Prions and transfusion medicine.

BACKGROUND AND OBJECTIVES: There is growing concern at national and international levels that blood supplies might be contaminated with Creutzfeldt-Jakob disease (CJD) agents (prions). MATERIALS AND METHODS: This review on CJD and blood focuses on experimental and epidemiological data, available tests, factors contributed by donors and recipients, and production processes. All of this has been recently detailed in excellent reports and opinions which are summarised here. RESULTS: Solid evidence from experimentally infected animals, and fragmentary evidence from naturally infected humans, ind…

[Prions and their biology].

Prion diseases or transmissible spongiform encephalopathies are fatal neurodegenerative disorders featured by an aberrant metabolism of a cellular membrane glycoprotein, the prion protein (PrP-C). PrP-C is being related to Cu (II) homeostasis and postulated as candidate for cell signaling and cell adhesion functions. Under pathological conditions PrP-C converts into a conformational isomer (PrP*, PrP-res, PrP-Sc, PrP-Creutzfeldt-Jakob disease, etc.). In addition to divergent biochemical characteristics, the latter form displays the property of recognizing the normal protein and transforming it

[Prions and infantile pathology].

The authors briefly review the problem of prions pathology (with particular emphasis on children). The outstanding interest of the subject justifies this review.

Prions and prion proteins.

Neurodegenerative diseases of animals and humans including scrapie, bovine spongiform encephalopathy, and Creutzfeldt-Jakob disease are caused by unusual infectious pathogens called prions. There is no evidence for a nucleic acid in the prion, but diverse experimental results indicate that a host-derived protein called PrPSc is a component of the infectious particle. Experiments with scrapie-infected cultured cells show that PrPSc is derived from a normal cellular protein called PrPC through an unknown posttranslational process. We have analyzed the amino acid sequence and posttranslational mo…

Inactivation Methods for Prions.

Incidences of iatrogenic Creutzfeldt-Jakob disease (iCJD) are caused by transplantation of prion-contaminated hormones, cornea and dura mater as well as contact with prion- contaminated medical devices, such as stereotactic electrodes, used in neurosurgery. Because prions are highly resistant and difficult to inactivate, prion contamination is a severe risk when medical instruments are reused after surgical procedures involving suspicious and confirmed cases of patients with prion diseases. Therefore, when high-risk procedures such as cerebral surgery, craniotomy surgery, orthopaedic spinal su…

Here we review potential interactions between prions and the proteasome outlining how the UPS may be implicated in prion-mediated neurodegeneration.

Prions, BSE and food.

Biochemical and biophysical properties of prions including possible inactivation methods are reviewed. Possible molecular markers of transmissible spongiform encephalopathy (TSE) and mechanisms behind infectivity and correlation with clinical symptoms are discussed. The risk of Bovine Spongiform Encephalopathy (BSE) for humans i.e. variant Creutzfeldt-Jakob Disease (cCJD) is addressed in detail. The consequences of the emergence of the new cCJD and the lack of information on the infectivity of cCJD at the clinical stage of the disease in relation to the need to reconsider the biological concep…

Prions: a mystery unravelled?

Prions result in fatal degeneration of the central nervous system (CNS) in the form of diseases known as transmissible spongiform encephalopathies (TSEs). The discovery in 1996 of a new variant of Creutzfeldt-Jakob disease (a human TSE) and experimental confirmation that it is caused by the prion strain responsible for bovine spongiform encephalopathy (BSE) has greatly spurred research in this field. The mechanism underlying prion propagation is now reasonably clear. Prions multiply, in fact, by stimulating their hosts to produce proteins that are initially normal, but acquire an abnormal, pri…

Prions and blood products.

The transmission of Creutzfeldt-Jakob disease (CJD) by human pituitary-derived growth hormone has led to concerns that blood products might also provide a route for the iatrogenic transmission of CJD. A number of actions have been implemented by regulatory authorities to address such concerns, and numerous studies have been undertaken to determine whether or not there is a risk of CJD being transmitted in this manner. To date, no excess risk has been identified, leading to a growing consensus that there is little or no risk of long established forms of CJD being transmitted to recipients of bl

[Prions: definition and diseases].

In this article we review the concept and terminology of prions, their replication and some current hypothesis on the nature of these infectious agents causing neurodegenerative diseases. This revision also summarizes the etiopathogenic, epidemiological, clinical and neuropathological features of the prion diseases or human transmissible spongiform encephalopathies, and some methods for their early diagnosis. Finally, we discuss the possible link between the bovine spongiform encephalopathy and the new cases of Creutzfeldt-Jakob disease identified in the United Kingdom.

Prions and prion diseases.

Currently known transmissive spongiform encephalopathies in humans and animals are presented. Caused by prions, they are known as "prion disease". The results of extensive investigations of prions are described, as well as the theories about their nature. Relating to the occurrence of the epizootic of bovine spongiform encephalopathy in England, its causes and consequences as well as the research on possible relationship between this encephalopathy and the Creutzfeldt-Jakob's disease are discussed.

[Encephalopathy caused by prions].

The concept of prion encephalopathy has emerged from such previous notions as slow virus infections, spongiform encephalopathies or transmissible dementias. The term prion (Prusiner, 1982) is now used in preference to unconventional agents. Proteins and genes of prions have recently been identified by molecular biology. Exactly how prion proteins are amplified in cells is still unknown. It has been demonstrated that amyloid deposits in scrapie-infected brain, Creutzfeldt-Jakob, Gerstmann-Straüssler and Kuru diseases are composed of prion proteins. Prion encephalopathies are good models to stud…

The Structure of PrPSc Prions.

PrPSc (scrapie isoform of the prion protein) prions are the infectious agent behind diseases such as Creutzfeldt-Jakob disease in humans, bovine spongiform encephalopathy in cattle, chronic wasting disease in cervids (deer, elk, moose, and reindeer), as well as goat and sheep scrapie. PrPSc is an alternatively folded variant of the cellular prion protein, PrPC, which is a regular, GPI-anchored protein that is present on the cell surface of neurons and other cell types. While the structure of PrPC is well studied, the structure of PrPSc resisted high-resolution determination due to its general …

Prions, proteinase K and infectivity.

OBJECTIVES: Systemic lupus erythematosus (SLE) is a chronic autoimmune disease characterised by a complex aetiology that encompasses genetic, environmental, and immunological factors. A comprehensive understanding of SLE aetiology is essential for the development of effective therapeutic strategies and the enhancement of patient outcomes. This study aims to provide a thorough bibliometric analysis of research concerning the aetiology of SLE conducted over the past decade. METHODS: A bibliometric analysis was conducted based on articles published between January 2014 and December 2023, retriev
