Tracing the origin of myofibroblasts in kidney fibrosis.

Kidney fibrosis is a key pathological feature of chronic kidney disease (CKD), characterized by the activation and accumulation of αSMA-positive myofibroblasts. The cellular origin of myofibroblasts in kidney fibrosis has been a subject of extensive research and debate. Previous studies suggested that myofibroblasts might arise from various cellular sources through mesenchymal transition processes, including epithelial-mesenchymal transition (EMT) of tubular epithelial cells (TECs), endothelial-mesenchymal transition (EndoMT), and macrophage-mesenchymal transition (MMT) of bone marrow-derived …

Macrophages Transdifferentiate Into Myofibroblasts During Ocular Surface Inflammation.

Ocular surface inflammation and injury often lead to fibrosis and impaired vision. Myofibroblasts directly promote fibrosis by producing excessive extracellular matrix (ECM) proteins and alpha-smooth muscle actin (ɑ-SMA). In addition, macrophages have been shown to promote fibrosis by secreting pro-fibrotic factors to stimulate myofibroblast formation. However, the capacity of macrophages to directly remodel ECM remains understudied. This study utilized a mouse model of corneal injury and fibrosis to determine whether macrophages transform into myofibroblast-like cells during ocular surface in…

Shedding of proangiogenic microvesicles from hypertrophic scar myofibroblasts.

Hypertrophic scars are a common complication of burn injuries and represent a major challenge in terms of prevention and treatment. These scars are characterized by a supraphysiological vascular density and by the presence of pathological myofibroblasts (Hmyos) displaying a low apoptosis propensity. However, the nature of the association between these two hallmarks of hypertrophic scarring remains largely unexplored. Here, we show that Hmyos produce signalling entities known as microvesicles that significantly increase the three cellular processes underlying blood vessel formation: endothelial…

Effect of Keratinocytes on Myofibroblasts in Hypertrophic Scars.

Myofibroblasts play a central role in matrix formation and wound contraction during wound healing and undergo apoptosis at the end of the healing. Hypertrophic scarring is a pathologic condition in which myofibroblasts persist in the tissue. It has been hypothesized that abnormalities in epidermal-dermal crosstalk underlie this pathology. Therefore, in this study, we investigated whether myofibroblasts are affected by keratinocytes. Transforming growth factor beta-induced myofibroblasts (Imyo) and myofibroblasts from hypertrophic scar tissue (Hmyo) were characterized using microarrays. Keratin…

Origin of myofibroblasts and cellular events triggering fibrosis.

Renal fibrosis is a major hallmark of chronic kidney disease that is considered to be a common end point of various types of renal disease. To date, the biological meaning of fibrosis during the progression of chronic kidney diseases is unknown and possibly depends on the cell type contributing to extracellular matrix production. During the past decade, the origin of myofibroblasts in the kidney has been intensively investigated. Determining the origins of renal myofibroblasts is important because these might account for the heterogeneous characteristics and behaviors of myofibroblasts. Curren…

Myofibroblasts: trust your heart and let fate decide.

Cardiac fibrosis is a substantial problem in managing multiple forms of heart disease. Fibrosis results from an unrestrained tissue repair process orchestrated predominantly by the myofibroblast. These are highly specialized cells characterized by their ability to secrete extracellular matrix (ECM) components and remodel tissue due to their contractile properties. This contractile activity of the myofibroblast is ascribed, in part, to the expression of smooth muscle α-actin (αSMA) and other tension-associated structural genes. Myofibroblasts are a newly generated cell type derived largely from…

Myofibroblasts and extracellular matrix glycoproteins in palmar fibromatosis.

We investigated immunohistochemically the presence of myofibroblasts and their surrounding extracellular matrix (ECM) in palmar fibromatosis nodules, obtained from 30 patients. Histologically, the three following phases were identified in palmar fibromatosis nodules: proliferative, involutional, and residual. A variable association of these phases was simultaneously detected within the same nodule. In most cases (20/30), we found that involutional and residual phases coexisted, while the association of proliferative/involutional/residual and proliferative/involutional phases was observed in 8 …

Calcium-Signalling in Human Glaucoma Lamina Cribrosa Myofibroblasts.

Glaucoma is one of the most common causes of treatable visual impairment in the developed world, affecting approximately 64 million people worldwide, some of whom will be bilaterally blind from irreversible optic nerve damage. The optic nerve head is a key site of damage in glaucoma where there is fibrosis of the connective tissue in the lamina cribrosa (LC) extracellular matrix. As a ubiquitous second messenger, calcium (Ca2+) can interact with various cellular proteins to regulate multiple physiological processes and contribute to a wide range of diseases, including cancer, fibrosis, and gla

Origin and function of myofibroblasts in the liver.

Liver fibrosis contributes to many of the devastating complications of viral, toxic, fatty, and cholestatic liver disease. Understanding the cell populations that promote liver fibrosis and the molecular pathways through which they operate is essential for the development of antifibrotic therapies. The authors review the origins and functions of hepatic myofibroblasts, focusing on hepatic stellate cells, the main contributors to organ fibrosis, and portal fibroblasts, an insufficiently characterized population that may have a specialized function in promoting periductular fibrosis, but a limit…

Myofibroblasts and colonic anastomosis healing in Wistar rats.

BACKGROUND: The myofibroblasts play a central role in wound healing throughout the body. The process of wound healing in the colon was evaluated with emphasis on the role of myofibroblasts. METHODS: One hundred male Wistar rats weighing 274 ± 9.1 g (mean age: 3.5 months) were used. A left colonic segment was transected and the colon was re-anastomosed. Animals were randomly divided into two groups. The first group experimental animals (n = 50) were sacrificed on postoperative day 3, while the second group rats (n = 50) were sacrificed on postoperative day 7. Healing of colonic anastomosis was…

Stromal myofibroblasts predict disease recurrence for colorectal cancer.

PURPOSE: Myofibroblasts, which are specifically differentiated fibroblasts, are thought to play a central role in the desmoplastic reaction, a dynamic stromal change closely associated with cancer development. Although fundamental studies suggest that myofibroblasts may either facilitate or inhibit cancer progression, cumulative evidence supports their role in promoting tumor progression. The aim of this study was to assess the value of myofibroblasts in the cancer stroma as an indicator of disease recurrence after colorectal cancer surgery. EXPERIMENTAL DESIGN: Using computer-assisted image …

The myofibroblast in Dupuytren's contracture.

Dupuytren's contracture nodules, but not cords, contain myofibroblasts. These cells, which combine many electron microscopic, physiologic, and immunohistochemical characteristics of fibroblasts and smooth muscle cells, are probably the active force of contraction. Prominent myofibroblasts and intracellular microtubules correlate with increased likelihood of clinical recurrence after surgery. Tissue culture of cells derived from Dupuytren's contracture myofibroblasts show consistently slower cell replication than from fibroblasts and show persistence of electron microscopic characteristics in e…

The myofibroblast and the anophthalmic socket.

One of the greatest advances in the understanding of wound healing was the identification and characterization of the myofibroblast by Gabbiani in 1971. Since that time this contractile cell has been found in the early stages of wound healing and in many pathologic states. In a recent study, the myofibroblast was found in healing and contracting anophthalmic sockets.

Antiproliferative treatment of myofibroblasts prevents arrhythmias in vitro by limiting myofibroblast-induced depolarization.

AIMS: Cardiac fibrosis is associated with increased incidence of cardiac arrhythmias, but the underlying proarrhythmic mechanisms remain incompletely understood and antiarrhythmic therapies are still suboptimal. This study tests the hypothesis that myofibroblast (MFB) proliferation leads to tachyarrhythmias by altering the excitability of cardiomyocytes (CMCs) and that inhibition of MFB proliferation would thus lower the incidence of such arrhythmias. METHODS AND RESULTS: Endogenous MFBs in neonatal rat CMC cultures proliferated freely or under control of different dosages of antiproliferativ

Targeting Myofibroblasts as a Treatment Modality for Dupuytren Disease.

PURPOSE: Currently, no treatment corrects the contractile nature of Dupuytren myofibroblasts (DMFs) or prevents recurrence following surgery. Antifibrotic and proadipogenic growth factors are released when adipose-derived stem cells (ASCs) are cultured with platelet-rich plasma (PRP), a platelet concentration from whole blood. Reprograming myofibroblasts into adipocytes via growth factors is proposed as a powerful potential tool to target fibrosis. We aimed to assess whether the combination of ASCs and PRP reprograms DMFs into adipocytes in vitro and alters their contractile nature in vivo. M…

Origin and role of hepatic myofibroblasts in hepatocellular carcinoma.

Hepatocellular carcinoma (HCC) is the most common primary liver cancer and is the second leading cause of cancer-related death worldwide. Fibrosis and cirrhosis are important risk factors for the development of HCC. Hepatic myofibroblasts are the cells responsible for extracellular matrix deposition, which is the hallmark of liver fibrosis. It is believed that myofibroblasts are predominantly derived from hepatic stellate cells (HSCs), also known as Ito cells. Nevertheless, depending on the nature of insult to the liver, it is thought that myofibroblasts may also originate from a variety of ot…

Origin of myofibroblasts in the fibrotic liver in mice.

Hepatic myofibroblasts are activated in response to chronic liver injury of any etiology to produce a fibrous scar. Despite extensive studies, the origin of myofibroblasts in different types of fibrotic liver diseases is unresolved. To identify distinct populations of myofibroblasts and quantify their contribution to hepatic fibrosis of two different etiologies, collagen-α1(I)-GFP mice were subjected to hepatotoxic (carbon tetrachloride; CCl4) or cholestatic (bile duct ligation; BDL) liver injury. All myofibroblasts were purified by flow cytometry of GFP(+) cells and then different subsets ide…
