Orlistat is as beneficial as metformin in the treatment of polycystic ovarian syndrome.

The objective of this study was to evaluate and compare the effect of treatment with orlistat vs. metformin on the hormonal and biochemical features of patients with polycystic ovarian syndrome (PCOS). Twenty-one Caucasian women with PCOS [mean (+/-SEM) age 27 +/- 0.9 yr and body mass index 36.7 +/- 3.3 kg/m(2)] participated in this prospective, randomized, open-labeled study. All subjects had an 8-wk run-in period of dietary modification and then randomized to receive either metformin (500 mg three times daily) or orlistat (120 mg three times daily) for 3 months. Weight, blood pressure, and f

Orlistat impairs Chlamydia trachomatis development by disrupting its developmental cycle.

Chlamydia trachomatis is a common sexually transmitted bacterial pathogen that often causes urogenital infection and may progress to chronic inflammation and fibrosis due to its clinically silent nature. Although azithromycin and doxycycline are widely used for treatment, therapeutic failure has been documented in clinical cases. In this study, we demonstrate that orlistat disrupts the chlamydial developmental cycle both in vitro and in vivo. Orlistat inhibits early stages of chlamydial infection, reticulate body replication, and reticulate body-to-elementary body differentiation. Transcriptom

Orlistat abuse in a case of bulimia nervosa: the changing Indian society.

Eating disorders like bulimia nervosa, generally considered to be rare in Asian countries, are currently on the rise among the youth, probably secondary to rapid westernization and globalization of the orient. Clinical manifestations of these disorders, which were previously thought to be different in the oriental countries, are now also often seen to parallel their western counterparts. However, detailed clinical descriptions of such cases from Asian countries, documenting the societal change, are missing. In addition, the possibility of abuse of various anti-obesity drugs as a part of bulimi

Safety profile of orlistat: results of a prescription-event monitoring study.

INTRODUCTION: Orlistat, the first of a new class of drugs for the treatment of obesity, was launched in the UK in December 1998. The prescribing information recommends that treatment with orlistat should be discontinued after 12 weeks if the patient has not achieved a specified loss of weight. OBJECTIVE: To monitor the safety of orlistat prescribed in the primary care setting in England using prescription-event monitoring (PEM). METHODS: A postmarketing surveillance study using the observational cohort technique of PEM. Patients were identified from dispensed prescriptions issued by primary

Review article: malnutrition and maltreatment--a comment on orlistat for the treatment of obesity.

The prevalence of obesity has doubled in the last 10 years and is now reaching epidemic proportions. There is a significant comorbidity and financial cost associated with this disorder. Orlistat is an intestinal lipase inhibitor that is approved for the treatment of obesity. Recent randomized, double-blind, placebo-controlled trials have demonstrated the benefit of orlistat used in conjunction with a hypocaloric (low-fat) diet in facilitating weight reduction and the long-term maintenance of this weight loss. Patients treated with orlistat lost a greater amount of initial body weight compared

Pattern of orlistat exposures in children aged 5 years or less.

On February 7, 2007, orlistat became the first weight-loss drug approved by the United States Food and Drug Administration for over-the-counter sales. However, information on exposures among young children is limited. The objective of this study was to describe the pattern of orlistat exposures among young children reported to poison control centers. The pattern of all exposures to orlistat alone among patients < or = 5 years old reported to six poison control centers during 1999-2005 was identified with respect to various factors. There were 107 cases. The average age was 21.4 months. There w

Long-term persistence with orlistat and sibutramine in a population-based cohort.

OBJECTIVE: Orlistat and sibutramine are widely prescribed antiobesity agents that are approved for 2 years of continuous use. Previous 1-4-year randomized, placebo-controlled trials of these drugs have reported average weight losses of <5 kg, significant adverse effects and attrition rates of up to 60%. The objective of this study was to determine the long-term persistence with orlistat and sibutramine therapy outside a clinical trial setting. DESIGN, SETTING AND PATIENTS: Population-based administrative data from British Columbia, Canada, were used to create an inception cohort of orlistat a

Comparison of efficacy of sibutramine or orlistat versus their combination in obese women.

OBJECTIVE: Sibutramine and orlistat are currently used for weight loss. We aimed to investigate the effect of orlistat and sibutramine combination therapy in treatment of obese women. SUBJECTS AND DESIGN: Study population consisted of 89 obese women who had a body mass index > or = 30 kg/m2, were normotensive, and had normal glucose tolerance. All patients were placed on a diet which contained fat approximately 30% of total calorie intake and the diet was designed to cause an energy deficit of approximately 2.51-3.56 megajoule/day. At the first month of diet (baseline), all patients were rand

Orlistat: a second look. At best, a minor adjunct to dietary measures.

(1) Treatments for obesity are disappointing, and none has yet shown an effect on morbidity or mortality. Non drug treatments have not been assessed adequately. Long-term maintenance of weight loss requires long-term patient management. (2) Orlistat, a gastrointestinal lipase inhibitor, is licensed in Europe for the treatment of obesity, in combination with a low-calorie diet. (3) The risk-benefit ratio of orlistat could not be estimated from the initial assessment file in 1999. There were fears over a possible increase in the risk of breast cancer. (4) Few new efficacy data have been obtained

An integrated analysis of liver safety data from orlistat clinical trials.

OBJECTIVE: Orlistat is an oral gastrointestinal lipase inhibitor and is indicated for treatment of obesity in combination with a hypocaloric diet. Post-marketing reports of adverse reactions revealed hints for possible drug-induced liver injury which has prompted changes to the product information. Orlistat's development program, involving over 30,000 patients, did not indicate a hepatic safety issue. METHODS: We analyzed liver function test data from randomized clinical trials of orlistat, using i) meta-analysis of published study safety data, ii) time-to-event analysis for individual patien

Severe hepatotoxicity associated with orlistat: clinical presentation, risk factors and outcomes.

BACKGROUND AND AIMS: Orlistat, a widely prescribed anti-obesity agent, has been associated with rare but severe hepatotoxicity. The risk profile remains poorly characterised in vulnerable populations, particularly elderly individuals and those with concurrent alcohol use. This study presents a fatal case of OILI and systematically reviews published cases to delineate the clinical spectrum of the injury and highlight features that may be relevant to vulnerable populations. METHODS: We report a fatal case of OILI in a 66-year-old man with chronic alcohol use. A systematic literature review was

Orlistat Resensitizes Sorafenib-Resistance in Hepatocellular Carcinoma Cells through Modulating Metabolism.

Sorafenib is one of the options for advanced hepatocellular carcinoma treatment and has been shown to extend median overall survival. However, sorafenib resistance often develops a few months after treatment. Hence, developing various strategies to overcome sorafenib resistance and understand the possible mechanisms is urgently needed. We first established sorafenib-resistant hepatocellular carcinoma (HCC) cells. Then, we found that sorafenib-resistant Huh7 cells (Huh7/SR) exhibit higher glucose uptakes and express elevated fatty acid synthesis and glucose metabolism-related proteins than thei

Safety assessment of FDA-approved (orlistat and lorcaserin) anti-obesity medications.

INTRODUCTION: Options for treating obesity remain limited despite it being a chronic, recurrent and morbid condition. New drugs that are proposed for its treatment encounter strong reluctance by regulatory agencies and many doctors. AREAS COVERED: This review will focus on the safety of an older drug, orlistat (the only one still approved in the European Union) and a newer recently FDA-approved one, lorcaserin. Both are approved as long-term monotherapy for obesity in the United States of America and they have demonstrated median weight loss of nearly 3% over placebo. EXPERT OPINION: Researc

Orlistat Therapy for Children With Type 1 Hyperlipoproteinemia: A Randomized Clinical Trial.

CONTEXT: Patients with type 1 hyperlipoproteinemia (T1HLP), a rare genetic disorder, have extreme chylomicronemia and recurrent episodes of acute pancreatitis. Currently, the only therapeutic option is to consume an extremely low-fat diet because the triglyceride-lowering medications are not efficacious. OBJECTIVE: To determine the efficacy of orlistat, a gastric and pancreatic lipase inhibitor, in reducing serum triglyceride levels in patients with T1HLP. DESIGN AND SETTING: We conducted a randomized, open-label, clinical trial with a four-period, two-sequence ("orlistat" and "off orlistat"

Orlistat Confers Neuroprotection in Traumatic Brain Injury by Modulating Microglial Lipid Metabolism.

Traumatic brain injury (TBI) represents a major cause of mortality and disability worldwide, particularly affecting young adults and elderly populations. This study investigates the neuroprotective potential of orlistat (ORL), a gastrointestinal lipase inhibitor, in a murine TBI model. Behavioral, histological, and molecular analyses demonstrated that ORL significantly attenuated TBI-induced neurological damage. Microglial depletion experiments revealed that ORL's neuroprotective effects were largely mediated through microglial modulation. In vitro and in vivo studies showed that ORL suppresse

Orlistat reverses intratesticular lactate transport decline and infertility in male obese rats.

Obesity and its accompanying complications predispose to abnormal testicular glucose metabolism, penile erectile dysfunction and subfertility. This study examined the potentials of orlistat in attenuating erectile dysfunction and fertility decline in high-fat diet (HFD)-induced obesity in male rats. Eighteen adult male Sprague-Dawley rats whose weights were between 250 and 300 g were divided into three groups (n = 6/group) namely: normal control (NC), HFD and HFD + orlistat (10 mg/kg body weight/day co-administered for 12 weeks) (HFD+O). During the 11th and 12th week, mating behaviour and fert
