We describe a novel immuno-scanning electron microscopy (SEM) technique that combines both Tokuyasu's cryosectioning and section SEM methods. In this technique, semithin cryosections, cut according to the Tokuyasu method, were adhered to glass microscope slides, immunostained for bio-molecules of interest and observed by confocal laser scanning microscopy. The same sections were subsequently embedded in epoxy resin and ultrathin sections were cut on an ultramicrotome. These were then observed by SEM using a backscattered electron detector. PURPOSE OF REVIEW: Cryogenic electron microscopy (cryo-EM) resolves the structures of proteins and their complexes under near-native conditions that are often inaccessible to other structural techniques. Cryo-EM has recently contributed significant advances in basic knowledge to the field of coagulation. New paradigms regarding individual domain role in protein-protein interactions have emerged, casting the main reactions of the coagulation cascade in a much-needed structural context. A universal feature of seed plants is that their phloem consists of a continuous sieve-tube system extending throughout the plant, which is highly pressurized due to its sugar content. Light microscopy of granuloma annulare shows mucin deposition with lympho-histiocytic infiltrate. We describe the ultrastructural three-dimensional aspects of a typical case of granuloma with characteristic histopathological findings. At the ultrastructural level, affected collagen bundles and granular mucin deposition were observed. Round cells corresponding to lymphocytes were identified. Bigger oval cells, corresponding to isolated or palisading histiocytes were also found. The ultrastructural aspects overlap with light microscopy and contribute to its iconographic documentation.

OBJECTIVE: To describe the findings on the presbylarynx under scanning electron microscopy.
STUDY DESIGN: Cadaver study.
SETTING: Universidade Estadual Paulista (Botucatu, São Paulo, Brazil).
SUBJECTS AND METHODS: Sixteen vocal folds were removed during necropsies and distributed into 2 age groups: control (n = 8; aged 30-50 years) and elderly (n = 8; aged 75-92 years). The right vocal fold was dissected, fixed in glutaraldehyde 2.5%, and prepared for scanning electron microscopy. The thickness of the epithelium was measured using a scandium morphometric digital program.

Tinea nigra is a rare superficial mycosis caused by Hortaea werneckii. This infection presents as asymptomatic brown to black maculae mostly in palmo-plantar regions. We performed scanning electron microscopy of a superficial shaving of a tinea nigra lesion. The examination of the outer surface of the sample showed the epidermis with corneocytes and hyphae and elimination of fungal filaments. Lichen sclerosus is an acquired inflammatory condition characterized by whitish fibrotic plaques, with a predilection for the genital skin. We performed scanning electron microscopy of the dermis from a lesion of lichen sclerosus. Normal collagen fibers could be easily found in deeper layers of the specimen, as well as the transition to pathologic area, which seems homogenized. With higher magnifications in this transitional area collagen fibers are adherent to each other, and with very high magnifications a pearl chain aspect became evident along the collagen fibers. Hyperplastic carotid bodies from an 80-year-old woman with sustained systemic hypertension were examined ultrastructurally and compared with those from six normal subjects. Hyperplasia involved both elongated sustentacular and Schwann's cells, both of which were more numerous than normal. They formed concentric whorls around a diminished core of chief cells.
