The mutagenic activity of chlorpromazine hydrochloride based on the Ames plate incorporation test and the modified fluctuation test in the presence and absence of liver microsomal enzyme (S9 fraction) and NADPH was determined. Benzonalum mutagenic activity was demonstrated in a dominant lethal test on mice. The late spermatids stage proved to be the most sensitive in this respect. The benzonalum did not evoke any cytogenetic activity, that was shown by counting chromosome aberrations in metaphases of the bone marrow cells in vivo and did not induce gene mutations in tester trains of Salmonella typhimurium in vitro and in vivo (a host-mediated assay).

This paper reports the use of polyurethane foam (PUF) in the sampling of airborne mutagens too volatile to be retained by glass fibre filters. Air was sampled simultaneously with a glass fibre filter and PUF plugs, and the two extracts of the samples were tested for mutagenicity and analysed for polycyclic aromatic hydrocarbons (PAH). Stable, tandem dicentric chromosomes were discovered in two mutant cell colonies resulting from exposure of L5178Y mouse lymphoma cells to chemical mutagens. These unusual dicentrics were present in all metaphase cells examined from these colonies, even after approximately 65 cell generations in culture. Observation of cells in metaphase and anaphase suggests that the interstitial centromere in these dicentrics is non-functional, and that the terminal centromere is solely responsible for their orderly anaphase segregation.
