Experiments were performed to improve the quantitative analysis of passive cutaneous anaphylaxis (PCA) by labelling the antigen with radioactive iodine. The ability of injected rat IgE myeloma protein IR162 to inhibit passive and active cutaneous anaphylaxis in Lewis rats was investigated. IgE injected i.p. 24 hr before the sensitization with IgE anti-ovalbumin (OVA) completely inhibited both IgE- and IgG2a-induced passive cutaneous anaphylactic (PCA) reactions at a dose (2.5 mg/100 g body weight) that resulted in peak serum concentrations of 150 micrograms IgE IR162/ml. The studies reported here demonstrated that adult chickens, when immunized with bovine serum albumin, respond with the production of serum antibodies which will mediate passive cutaneous anaphylactic (PCA) reactions in 10-day-old chicks but not in adults. This difference is at least in part due to differences in histamine sensitivity between chicks and adults. The molecule responsible for this PCA activity was indistinguishable from the predominant 7S chicken immunoglobulin antigenically and by a combination of gel filtration on Sephadex G-200 and ion exchange chromatography on DEAE-cellulose.

Passive cutaneous anaphylaxis was produced in chickens with bovine serum albumin (BSA) and anti-BSA chicken serum. Colloidal carbon was then given intravenously to identify the leaky vessels. Microscopic examination of cleared unstained skin revealed the topography of labelled vessels. The carbon labelling was confined to venules and small veins only. Examination of the time-course of permeability response revealed a biphasic pattern.
