Identification of the first Oomycete annexin as a (1-->3)-beta-D-glucan synthase activator.

(1-->3)-beta-D-Glucans are major components of the cell walls of Oomycetes and as such they play an essential role in the morphogenesis and growth of these microorganisms. Despite the biological importance of (1-->3)-beta-D-glucans, their mechanisms of biosynthesis are poorly understood. Previous studies on (1-->3)-beta-D-glucan synthases from Saprolegnia monoica have shown that three protein bands of an apparent molecular weight of 34, 48 and 50 kDa co-purify with enzyme activity. However, none of the corresponding proteins have been identified. Here we have identified, purified, sequenced an…

Design, Synthesis, and Biological Evaluation of Novel Camphor-Based Hydrazide and Sulfonamide Derivatives as Laccase Inhibitors against Plant Pathogenic Fungi/Oomycetes.

To discover novel natural product-based fungicidal agrochemicals, 41 novel camphanic acid hydrazide and camphor sulfonamide derivatives were designed, synthesized, and tested for their antifungal profile against four plant pathogenic fungi and three oomycetes. As a result, some derivatives presented pronounced inhibitory activities toward Botryosphaeria dothidea, Fusarium graminearum, Phytophthora capsici, and Phytophthora nicotianae. Especially, compound 4b demonstrated the most potent anti-B. dothidea activity (EC50 = 1.28 mg/L), much stronger than positive control chlorthalonil. The in vivo…

CP32, a novel anti-oomycete cyclic peptide, interferes with cell-wall deposition in Phytophthora infestans.

Phytophthora infestans, the causal agent of late blight in potato and tomato, is a historically devastating pathogen that continues to pose a major threat to global food security. Although conventional fungicides remain a cornerstone of crop protection, growing concerns over their environmental impact and long-term sustainability have driven the search for safer and more selective alternatives. In this context, next-generation fungicides with high specificity and minimal ecological footprint are urgently needed. This study introduces CP32, a small cyclic peptide, identified via yeast two-hybri

Detection of genomic DNA of the crayfish plague fungus Aphanomyces astaci (Oomycete) in clinical samples by PCR.

A diagnostic procedure, based on a polymerase chain reaction method (PCR) was developed to detect infection of crayfish with the Oomycete Aphanomyces astaci. A set of oligonucleotide primers was designed to specifically amplify A. astaci DNA in the ITS region surrounding the 5.8S rDNA gene. The PCR amplifies a 115bp amplicon. The specificity of the primers was demonstrated by testing on 27 A. astaci strains and against 20 non-A. astaci Oomycetes and 5 fungal species. Most of the non-A. astaci Oomycete or fungal species included in the study are either known parasites of freshwater crayfish cut…

Reprint of: The diversity of oomycetes on crayfish: Morphological vs. molecular identification of cultures obtained while isolating the crayfish plague pathogen.

Numerous oomycetes colonise the crayfish cuticle, the best known being the crayfish plague pathogen Aphanomyces astaci. Although other oomycetes associated with crayfish complicate the isolation and molecular detection of A. astaci, their diversity is little known. To improve this knowledge, we analysed 95 oomycete isolates obtained during attempts to isolate A. astaci from crayfish presumably infected by this pathogen. We characterized the isolates morphologically and by sequencing of the nuclear internal transcribed spacer (ITS) region. We identified 13 taxa by molecular analysis. Ten of the…

Design, synthesis, anti-oomycete, and anti-fungal activities of novel paeonol derivatives containing carbamate moiety.

Paeonol is the main active ingredient in the Chinese medicinal herb peony root bark. In order to discover new compounds with high efficiency and broad-spectrum pesticide biological activity, twenty novel paeonol carbamate derivatives (1a-c, 2a-c, 3a-c, 4a-c, 5a-c, and 1-5dd') were designed and synthesized by ingeniously introducing carbamate pharmacophores, and their structures were well characterized by 1H NMR, 13C NMR, and HRMS. The stereochemical configurations of two target compounds 2c and 3c were unambiguously subjected to single-crystal X-ray diffraction. Moreover, we evaluated the bioa

Fungal and bacterial-derived CAZymes targeting fungal and oomycete cell walls for biological control of crop diseases.

Cell wall-degrading Carbohydrate-Active enZymes (CAZymes) have substantial potential for biological control of crop diseases by targeting the cell walls of filamentous plant pathogens (fungi and oomycetes) and could substitute for environmentally hazardous pesticides. The cell walls of fungi and oomycetes share common components but also differ in important ways, and there have been considerable recent advances in understanding dynamic structural differences across fungal species. The damage caused to cell walls by CAZymes and the potential of CAZymes in biocontrol are reviewed using studies w…

New Insights into the Anti-Oomycete Mechanisms of a Leaf Extract from Glycyrrhiza glabra Against Pseudoperonospora cubensis.

Cucurbit downy mildew, caused by Pseudoperonospora cubensis, is responsible for high economic losses worldwide in cucumber production. Synthetic pesticides or copper-based products are still important tools to manage the disease. However, the pathogen has developed resistance against common fungicides rather quickly, and there is a need for alternative plant-protecting agents. Glycyrrhiza glabra leaf extract is known for its antifungal activity and was highly effective in former bioassays and semi-commercial trials against downy mildew of cucumber. To elucidate the active ingredients and the m

Essential Oils of Sage, Rosemary, and Bay Laurel Inhibit the Life Stages of Oomycete Pathogens Important in Aquaculture.

Saprolegnia parasitica, the causative agent of saprolegniosis in fish, and Aphanomyces astaci, the causative agent of crayfish plague, are oomycete pathogens that cause economic losses in aquaculture. Since toxic chemicals are currently used to control them, we aimed to investigate their inhibition by essential oils of sage, rosemary, and bay laurel as environmentally acceptable alternatives. Gas Chromatography-Mass Spectrometry (GC-MS) analysis showed that the essential oils tested were rich in bioactive volatiles, mainly monoterpenes. Mycelium and zoospores of A. astaci were more sensitive c

In vitro susceptibility of the oomycete Pythium insidiosum to metallic compounds containing cadmium, lead, copper, manganese or zinc.

Pythium insidiosum is an aquatic oomycete that causes pythiosis, an important and severe disease of difficult treatment that affects humans, domestic and wild animals. This infection is often described in horses in Brazil and humans in Thailand. In clinical practice, we have observed many cases that do not respond to available therapies, indicating the need to explore alternative therapeutic approaches. In this sense, studies using metal compounds in conjunction with available antimicrobial agents have been demonstrated greater antimicrobial activity. Thus, in this research, we tested in vitro

The diversity of the pathogenic Oomycete (Aphanomyces astaci) chitinase genes within the genotypes indicate adaptation to its hosts.

The aim of this work was to evaluate the genetic diversity of the crayfish plague pathogen Aphanomyces astaci (Oomycete) among different isolates and genotypes. Partial chitinase genes were cloned and sequenced from 28 A. astaci isolates including four of the five previously identified RAPD (random amplification of polymorphic DNA)-genotypes. The cloned chitinase sequences (n=176) formed three main groups, CHI1, CHI2 and CHI3, with the CHI2 group then further divided into three subgroups, CHI2A, CHI2B and CHI2C. Some of these chitinases were specific for certain genotypes of A. astaci, as CHI2

Chitin synthases from Saprolegnia are involved in tip growth and represent a potential target for anti-oomycete drugs.

Oomycetes represent some of the most devastating plant and animal pathogens. Typical examples are Phytophthora infestans, which causes potato and tomato late blight, and Saprolegnia parasitica, responsible for fish diseases. Despite the economical and environmental importance of oomycete diseases, their control is difficult, particularly in the aquaculture industry. Carbohydrate synthases are vital for hyphal growth and represent interesting targets for tackling the pathogens. The existence of 2 different chitin synthase genes (SmChs1 and SmChs2) in Saprolegnia monoica was demonstrated using b…

Extraction, characterization and biological activity of a (1,3)(1,6)-β-d-glucan from the pathogenic oomycete Pythium insidiosum.

Pythiosis is a life-threatening infectious disease caused by the pathogenic oomycete Pythium insidiosum. This study is the first to evaluate the P. insidiosum glucan content and its biological activities. The enzymatic quantification of the glucans in P. insidiosum mycelia showed that the β-glucan content was 18.99%±3.59. The cell wall polysaccharide extract consisted of ∼81.7% carbohydrates (exclusively glucose) and ∼18.3% residual amino acids and peptides. The results from monosaccharide composition, methylation and 1D/2D NMR spectroscopy analyses indicated the presence of a highly branched
