Prevotella denticola promotes caries by inducing oral microbial dysbiosis.

BACKGROUND: While the role of oral microbial dysbiosis in cariogenicity is widely appreciated, it remains unclear whether some resident microorganisms are active contributors in this process. Recently, increasing evidence showed that Prevotella spp. are associated with caries whereas the identified roles at species level remain undefined. This study aimed to analyze the effects of Prevotella denticola on microbial ecosystems and pathogenic mechanisms associated with caries. METHODS: Here, we applied metagenomics to supragingival microbiota (dental plaque) samples from 62 adults to identify dis…

Prevotella mikamonis sp. nov., isolated from equine clinical specimens.

In this study, five strains of anaerobic, Gram-negative, rod-shaped bacteria were characterized phenotypically, biochemically and genotypica … n 73.1% and 28.6%, respectively, compared with the type strains of closely related species, such as Prevotella phocaeensis and Prevotella merdae. Based on these phenotypic, phylogenetic and genomic data, a novel species of the genus Prevotella, Prevotella mikamonis sp. nov., is proposed, with GTC17253T (CCUG77931T) as the type strain.

Current research progress on Prevotella intermedia and associated diseases.

Prevotella intermedia is a Gram-negative anaerobic bacterium that is a common pathogen of periodontitis. Recent studies have revealed that P. intermedia is closely associated with a variety of diseases involving multiple systems. Under the action of its virulence factors such as cysteine protease and adhesins, P. intermedia has the ability to bind and invade various host cells including gingival fibroblasts. It can also copolymerize a variety of pathogenic bacteria, leading to interference with the host's immune inflammatory response and causing various diseases. In this article, we review the…

Refractory pneumonia caused by Prevotella heparinolytica: a case report.

BACKGROUND: Prevotella heparinolytica is a Gram-negative bacterium that is commonly found in the oral, intestinal, and urinary tracts. It has been extensively studied in lower respiratory tract infections in horses, which has heparinolytic activity and can secrete heparinase and further induces virulence factors in cells and causes disease. However, no such cases have been reported in humans. CASE PRESENTATION: A 58-year-old male patient from China presented to the respiratory clinic in Suzhou with a productive cough producing white sputum for 20 days and fever for 3 days. Prior to this visit…

Bacteremia due to Prevotella oris of probable hepatic origin.

Prevotella species are Gram-negative anaerobic bacilli mainly involved in oral cavity infections. We report a rare case of bacteremia due to P. oris of probable hepatic origin. A 70-year-old man with a history of diabetes mellitus was admitted for general malaise, fever and dyspnea. The patient was admitted to the ICU due to septic shock. Two sets of blood cultures yielded a pure culture of an anaerobic microorganism identified as P. oris by MALDI-TOF MS. The same microorganism was isolated from a hepatic abscess drainage. Only resistance to penicillin was documented. Treatment with piperacill…

Prevotella intermedia Synergistically Exacerbates Pneumonia Induced by Oral Streptococci.

BACKGROUND: The precise mechanisms of respiratory infection caused by oral anaerobic bacteria remain elusive. Unexpectedly, bacterial microbiota analysis with 16S rRNA revealed "hidden" mixed infections of anaerobic bacteria and commensal oral Streptococcus species in patients with community-acquired pneumonia. The study aimed to elucidate the mechanisms by which Prevotella intermedia exacerbates oral streptococcal pneumonia. METHODS: Oral streptococci were oropharyngeally administered with the culture supernatants of P intermedia (PiSup) in mice to assess survival, microbial burden, inflamma…

The immune response to Prevotella bacteria in chronic inflammatory disease.

The microbiota plays a central role in human health and disease by shaping immune development, immune responses and metabolism, and by protecting from invading pathogens. Technical advances that allow comprehensive characterization of microbial communities by genetic sequencing have sparked the hunt for disease-modulating bacteria. Emerging studies in humans have linked the increased abundance of Prevotella species at mucosal sites to localized and systemic disease, including periodontitis, bacterial vaginosis, rheumatoid arthritis, metabolic disorders and low-grade systemic inflammation. Intr…

Isolation, identification, and quantification of Prevotella intermedia and Prevotella nigrescens in subgingival plaque samples of periodontally healthy and chronic periodontitis patients through chromogenic in situ hybridization.

BACKGROUND: The genus Prevotella comprising over 50 identified species is a normal commensal of the oral cavity. The commonly studied species are Prevotella intermedia (PI) and Prevotella nigrescens (PN). The molecular technique, used for isolating and identifying these species, requires specialized equipment, making them not feasible in a standard laboratory. Chromogenic in situ hybridization (CISH) can be performed in routine laboratory and is economical and less time-consuming. Hence, the aim of this study was to isolate, identify, and quantify PI and PN from subgingival plaque samples of c…

Prevotella copri impairs bone mass via osteoclast activation in mice.

Prevotella copri, a predominant commensal bacterium in the human gut, exhibits positive or negative correlations with multiple metabolic disorders through its abundance dynamics. Although its association with rheumatoid arthritis pathogenesis is established, the effect of P. copri on bone metabolism remains elusive. This study demonstrates that P. copri exposure induces systemic bone metabolic imbalance in both normal physiological murine models and in vitro-cultured bone marrow-derived adherent cells (BMDCs), which is characterized by exacerbated osteoclastogenesis. Notably, succinate acts as…
