Physiological significance of neurotensin in pituitary glycoprotein hormone release as revealed by in vivo and in vitro studies with neurotensin antiserum.

The neuropeptide, neurotensin, is localized to neurons within the hypothalamus which project to the median eminence. It is released from the terminals in the median eminence into the hypophyseal portal vessels and is carried to the gland. The content in the pituitary gland cells may be partly related to the delivery of the peptide via the portal vessels, but it also appears to be produced directly in pituitary cells. The peptide has actions on the release of glycoprotein hormones from the pituitary and in the present experiments, we attempted to determine whether these actions of the peptide w…

Distribution of neurotensin in the canine gastrointestinal tract.

The distribution of immunoreactive neurotensin in the canine gastrointestinal tract from the esophagus to the rectum, as well as in the pancreas, was determined by a specific neurotensin radioimmunoassay. Immunoreactive neurotensin was found throughout the gastrointestinal tract and in the pancreas. The highest concentrations of immunoreactive neurotensin were found in the mucosal extracts of the jejunum (422 +/- 68 ng/gm) and ileum (3025 +/- 289 ng/gm). Small but substantial amounts of immunoreactive neurotensin were found in the esophagus, fundus (includes fundus and corpus), antrum, duodenu…

The role of neurotensin in human gallbladder motility.

Gallbladder contraction in response to a fatty meal is thought to be caused by release of cholecystokinin (CCK). We have previously demonstrated a close correlation between circulating concentrations of CCK and contraction of the gallbladder in normal humans and in gallstone patients. Recent studies in animals, however, have shown that other potentially cholecystokinetic hormonal agents are released by a fatty meal, which suggests that other hormones may be involved in postprandial gallbladder contraction. Neurotensin, a 13-amino acid peptide, is released by fat; we have shown it to cause gall…

Plasma neurotensin in term and preterm neonates.

The new ileal peptide neurotensin has not been studied hitherto in the human neonate. Plasma concentrations of neurotensin were measured by radioimmunoassay in 276 healthy term or preterm infants either at birth or pre- or post-prandially during the neonatal period. In addition, a group of 10 6-day-old preterm infants were studied who had never been enterally fed on account of hyaline membrane disease. Plasma neurotensin values were obtained also in 12 healthy fasting adults. Term infants had higher plasma neurotensin concentration than preterm infants at birth. Both groups showed a significan…

Hyperglycemic effect of neurotensin, a hypothalamic peptide.

A hypotensive, gut-contracting peptide neurotensin (NT), recently isolated from bovine hypothalami, has been found to produce hyperglycemia within minutes after iv injection into anesthetized rats. The dose-response relationship (deltaglucose, 15 min after injection) was linear over the range 30-200 pmol/100 g BW. NT did not alter the disappearance rate of [14C]glucose from plasma during the development of the hyperglycemia. However, the peptide caused a fall in liver glycogen (52 +/- 6.5 to 41 +/- 3.3 mg/g) and a 7-fold increase in the activity of the 5'-AMP independent form of liver glycogen…

Purification and primary structure of alligator neurotensin.

The gastrointestinal neurohormones of reptiles have been poorly characterized structurally. Neurotensin has been purified to apparent homogeneity from an extract of the small intestine of the alligator, Alligator mississipiensis. The primary structure of the peptide (pGlu-Leu-His-Val-Asn-Lys-Ala-Arg-Arg-Pro-Tyr-Ile-Leu) is identical to that of chicken neurotensin. The data provide further evidence for a close phylogenetic relationship between crocodilians and birds.

[Immunoreactive neurotensin in children with gastrointestinal diseases].

Neurotensin is a 13-amino acid hormonal peptide, localized to specific areas in both the brain and gastrointestinal tract of many mammalian species. It has been described as causing a wide variety of central and peripheral actions. At present there is almost no information concerning the possible physiological and pathophysiological significance of this peptide especially in childhood. Therefore we investigated plasma concentrations of neurotensin-like immunoreactivity in 113 healthy and 103 children suffering from various gastrointestinal diseases. The fasting levels (48.7 +/- 17.4 pg/ml) wer…

Neurotensin actions in the retina: mechanisms and variability.

The effects of neurotensin on mudpuppy retinal cells were studied using extracellular and intracellular electrophysiological recording techniques and bath application of the peptide. Ganglion and amacrine cells (but not bipolar or horizontal cells) were reversibly depolarized by low micromolar concentrations of neurotensin. Depolarizations also occurred with neurotensin application during cobalt block of synaptic transmission and were accompanied by decreased input resistances. This suggests neurotensin may act directly on amacrine and ganglion cells as a conventional excitatory transmitter. H…

Neurotensin-induced polydipsia: a structure-activity study.

Neurotensin (NT) and equimolar concentrations of NT fragments were microinjected into the lateral cerebral ventricle of water-deprived rats. NT (10 micrograms) increased water intake 146% in the first 15 min following injection. While NT1-11 and NT10-13 had no effect on water intake, NT8-13 increased water intake 101%. The polydipsia produced by NT8-13 was statistically equivalent to that produced by NT. The results are discussed in relation to a proposed model of the NT receptor.

Neurotensin protects pancreatic beta cells from apoptosis.

The survival of pancreatic beta cells depends on the balance between external cytotoxic and protective molecular systems. The neuropeptide neurotensin (NT) has been shown to regulate certain functions of the endocrine pancreas including insulin and glucagon release. However, the mechanism of action of NT as well as the identification of receptors involved in the pancreatic functions of the peptide remained to be studied. We demonstrate here that NT is an efficient protective agent of pancreatic beta cells against cytotoxic agents. Both beta-TC3 and INS-1E cell lines and the mouse pancreatic is…

A marine analgesic peptide, Contulakin-G, and neurotensin are distinct agonists for neurotensin receptors: uncovering structural determinants of desensitization properties.

Neurotensin receptors have been studied as molecular targets for the treatment of pain, schizophrenia, addiction, or cancer. Neurotensin (NT) and Contulakin-G, a glycopeptide isolated from a predatory cone snail Conus geographus, share a sequence similarity at the C-terminus, which is critical for activation of neurotensin receptors. Both peptides are potent analgesics, although affinity and agonist potency of Contulakin-G toward neurotensin receptors are significantly lower, as compared to those for NT. In this work, we show that the weaker agonist properties of Contulakin-G result in inducin…

Neurotensin and its receptors in the control of glucose homeostasis.

The pharmacological roles of the neuropeptide neurotensin through its three known receptors are various and complex. Neurotensin is involved in several important biological functions including analgesia and hypothermia in the central nervous system and also food intake and glucose homeostasis in the periphery. This review focuses on recent works dealing with molecular mechanisms regulating blood glucose level and insulin secretion upon neurotensin action. Investigations on crucial cellular components involved in the protective effect of the peptide on beta cells are also detailed. The role of …

Neurotensin pulmonary metabolism in normal and asthmatic subjects.

BACKGROUND: The role of pulmonary metabolism of endogenous neurotensin (NT) in asthma is still unclear. Information on this subject in humans is scarce. OBJECTIVES: Evaluation of the pulmonary metabolism of the endogenous NT in asthmatic subjects during symptom-free periods and after a methacoline challenge test and in healthy individuals. METHODS: Ten asthmatic subjects (aged 34 to 70 years), diagnosed with extrinsic (n = 5), atopic (n = 3), and mixed asthma (n = 2), were compared to a group of 10 healthy individuals (aged 45 to 69 years). The asthmatic group of patients was evaluated with …

Effects of neurotensin on the healing of experimental anastomosis of the colon.

The leakage of colonic anastomoses increases perioperative morbidity significantly. The purpose of the study was to investigate the influence of neurotensin, an intestinal trophic peptide, on the healing of colonic anastomosis. Forty-two Wistar-albino rats were separated into three equal groups: Group 1 (control group)--segmental resection of the left colon and end-to-end anastomosis; Group 2 (dexamethasone group)--resection and anastomosis, plus 0.1 mg/kg/day of dexamethasone; Group 3 (neurotensin group)--same surgical procedure plus 300 micrograms/kg/day of neurotensin. Bursting pressure and…

Alteration of neurotensin receptors in MPTP-treated mice.

We examined the sequential changes in neurotensin receptors in the striatum and substantia nigra of mouse brains lesioned with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) by receptor autoradiography, in comparison with the alterations in dopamine uptake sites. The mice received four intraperitoneal injections of MPTP (10 mg/kg) at 1-h intervals and then the brains were analyzed at 6 h and 1, 3, 7, and 21 days after the treatments. [3H]Neurotensin and [3H]mazindol were used to label neurotensin receptors and dopamine uptake sites, respectively. [3H]Neurotensin binding was significantly …

Antidiuretic effects of neurotensin in chloralose anaesthetized dogs.

1. Effects of cerebroventricular and/or intravenous infusions of neurotensin (NT), an endogenous tridecapeptide, on haemodynamics and renal function were investigated in chloralose anaesthetized dogs. 2. Cerebroventricular infusions (i.c.v.) of NT (10-6 mol/L and 10-5 mol/L, 0.1 mL/min) for 30 min did not produce any significant alterations in the measured variables. In the vagotomized dogs, intravenous (i.v.) infusion of NT (10(-5) mol/L) at a rate of 0.1 mL/min for 30 min significantly lowered the arterial blood pressure and glomerular filtration rate; these effects were accompanied by prono…

Microinjection of neurotensin into the CNS induces hyperdipsia in the rat.

Neurotensin (NT) is a neuropeptide and putative neurotransmitter that has been shown to exert a variety of effects on digestive and ingestive processes. In order to address the possibility that NT might play a role in the regulation of water intake as well, the peptide was infused into the lateral cerebral ventricle, amygdala, ventral tegmental area, lateral hypothalamus, and preoptic area of the anterior hypothalamus of rats deprived of water for 16 hours. Neurotensin produced a significant and dose-dependent increase in water intake when injected into the ventricular system but had no effect…

Neurotensin: a central neuromodulator of gastrointestinal motility in the dog.

The effects of intracerebroventricular and intravenous injection of neurotensin on gastrointestinal and colonic motility were examined in fasted and fed conscious, intact, and vagotomized dogs. When administered intracerebroventricularly at 20 ng X kg-1 or higher doses in the fasted state, neurotensin reduced the duration of the periods of gastric motility for 3-4 h. During this time the jejunal migrating motor complex was replaced by isolated phases of regular activity occurring at a rhythm of 2-3/h, while colonic motility was unaffected. These effects were abolished after vagotomy and were n…
