Double prenylation of SNARE protein Ykt6 is required for lysosomal hydrolase trafficking.

Ykt6 is an evolutionarily conserved SNARE protein regulating Golgi membrane fusion and other diverse membrane trafficking pathways. Unlike most SNARE proteins, Ykt6 lacks a transmembrane domain but instead has a tandem cysteine motif at the C-terminus. Recently, we have demonstrated that Ykt6 undergoes double prenylation at the C-terminal two cysteines first by farnesyltransferase and then by a newly identified protein prenyltransferase named geranylgeranyltransferase type-III (GGTase-III). GGTase-III consists of a novel α subunit prenyltransferase alpha subunit repeat containing 1 (PTAR1) and…

Defective Protein Prenylation in a Spectrum of Patients With Mevalonate Kinase Deficiency.

The rare autoinflammatory disease mevalonate kinase deficiency (MKD, which includes HIDS and mevalonic aciduria) is caused by recessive, pathogenic variants in the MVK gene encoding mevalonate kinase. Deficiency of this enzyme decreases the synthesis of isoprenoid lipids and thus prevents the normal post-translational prenylation of small GTPase proteins, which then accumulate in their unprenylated form. We recently optimized a sensitive assay capable of detecting unprenylated Rab GTPase proteins in peripheral blood mononuclear cells (PBMCs) and showed that this assay distinguished MKD from ot…

Protein prenylation and human diseases: a balance of protein farnesylation and geranylgeranylation.

The protein prenylation is one of the essential post-translational protein modifications, which extensively exists in the eukaryocyte. It includes protein farnesylation and geranylgeranylation, using farnesyl pyrophosphate (FPP) or geranylgeranyl pyrophosphate (GGPP) as the substrate, respectively. The prenylation occurs by covalent addition of these two types of isoprenoids to cysteine residues at or near the carboxyl terminus of the proteins that possess CaaX motif, such as Ras small GTPase family. The attachment of hydrophobic prenyl groups can anchor the proteins to intracellular membranes…

Prenylated phenylpropanoids from enzyme catalysis and their antibacterial activities.

Phenylpropanoids are a large class of natural products with C6-C3 as the backbone. Research has shown that prenyl units can increase the lipophilicity of compounds, enhance their affinity with biofilms, and improve their bioavailability. In this work, the prenylations of phenylpropanoids with three prenyl donors including dimethylallyl diphosphate (DMAPP), geranyl diphosphate (GPP), and farnesyl diphosphate (FPP) by two fungal prenyltransferases were investigated. A total of 11 products (1D1a, 1D1 - 1D3, 1G, 2D1 - 2D4, 2G, and 3G) were obtained, two of which are dearomative prenylated products…

Biosynthetic allene and alkyne formation by enzymatic prenyl demethylation.

Allenes and alkynes are versatile functional groups in chemical biology and drug discovery. However, how nature biosynthetically installs them in natural products, especially allenes, remains poorly understood. Here we uncovered that allenes and alkynes can be formed enzymatically through oxidative C(sp2)-demethylation of the common five-carbon prenyl group. Two fungal cytochrome P450 monooxygenases, PpnB and NseB, from the penipratynolene and sinuxylamide biosynthetic pathways, respectively, were shown to catalyze oxidative removal of a C(sp2)-methyl group in O-prenyl L-tyrosine to afford O-h

Mevalonate Metabolism Orchestrates Platelet Hyperactivation via Protein Prenylation to Aggravate Septic Thrombosis.

Sepsis is a life-threatening systemic inflammatory disorder that triggers disseminated intravascular coagulation and microthrombosis, in which exaggerated platelet activation exacerbates disease progression. The mevalonate pathway is responsible for the biosynthesis of vital isoprenoids and sterols, also playing an important role in the prenylation modification of proteins. We found the mevalonate metabolic pathway is highly activated in platelets during infection. The interruption of this pathway, using platelet specific mevalonate kinase deficiency mice model, could significantly suppress pl…

Host prenylation promotes membrane association of the Salmonella effector GtgE during infection.

The Rab32 antimicrobial pathway restricts Salmonella Typhi replication in murine macrophages. The broad-host pathogen Salmonella Typhimurium … acuole, whereas the C225S variant showed reduced membrane association. These findings identify host prenylation as a mechanism that may direct GtgE to intracellular membranes where Rab32 is accessible for cleavage.

Alteration of RhoA Prenylation Ameliorates Cardiac and Vascular Remodeling in Spontaneously Hypertensive Rats.

BACKGROUND: In our previous study, farnesyl pyrophosphate synthase (FPPS) was shown to be increased in spontaneously hypertensive rats (SHR) and in mice with angiotensin-II induced cardiac hypertrophy. Overexpression of FPPS induced cardiac hypertrophy and fibrosis in mice, accompanied by an increase in the synthesis of farnesyl pyrophosphate (FPP) and geranylgeranyl pyrophosphate (GGPP). In the present study, we investigated the mechanisms of reversing cardiovascular remodeling in SHR by inhibiting FPPS. METHODS AND RESULTS: Six-week-old rats were given vehicle or an FPPS inhibitor (alendron

Catalytic prenyl conjugate additions for synthesis of enantiomerically enriched PPAPs.

Polycyclic polyprenylated acylphloroglucinols (PPAPs) are a class of >400 natural products with a broad spectrum of bioactivity, ranging from antidepressant and antimicrobial to anti-obesity and anticancer activity. Here, we present a scalable, regio-, site-, and enantioselective catalytic method for synthesis of cyclic β-prenyl ketones, compounds that can be used for efficient syntheses of many PPAPs in high enantiomeric purity. The transformation is prenyl conjugate addition to cyclic β-ketoesters promoted by a readily accessible chiral copper catalyst and involving an easy-to-prepare and is

Targeting a uniquely nonspecific prenyl synthase with bisphosphonates to combat cryptosporidiosis.

Cryptosporidiosis is a neglected disease without a wholly effective drug. We present a study demonstrating nitrogen-containing bisphosphonates (N-BPs) to be capable of inhibiting Cryptosporidium parvum at low micromolar concentrations in infected MDCK cells. Predictably, the mechanism of action is based on inhibition of biosynthesis of isoprenoids but the target enzyme is unexpectedly a distinctive C. parvum enzyme dubbed nonspecific polyprenyl pyrophosphate synthase (CpNPPPS). This enzyme produces various isoprenoid products larger than FPP and is inhibited by N-BPs at subnanomolar concentrat

Prenylation: A key regulatory point in tumor metabolism and a potential therapeutic target.

Prenylation is an important post-translational lipid modification that enhances protein hydrophobicity and regulates subcellular localization. It plays a critical role in multiple signaling pathways and metabolic processes and is closely associated with the development and progression of various diseases, particularly in tumor initiation, progression, and metastasis. However, its systemic regulatory roles and therapeutic vulnerabilities in cancer remain incompletely understood. In this review, we summarize the molecular mechanisms of protein prenylation and its biological functions, with a par…

The role of protein prenylation in kidney diseases: Molecular basis and therapeutic implications.

Kidney diseases may lead to a life-threatening condition, with their global prevalence increasing day by day. The progressive nature of kidney diseases is driven by a complex interplay of cellular and molecular events, among which protein prenylation plays a pivotal role. Protein prenylation, a lipid-mediated post-translational modification, is essential for the functional regulation of small guanosine triphosphatase binding proteins, including Ras, Rac, Rab, and Rho, which govern cell signaling, proliferation, and membrane dynamics. Aberrant prenylation disrupts these signaling cascades, cont…

Diprenylated cyclodipeptide production by changing the prenylation sequence of the nature's synthetic machinery.

Ascomycetous fungi are often found in agricultural products and foods as contaminants. They produce hazardous mycotoxins for human and animals. On the other hand, the fungal metabolites including mycotoxins are important drug candidates and the enzymes involved in the biosynthesis of these compounds are valuable biocatalysts for production of designed compounds. One of the enzyme groups are members of the dimethylallyl tryptophan synthase superfamily, which mainly catalyze prenylations of tryptophan and tryptophan-containing cyclodipeptides (CDPs). Decoration of CDPs in the biosynthesis of mul…
