Locally Injectable Hydrogels for Tumor Immunotherapy.

Hydrogel-based local delivery systems provide a good delivery platform for cancer immunotherapy. Injectable hydrogels can directly deliver antitumor drugs to the tumor site to reduce systemic toxicity and achieve low-dose amplification immunotherapy. Therefore, it may overcome the problems of low drug utilization rate and the systemic side effects in cancer immunotherapy through systemic immune drugs, and it provides simple operation and little invasion at the same time. This study aimed to review the research progress of injectable hydrogels in tumor immunotherapy in recent years. Moreover, t

Interconnected Toroidal Hydrogels for Islet Encapsulation.

Islet encapsulation and transplantation promises to improve upon current treatments for type 1 diabetes mellitus, though several limitations remain. Macroscale devices have been designed for in vivo transplantation and retrieval, but traditional geometries do not support clinically adequate mass transfer of nutrients to and insulin from the encapsulated tissue. Microcapsule technologies have improved mass transfer properties, but their clinical translation remains challenging as their complete retrieval is difficult, should the graft become a safety concern. Here, the design, characterization

Tunable antibiotic delivery from gellan hydrogels.

Hydrogels are used extensively in wound management. Many wounds are highly susceptible to infection and hydrogels can provide localized antibacterial delivery to treat and prevent this infection. There are several key considerations in designing antibacterial hydrogels for wound therapy, including preserving activity of encapsulated antibacterial agents, controlling drug release timescales and concentrations, and having the ability to conform to various wound configurations. In this work, we have used gellan, a U.S. Food and Drug Administration approved food additive, to develop antibiotic loa

Biocompatibility of injectable resilin-based hydrogels.

Vocal folds are connective tissues housed in the larynx, which can be subjected to various injuries and traumatic stimuli that lead to aberrant tissue structural alterations and fibrotic-induced biomechanical stiffening observed in patients with voice disorders. Much effort has been devoted to generate soft biomaterials that are injectable directly to sites of injury. To date, materials applied toward these applications have been largely focused on natural extracellular matrix-derived materials such as collagen, fibrin or hyaluronic acid; these approaches have suffered from the fact that mater

Polymeric hydrogels for oral insulin delivery.

The search for an effective and reliable oral insulin delivery system has been a major challenge facing pharmaceutical scientists for over many decades. Even though innumerable carrier systems that protect insulin from degradation in the GIT with improved membrane permeability and biological activity have been developed, yet a clinically acceptable device is not available for human application. Efforts in this direction are continuing at an accelerated speed. One of the preferred systems widely explored is based on polymeric hydrogels that protect insulin from enzymatic degradation in acidic s

Supramolecular Mechanophore Additives Enable Mechanochromic Hydrogels.

Mechanochromic hydrogels that report deformation through optical signals require mechanophores that function in aqueous environments, are activated at low force, and allow integration into the hydrogel matrix. Here, we report a new strategy to address these challenges that involves deploying a supramolecular mechanophore as a water-soluble macromolecular additive. The mechanophore developed for this purpose features a loop-forming architecture, in which a naphthalene diimide (NDI) quencher and a 9,10-bis(phenylethynyl)anthracene (BPEA) fluorophore are connected by a short spacer. Intramolecula

Electrically Conductive Hydrogels for Wound Healing.

SIGNIFICANCE: Wound healing is a complex, tightly regulated process involving a range of enzymes, growth factors, and cytokines that coordinate cellular activities essential for tissue repair and wound closure. However, in cases of extensive or severe injury, the intrinsic repair mechanisms are often insufficient, underscoring the need for advanced therapeutic strategies to accelerate healing and minimize scar formation. RECENT ADVANCES: Electrically conductive hydrogels (ECHs), combining the advantageous properties of hydrogels with the physiological and electrochemical characteristics of co

Antifouling Activity of Bottlebrush Network Hydrogels.

Mitigating the attachment of microorganisms to polymer biomaterials is critical for preventing hospital-acquired infections. Two chemical strategies to mitigate fouling include fabricating fouling-resistant surfaces, which typically present hydrophilic polymers, such as polyethylene glycol (PEG), or creating fouling-release surfaces, which are generally hydrophobic featuring polydimethylsiloxane (PDMS). Despite the demonstrated promise of employing PEG or PDMS, amphiphilic PEG/PDMS copolymer materials remain understudied. Here, for the first time, we investigated if phase-separated amphiphilic

Designed modular protein hydrogels for biofabrication.

Designing proteins that fold and assemble over different length scales provides a way to tailor the mechanical properties and biological performance of hydrogels. In this study, we designed modular proteins that self-assemble into fibrillar networks and, as a result, form hydrogel materials with novel properties. We incorporated distinct functionalities by connecting separate self-assembling (A block) and cell-binding (B block) domains into single macromolecules. The number of self-assembling domains affects the rigidity of the fibers and the final storage modulus G' of the materials. The mech

Advances in Hydrogels for Periodontitis Treatment.

Periodontitis is a common condition characterized by a bacterial infection and the disruption of the body's immune-inflammatory response, which causes damage to the teeth and supporting tissues and eventually results in tooth loss. Current therapy involves the systemic and local administration of antibiotics. However, the existing treatments cannot exert effective, sustained release and maintain an effective therapeutic concentration of the drug at the lesion site. Hydrogels are used to treat periodontitis due to their low cytotoxicity, exceptional water retention capability, and controlled dr

Strong, tough and anisotropic bioinspired hydrogels.

Soft materials are widely used in tissue engineering, soft robots, wearable electronics, etc. However, it remains a challenge to fabricate soft materials, such as hydrogels, with both high strength and toughness that are comparable to biological tissues. Inspired by the anisotropic structure of biological tissues, a novel solvent-exchange-assisted wet-stretching strategy is proposed to prepare anisotropic polyvinyl alcohol (PVA) hydrogels by tuning the macromolecular chain movement and optimizing the polymer network. The reinforcing and toughening mechanisms are found to be "macromolecule crys

Polysaccharide-based hydrogels for cartilage regeneration.

Cartilage defect is one of the common tissue defect clinical diseases and may finally lead to osteoarthritis (OA) which threat patients' physical and psychological health. Polysaccharide is the main component of extracellular matrix (ECM) in cartilage tissue. In the past decades, polysaccharide-based hydrogels have shown great potential for cartilage regeneration considering unique qualities such as biocompatibility, enhanced cell proliferation, drug delivery, low toxicity, and many others. Structures such as chain length and chain branching make polysaccharides have different physical and che

Poroviscoelasto-plasticity of agarose-based hydrogels.

Agarose gels are excellent candidates for tissue engineering as they are tunable, viscoelastic, and show a pronounced strain-stiffening response. These characteristics make them ideal to create in vitro environments to grow cells and develop tissues. As in many other biopolymers, viscoelasticity and poroelasticity coexist as time-dependent behaviors in agarose gels. While the viscoelastic behavior of these hydrogels has been considered using both phenomenological and continuum models, there remains a lack of connection between the underlying physics and the macroscopic material response. Recent Advances in Antimicrobial Peptide Hydrogels.

Advances in the number and type of available biomaterials have improved medical devices such as catheters, stents, pacemakers, prosthetic joints, and orthopedic devices. The introduction of a foreign material into the body comes with a risk of microbial colonization and subsequent infection. Infections of surgically implanted devices often lead to device failure, which leads to increased patient morbidity and mortality. The overuse and improper use of antimicrobials has led to an alarming rise and spread of drug-resistant infections. To overcome the problem of drug-resistant infections, novel

Preparation of DNA-crosslinked polyacrylamide hydrogels.

Mechanobiology is an emerging scientific area that addresses the critical role of physical cues in directing cell morphology and function. For example, the effect of tissue elasticity on cell function is a major area of mechanobiology research because tissue stiffness modulates with disease, development, and injury. Static tissue-mimicking materials, or materials that cannot alter stiffness once cells are plated, are predominately used to investigate the effects of tissue stiffness on cell functions. While information gathered from static studies is valuable, these studies are not indicative o

Release of drugs from polymeric hydrogels.

Hydrogels were prepared from 2-hydroxyethyl methacrylate and ethylene glycol dimethacrylate as cross-linking agent. Two different polymerization techniques were followed, namely bulk polymerization initiated by dibenzoyl peroxide and solution polymerization, in water, using a redox initiator system. Differences in the water desorption from swollen samples between the two types of the above products were recorded and attributed to the fact that those hydrogels were obtained in glassy or rubbery state, according to the polymerization technique. Also, the delivery of salicylic acid from samples o

Cellulose Hydrogels Derived from Pineapple Bagasse for Potential Dental Applications: Chlorhexidine-Loaded Hydrogels with Antibacterial and Cytocompatible Properties.

Pineapple fibers were used as a sustainable raw material to isolate native cellulose from alkaline-acid treatment. The cellulose fibers were regenerated into transparent and flexible cellulose hydrogels using the lithium chloride/N,N-dimethylacetamide (LiCl/DMAc) solvent system, followed by a phase-inversion process of the cellulose solution under ethanol vapor. Chlorhexidine was incorporated into the hydrogels to provide antibacterial properties. The concentration of chlorhexidine ranged from 0.1 to 0.8 wt%. The prepared hydrogels showed better early onset cytocompatibility than the cell cult
