Urinary excretion of propylene glycol butyl ether (PGBE) and propylene glycol methyl ether (PGME) in healthy human participants and workers.

Exposure to organic solvents can affect the central nervous system. Propylene glycol ethers such as propylene glycol methyl ether (PGME) and propylene glycol butyl ether (PGBE) are organic solvents widely used in commercial products including cleaning products. PGME has an established Swiss occupational exposure limit and a biomonitoring limit value while PGBE does not, which is probably due to the absence of toxicokinetic data. Our aim was to (1) Characterize PGBE elimination toxicokinetics in healthy human participants (N = 11) exposed under controlled conditions; (2) Provide urinary elimina…

Modification of Polylactide with Triblock and Diblock Copolymers of Ethylene Glycol and Propylene Glycol.

Polylactide (PLA) was melt blended with block copolymers of ethylene glycol and propylene glycol: a triblock copolymer (PPG-b-PEG-b-PPG) with a molar mass of 2700 g/mol and 40 wt% PEG content, and a diblock copolymer (PPG-b-PEG) with a molar mass of 4000 g/mol and 50 wt% PEG content. The structure as well as the thermal and mechanical properties of both amorphous and crystallized blends were investigated. Due to the copolymers' chemical composition and the resulting phase structure, the 10 wt% amorphous blends with PPG-b-PEG-b-PPG and PPG-b-PEG, with Tg values of 38 °C and 46 °C, respectively,…

Propylene glycol toxicity from compulsive corn starch ingestion.

INTRODUCTION: Propylene glycol (PG) is usually considered safe, however, toxicity can develop with high doses or when used for prolonged periods of time. PG can be found in some medications as well as some food products. We report a case of likely PG toxicity that occurred after compulsive daily ingestion of large amounts of corn starch. CASE REPORT: Our patient initially presented to an outside hospital (OSH) via ambulance for altered mental status. Her mental status improved after her blood sugar of 25 was corrected. On admission to OSH Emergency Department her initial vital signs included …

Propylene glycol enhances anti-inflammatory effects of phenylbutazone.

1. The interference of propylene glycol with anti-inflammatory effects of phenylbutazone was investigated. 2. Inhibitory effect of phenylbutazone on both carrageenin-induced edema and the cotton pellet granuloma was increased when propylene glycol was used as solvent. 3. Propylene glycol given alone inhibited carrageenin-induced edema and pleurisy, as well as granulomatous tissue formation. 4. Some pharmacokinetic parameters of phenylbutazone were also changed by propylene glycol administered simultaneously. 5. These results suggest that propylene glycol probably increases the anti-inflammator…

Histological aspects of skin reactions to propylene glycol.

In view of the increased use of dermatological preparations containing a high concentration of propylene glycol (PG) for use under occlusive dressings, patch tests were performed with pure PG in a group of 98 out-patients at a skin clinic. No indication of a possible allergic contact dermatitis to PG could be found in the history of any of these patients. In 11 of them, the patch test reactions after a 48-h application were positive. In eight cases the reaction was scored as one plus, in two cases as two plus, and in the remaining case a three plus reaction was observed. In view of the negativ…

Propylene Glycol in Contact Dermatitis: A Systematic Review.

Propylene glycol (PG), an emollient and emulsifier found in cosmetics, medications, and food, has been granted the dubious honor of being named the American Contact Dermatitis Society's Allergen of the Year for 2018. Contact, systemic, and irritant cutaneous reactions have been documented for PG, which has become an increasingly common ingredient. Propylene glycol is as contentious as it is ubiquitous because it acts as both a weak sensitizer and an irritant, confounding the results of positive patch tests. This review serves to delve into what we know about PG from previous reports and studie…

Propylene glycol toxicity complicating use of barbiturate coma.

INTRODUCTION: Barbiturate coma is a necessary medical therapy in certain instances, such as the treatment of refractory status epilepticus or severe intracranial hypertension, but its use is often wrought with serious complications. Potential complications include hemodynamic instability, respiratory depression, and immunosuppression with frequent nosocomial infections. High doses of barbiturates may also lead to the accumulation of propylene glycol, the vehicle used in the intravenous formulations of both pentobarbital and phenobarbital, thereby yielding another less-recognized complication o…

Skin-sensitizing and irritant properties of propylene glycol.

In the several publications reviewed in this article, propylene glycol (PG; 1,2-propylene glycol) is described as a very weak contact sensitizer, if at all. However, particular exposures to PG-containing products might be associated with an elevated risk of sensitization. To identify such exposures, we analysed patch test data of 45 138 patients who have been tested with 20% PG in water between 1992 and 2002. Out of these, 1044 patients (2.3%) tested positively, 1083 showed a doubtful, follicular or erythematous reaction (2.4%) and 271 explicit irritant reactions (0.6%). This profile of patch …

The ototoxic potential of propylene glycol in guinea pigs.

Propylene glycol, previously indicated to be ototoxic and to produce "deafness," was studied in the middle ear space of guinea pigs. Ninety percent propylene glycol produced conductive middle ear problems, but there was no loss of hair cells above that found in normal untreated animals. Ten percent propylene glycol produced no negative effects in the middle ear or elsewhere. Topical ear drops containing high concentrations of propylene glycol are contraindicated in cases with perforation of the tympanic membrane.

A review of propylene glycol dinitrate toxicology and epidemiology.

Propylene glycol dinitrate (PGDN) is a rapidly metabolized, nitrated ester explosive propellant with acute cardiovascular effects at lower levels of exposure and methemoglobinemia and vascular collapse at higher ones. Exposure can be by dermal or inhalation routes. Toxicology has played an important role in setting workplace permissible exposure limits, which have been limited by vascular headaches and subtle, transient decrements in central nervous system performance. Less well characterized is the etiology of excess, long-term cardiac morbidity in PGDN-exposed workers. Human central nervous …

Patch Testing to Propylene Glycol: The Mayo Clinic Experience.

BACKGROUND: Propylene glycol (PG) is a solvent, vehicle, and humectant being used increasingly in a wide array of personal care products, cosmetics, and topical medicaments. Propylene glycol is a recognized source of both allergic and irritant contact dermatitis. OBJECTIVE: The aim of the study was to report incidence of positive patch tests to PG at Mayo Clinic. METHODS: We retrospectively reviewed records of all patients patch tested to PG from January 1997 to December 2016. RESULTS: A total of 11,738 patients underwent patch testing to 5%, 10%, or 20% PG. Of these, 100 (0.85%) tested pos…

Preclinical safety evaluation of inhaled cyclosporine in propylene glycol.

Cyclosporine inhalation solution has the potential to improve outcomes following lung transplantation by delivering high concentrations of an immunosuppressant directly to the allograft while minimizing systemic drug exposure and associated toxicity. The objective of these studies was to evaluate the potential toxicity of aerosolized cyclosporine formulated in propylene glycol when given by inhalation route to rats and dogs for 28 days. Sprague-Dawley rats received total inhaled doses of 0 (air), 0 (vehicle, propylene glycol), 7.4, 24.3, and 53.9 mg cyclosporine/kg/day. In a separate study, be…

Allergic contact dermatitis to propylene glycol in calcipotriene ointment.

A woman developed a pruritic exacerbation of her methotrexate-dependent psoriasis after applying topical calcipotriene ointment. Patch test reactions were positive to propylene glycol 1 percent and white petrolatum, but to none of the other ingredients of the medication. Physicians would be advised to patch test patients to both the therapeutic medication and the excipients of topical substances to which patients have adverse reactions. By so doing, an allergic reaction to one or more of the excipients may be found, thus the patient may be provided the benefit of the topical medication by usin…

Propylene glycol inactivates respiratory viruses and prevents airborne transmission.

Viruses are vulnerable as they transmit between hosts, and we aimed to exploit this critical window. We found that the ubiquitous, safe, inexpensive and biodegradable small molecule propylene glycol (PG) has robust virucidal activity. Propylene glycol rapidly inactivates a broad range of viruses including influenza A, SARS-CoV-2 and rotavirus and reduces disease burden in mice when administered intranasally at concentrations commonly found in nasal sprays. Most critically, vaporised PG efficiently abolishes influenza A virus and SARS-CoV-2 infectivity within airborne droplets, potently prevent…

Severe lactic acidosis after an iatrogenic propylene glycol overdose.

Propylene glycol is a diluent found in many intravenous and oral drugs, including phenytoin, diazepam, and lorazepam. Propylene glycol is eliminated from the body by oxidation through alcohol dehydrogenase to form lactic acid. Under normal conditions, the body converts lactate to pyruvate and metabolizes pyruvate through the Krebs cycle. Lactic acidosis has occurred in patients, often those with renal dysfunction, who were receiving prolonged infusions of drugs that contain propylene glycol as a diluent. We describe a 50-year-old man who experienced severe lactic acidosis after receiving an ac…
