Mercuric chloride-induced apoptosis is dependent on protein synthesis.

Apoptosis is a mode of cell death with morphologic and biochemical features that distinguish it from necrosis. Recent studies demonstrating that mercury compounds initiate apoptosis in cultured cells did not elucidate if the biochemical mechanism of apoptosis involved a dependence on macromolecular synthesis post-insult, i.e. programmed cell death. The objectives of this in vitro study were (1) to determine if HgCl2 cytotoxicity includes an apoptotic component, and (2) to determine if apoptosis is dependent on protein synthesis, i.e. proceeds by an inducible mechanism. Suspensions of mouse lym

Immune-mediated glomerulonephritis induced by mercuric chloride in mice.

The BALB/c mouse developed mesangial deposits of immune constituents and light microscopical changes characteristic of immune complex glomerulonephritis after 8 weeks' treatment with mercuric chloride given by s.c. injection. There were no signs of linear of granular immune deposits along the glomerular capillary basement membrane after 2 or 8 weeks. The antigen could not be identified. No antibodies to nuclear or renal structures were found. Using a histochemical method (silver amplification) mercury was detected by light and electron microscopy in tubular and glomerular structures. Mercury w…

Mercuric chloride-induced renal tubular necrosis in the rat.

A light microscopic study of the renal tubulonecrotic lesion in rats given a small dose of HgCl2 is described. The changes consist of a rapidly developing vacuolation of the cytoplasm with loss of basophilic staining within 4 h that leads to cell breakdown, fragmentation and dissolution by 48 h. Nuclear changes appear to set in later. Permanent patchy fibrotic lesions were found in the kidneys at 10 days. The animals pass a large amount of urine of low osmolarity, low Na+, K+ and Cl- for a period of 3 days accompanied by an increased water intake. Nevertheless there appeared to be no water or

Rhabdomyolysis and renal failure in acute mercuric chloride poisoning.

A case of acute renal failure after mercuric chloride poisoning is reported. Laboratory data revealed markedly elevated serum concentrations of aldolase and creatine phosphokinase, and the presence of pigment granular casts and myoglobin in the urine. The patient went into a diuretic phase after 12 days of oliguria and renal function returned to normal during the next 10 days. Besides direct nephrotoxicity, the contributory role of rhabdomyolysis in the pathogenesis of renal failure after mercuric chloride poisoning, has been stressed.

Experimental study on the estrogen-like effect of mercuric chloride.

Although mercuric chloride has toxicity on reproductive system, it is uncertain if such toxicity is induced by estrogen-like effect. To study whether mercuric chloride has the estrogen-like effect and its relevant mechanism, proliferation assay of MCF-7 human breast cancer cells, uterotrophic assay, peroxidase activity assay and estrogen receptor competitive binding assay were conducted to screen the estrogen-like effect of mercuric chloride. The MCF-7 cells proliferated in the stimulation of mercuric chloride and got to the peak at 10(-7) mol/l concentration. And this proliferation could be c…

DMPS as a rescue agent for the nephropathy induced by mercuric chloride.

The effectiveness of 2,3-dimercaptopropane-1-sulfonate (DMPS) as a rescue agent for the acute nephropathy induced by HgCl2 was studied in uninephrectomized (NPX) and sham-operated (SO) rats. NPX and SO rats that were given a toxic 2.5-mumol/kg dose of HgCl2 developed severe renal damage within 24 hr after the HgCl2 was administered. Renal injury was assessed by measuring plasma creatinine, creatinine clearance, fractional excretion of several biological markers, the rate of excretion of cellular enzymes and plasma solutes and severity of morphologically demonstrable necrosis in the pars recta

The effect of unilateral nephrectomy on the nephrotoxicity of mercuric chloride in the rat.

Unilateral nephrectomy (UNX) induces a dramatic change in single-kidney structure and function. Therefore, the effects of nephrotoxins may be altered. To evaluate this possibility, mercuric chloride (2 mg/kg, sc) was given to male, Sprague-Dawley rats 2 days following either UNX or sham surgery. Nonoliguric acute renal failure developed and was qualitatively similar in both groups. Glomerular filtration rate (GFR) reached a nadir on Day 2 and was reduced to a greater extent in the UNX group. Furthermore, recovery of GFR was slower and occurred to a lesser extent by Day 10 in the animals subjec…

Interaction of potassium dichromate with the nephrotoxins, mercuric chloride and citrinin.

Potassium dichromate enhanced the effects of mercuric chloride or citrinin on renal slice transport of the organic ions p-aminohippurate (PAH) and tetraethylammonium (TEA) under certain experimental conditions. At the doses employed and times studied no effects, or only minimal effects, were observed on renal slice transport when the toxins were tested individually. Potassium dichromate (10 mg/kg) administered subcutaneously (s.c.) in combination with mercuric chloride (4 mg/kg, s.c.) or citrinin 35 or 55 mg/kg administered intraperitoneally (i.p.), resulted in a marked depression of organic i…

Different histochemical findings in the brain produced by mercuric chloride and methyl mercury chloride in rats.

The chemical form of mercury reactive by the histochemical technique was studied by using rats and mice treated with mercuric chloride (HgCl2) and methyl mercury chloride (MeHgCl). Mercury granules were demonstrated histochemically in the brain of HgCl2-treated rats with higher levels of inorganic mercury. However, mercury granules were not demonstrated in the brain of MeHgCl-treated rats in spite of a considerably high organic mercury level. In rats and mice with significant biotransformation of injected MeHg to inorganic mercury, the appearance of the peak inorganic mercury level and mercury…

Flaxseed oil ameliorates mercuric chloride-induced liver damage in rats.

BACKGROUND: Mercury is a relentless pollutant, and its toxicity contributes to significant health problems due to exposure to the environment. The present study has determined the impact of flaxseed oil on mercuric chloride (HgCl2)-mediated hepatic oxidative toxicity in rats. METHODS: Twenty-four healthy male Wistar rats were divided into four groups with six animals in each group. Group-A was the Control group treated with saline; Group-B received 1.0 ml oral dosage of flaxseed oil; Group-C was given 200 µl intraperitoneal injection of HgCl2, and Group-D received 1.0 ml oral dosage of flaxse…

Glutathione monoethyl ester moderates mercuric chloride-induced acute renal failure.

Glutathione (GSH)-dependent reactions are an important cellular defense against ischemic or oxidative injury, although their role in toxin-induced renal cellular injury is less clear. Because of the known sulfhydryl reactivity of mercury (M), we hypothesized that GSH could modify mercuric chloride (MC)-induced acute renal failure (ARF). Therefore, we evaluated the effects of glutathione monoethyl ester (GE), which produces high intrarenal levels of GSH, on the nephrotoxicity of MC. GE treatment in normal rats did not alter their creatinine clearance (CCr), fractional sodium (CNa/CCr) or lysozy…

Methylmercury- and mercuric-chloride-induced alterations in rat epididymal sperm.

Four-week-old male albino rats weighing 70 +/- 5 g were treated intraperitoneally daily with 0, 5 and 10 micrograms methylmercuric chloride (MMC)/kg or 0, 50 and 100 micrograms mercuric chloride (MC)/kg body weight, respectively, over a period of 90 days. Studies were carried out a intermittent intervals, i.e. on days 0, 15, 30, 60 and 90 of the experiment. Gradual decrements in body and epididymal weights were observed from day 30 onwards in both the MMC- and MC-treated groups. Morphological deformations of epididymal epithelium were noted from day 30 onwards in the mercurial-treated groups. …

Amelioration of mercuric chloride-induced acute renal failure by dithiothreitol.

Experiments were conducted to determine if administration of the sulfhydryl reducing agent and metal chelator dithiothreitol (31 mg/kg body wt) alters the development of renal dysfunction in the first 3 hr after injection of mercuric chloride (3 mg/kg). Mercuric chloride alone resulted in elevation of urine flow rate and fractional excretion of solutes within 30 min of injection. In animals injected with dithiothreitol 60 min after mercuric chloride, urine flow rate and fractional excretion of solutes were reduced within 30 min to values intermediate between control and mercuric chloride-treat…

Mercuric chloride nephritis depends on host rather than kidney strain.

Brown-Norway (BN) rats are susceptible to the induction of an autoimmune glomerulonephritis (GN) by HgCl2 while Lewis (LEW) rats are resistant. When a kidney from a LEW rat (nonsusceptible) is transplanted into a binephrectomized (LEW X BN)F1 hybrid (susceptible) then HgCl2 injections into the recipient result in GN developing in the donor kidney. When a kidney from a BN or (LEW X BN)F1 hybrid (susceptible) is transplanted into a nonsusceptible rat, injections of HgCl2 into the recipient do not result in GN in the donor kidney. These observations show that kidneys from nonsusceptible rats func

Diphenyl diselenide potentiates nephrotoxicity induced by mercuric chloride in mice.

Following our long-standing interest in the mechanisms involved in selenium toxicity, the aim of this work was to extend our previous studies to gain a better understanding of mercuric chloride (HgCl₂) + diphenyl diselenide (PhSe)₂ toxicity. Mice received one daily dose of HgCl₂ (4.6 mg kg(-1) , subcutaneously) for three consecutive days. Thirty minutes after the last injection of HgCl₂, mice received a single dose of (PhSe)₂ (31.2 mg kg(-1) , subcutaneously). Five hours after (PhSe)₂ administration, mice were euthanized and δ-aminolevulinate dehydratase, catalase (CAT), glutathione S-transfer…

Protection against mercuric chloride nephrotoxicity by cold exposure in rats.

The influence of cold exposure (+ 1 degree C for 24 h) was studied in rats dosed i.p. with 0, 2.5 or 5.0 mumol HgCl2/kg. Cold exposure of controls caused an increased diuresis and solute elimination, except sodium, with almost no variations in urine alkaline phosphatase (ALP) and gammaglutamyl transferase (GGT). Proximal tubule brush border damage was demonstrated by a marked increase in ALP and GGT in HgCl2-dose animals at room temperature. Cold exposure protected against this kidney damage.
