Lysosomes in retinal health and disease.

Lysosomes play crucial roles in various cellular processes - including endocytosis, phagocytosis, and autophagy - which are vital for maintaining retinal health. Moreover, these organelles serve as environmental sensors and act as central hubs for multiple signaling pathways. Through communication with other cellular components, such as mitochondria, lysosomes orchestrate the cytoprotective response essential for preserving cellular homeostasis. This coordination is particularly critical in the retina, given its high metabolic rate and susceptibility to photo-oxidative stress. Consequently, im…

Lysosomes and essential fatty acid deficiency.

The hydrolytic activity usually associated with lysosomes increased in the homogenates and subcellular fractions of rat liver as a result of essential fatty acid (EFA) deficiency. The proportion of the total (tissue homogenate) activity found in each subcellular fraction, however, was unchanged by EFA deficiency.Lysosomes isolated from normal and EFA-deficient rat livers differed significantly in their stability to thermal and osmotic variations. This suggested that lysosomal membranes, like other membranes, were altered by EFA deficiency. In spite of increased tissue-bound hydrolytic activity …

Peripheral positioning of lysosomes supports melanoma aggressiveness.

Emerging evidence suggests that the function and position of organelles are pivotal for tumor cell dissemination. Among them, lysosomes stand out as they integrate metabolic sensing with gene regulation and secretion of proteases. Yet, how their function is linked to their position and how this controls metastasis remains elusive. Here, we analyze lysosome subcellular distribution in patient-derived melanoma cells and patient biopsies and show that lysosome spreading scales with melanoma aggressiveness. Peripheral lysosomes promote matrix degradation and cell invasion which is directly linked …

Tubular lysosomes in rat and gerbil pinealocytes.

Ultracytochemical demonstration of acid phosphatase (AcPase) in pinealocytes of rat and Mongolian gerbil (Meriones unguiculatus) has revealed an existence of tubular lysosomes (30-200 nm in diameter and more than 5 micron long) in their cytoplasm. The tubular lysosomes arise by bulging from GERL cisternae (Golgi apparatus, endoplasmic reticulum, lysosomes) and spread throughout the whole cell body without forming an anastomosing network. Numerous varicosities are characteristic for the tubular lysosomes whose similarity with grumose bodies has lead to conclusion that the vermiculate variety of…

Podocytes degrade endocytosed albumin primarily in lysosomes.

Albuminuria is a strong, independent predictor of chronic kidney disease progression. We hypothesize that podocyte processing of albumin via the lysosome may be an important determinant of podocyte injury and loss. A human urine derived podocyte-like epithelial cell (HUPEC) line was used for in vitro experiments. Albumin uptake was quantified by Western blot after loading HUPECs with fluorescein-labeled (FITC) albumin. Co-localization of albumin with lysosomes was determined by confocal microscopy. Albumin degradation was measured by quantifying FITC-albumin abundance in HUPEC lysates by Weste…

Flux of fatty acids through NPC1 lysosomes.

Niemann-Pick type C (NPC) is an autosomal recessive lipid storage disorder characterized by lysosomal accumulation of cholesterol and gangli … ll division permease transporters. Our results indicate that fatty acid flux through NPC1-deficient lysosomes is normal.

Genetic evidence for transmembrane acetylation by lysosomes.

Acetyl-CoA:alpha-glucosaminide N-acetyltransferase is a lysosomal-membrane enzyme deficient in a genetic disorder, Sanfilippo disease type C. The enzyme catalyzes the transfer of an acetyl group from cytoplasmic acetyl-coenzyme A (acetyl-CoA) to terminal alpha-glucosamine residues of heparan sulfate within the organelle. Previous kinetic experiments indicated that the enzyme carries out a transmembrane acetylation via a ping-pong mechanism; the reaction can therefore be dissected into two half reactions--acetylation of the enzyme, and transfer of the acetyl group to glucosamine. Cells derived

PLGA nanoparticles restore acidic pH and degradative function to compromised lysosomes with Cy3-labeling providing enhanced tracking to lysosomes.

Lysosomal dysfunction and elevated lysosomal pH are hallmark features of age-related neurodegenerative diseases including age-related macula … nine3 amine (Cy3). Nanoparticle uptake was systematically optimized, achieving over 90% delivery to lysosomes of induced pluripotent stem cell-derived retinal pigment epithelial (iPS-RPE) cells, although uptake rates varied among adjacent cells. Once internalized, nanoparticles demonstrated remarkable stability, with no detectable change in concentration, d …

Here, we summarize our current knowledge of the regulation of lysosome by G4 ligands, underlying their potential functional impact on lysosome biology and autophagic flux, as well as on the transcriptional regulation of lysosomal g …

Fibrillar α-synuclein toxicity depends on functional lysosomes.

Neurodegeneration in Parkinson's disease (PD) can be recapitulated in animals by administration of α-synuclein preformed fibrils (PFFs) into … ted uptake of α-synuclein PFFs into cells but that chloroquine did not stop α-synuclein uptake into lysosomes despite impairing lysosomal function and inhibiting α-synuclein toxicity. Together, these data suggested that α-synuclein PFFs are toxic in functional lysosomes in vitro. Therapeutic strategies that prevent α-synuclein fibril uptake into lysosomes m …
