered by the homologous antigen, but did not have an antagonistic effect on histamine itself. Moreover, all the drugs examined caused in vitro inhibition of antigen-mediated histamine release from peritoneal mast cells and chopped lung tissue of sensitized rats producing IgE antibodies. Doxantrazole had a synergistic effect on the inhibition of histamine release by isoproterenol, whereas the other drugs were devoid of this capacity. PRD-92 and N5'…

Histamine release and contrast media-induced renal vasoconstriction.

RATIONALE AND OBJECTIVES: The authors' purpose was to investigate the role of histamine release causing renal vasoconstriction induced by application of contrast media, an important element in contrast medium-induced nephrotoxicity. MATERIALS AND METHODS: Isometric contractions in rabbit segmental renal arteries stimulated with KCl and increasing concentrations of the ionic contrast medium diatrizoate and the nonionic agents iomeprol and iodixanol were studied both with and without increasing concentrations of the histamine H1 and H2 blockers diphenhydramine and cimetidine. Histamine concentr…

Exercise-induced allergies: the role of histamine release.

Exercise is a physical cause of allergic reactions, including exercise-induced anaphylaxis (EIAna), exercise-induced urticaria (EIU), exercise-induced asthma (EIA), and exercise-induced rhinitis (EIR). Since its first description in 1979, EIAna has been reported with variable clinical manifestations, with exercise alone, and in combination with food ingestion. Elevated serum histamine levels and cutaneous mast cell degranulation have been noted. Exercise-induced urticaria appears as small, punctate lesions that differ from the classic coalescent type seen with EIAna. Variant forms of EIAna wit

Theophylline reduces histamine release during pollen-induced rhinitis.

In an attempt to understand how theophylline achieves its in vivo therapeutic effect, a double-blind crossover study of the effect of theophylline on the immediate response of the upper airways to challenge with antigen was performed. Ten subjects with allergic rhinitis were challenged with increasing doses of antigen extract, and their responses were assessed by counting the number of sneezes and measuring the level of histamine, N-alpha-p-tosyl-L-arginine-methyl ester-esterase activity, and kinin in nasal secretions. One-week premedication with theophylline led to serum levels in the therape

Gatifloxacin-induced histamine release and hyperglycemia in rats.

Gatifloxacin, a fluoroquinolone antimicrobial agent, has been reported to cause both hypoglycemia and hyperglycemia in diabetic and non-diabetic patients. The purpose of the present study was to investigate the mechanism of gatifloxacin-induced hyperglycemia in normal and diabetic rats. Rats received a single intravenous injection of gatifloxacin and samples of their arterial blood were collected periodically. Diabetic rats were produced by the injection of streptozotocin and nicotinamide. In normal rats, the concentration of serum glucose decreased after the injection of gatifloxacin at 50mg/

Histamine-releasing factor enhances food allergy.

Food allergy occurs due to IgE- and mast cell-dependent intestinal inflammation. Previously, we showed that histamine-releasing factor (HRF), a multifunctional protein secreted during allergy, interacts with a subset of IgE molecules and that the HRF dimer activates mast cells in an HRF-reactive IgE-dependent manner. In this study, we investigated whether HRF plays any role in food allergy. Specifically, we determined that prophylactic and therapeutic administration of HRF inhibitors that block HRF-IgE interactions reduces the incidence of diarrhea and mastocytosis in a murine model of food al

Histamine-releasing factor and immunoglobulins in asthma and allergy.

Factors that can induce the release of histamine from basophils have been studied for more than 30 years. A protein termed histamine-releasing factor (HRF) was purified and molecularly cloned in 1995. HRF can stimulate histamine release and IL-4 and IL-13 production from IgE-sensitized basophils and mast cells. HRF-like activities were found in bodily fluids during the late phase of allergic reactions, implicating HRF in allergic diseases. However, definitive evidence for the role of HRF in allergic diseases has remained elusive. On the other hand, we found effects of monomeric IgE on the surv…

Release of creatine kinase by antigen, histamine, and catecholamines.

In mepyramine treated, ovalbumin sensitized anaesthetized guinea pigs protracted anaphylactic shock was produced by i.p. injection of antigen, and serum creatine kinase (CK) activities were determined 4, 6, or 17 hrs thereafter. Significant increases above nonsensitized controls were obtained. In nonanaesthetized guinea pigs shock course and serum CK increase were considerably accelerated. Histamine increased the serum CK only when given in high amounts (10 mg/kg) s.c., in the presence or absence of mepyramine. I.p. injection of histamine in mepyramine treated animals had no effect. Adrenaline

Histamine release in vitro: inhibition by catecholamines and methylxanthines.

Methylxanthines and catecholamines both inhibit antigenically induced histamine release from human leukocytes. They act synergistically to inhibit the reaction, but must be present when antigen is added; preincubation is not effective. Since both increase cellular levels of cyclic 3 ', 5'-adenosine monophosphate it is postulated that this compound plays a role in the regulation of allergic histamine release.
