Thyrotropin receptor knockout mice: studies on immunological tolerance to a major thyroid autoantigen.

Graves' disease involves a breakdown in self-tolerance to the TSH receptor (TSHR). Central T cell tolerance is established by intrathymic deletion of immature T lymphocytes that bind with high affinity to peptides from autoantigens (like the TSHR) expressed ectopically in the thymus. In TSHR-knockout mice, tolerance cannot be induced to the TSHR, which should, therefore, be a foreign antigen for these animals. To test this hypothesis, TSHR-knockout mice and wild-type controls were vaccinated (three injections) with TSHR DNA or control DNA. TSHR antibodies, measured by ELISA, binding to TSHR-ex

Antagonists of the human TSH receptor: in vitro and in vivo studies of their functional and immunological effects.

We have developed and characterized a prototype of a TSH receptor antagonist derived from the hCG molecule. This may be used to block human TSH receptor both functionally and immunologically, particularly in the study of Graves' disease. Our hCG derived TSH receptor blocker compares favorably with other substances (e.g. deglycosylated forms of TSH, synthetic peptides of the alpha or beta subunit of TSH) that hare been reported to inhibit bTSH binding or bTSH-stimulated cAMP response (Joshi et al., 1981; Morris et al., 1988). It has a much higher affinity for human TSH receptor than the TSH sub

[At the intersection of immunology and oncology: TAM receptors in the regulation of immune responses and tumorigenesis-related processes].

TAM receptor tyrosine kinases (TYRO3, AXL, MER) and their ligands, protein S (PROS1) and growth inhibition-specific protein 6 (GAS6), play a key role in maintaining homeostasis and regulating the immune response through involvement in efferocytosis, i.e., phagocytic removal of apoptotic cells and suppression of the innate immune response. Thus, their dysfunction leads, among others, to the development of autoimmune diseases. In turn, excessive production of TAM receptors correlates with the invasive phenotype of cancer cells, metastasis, drug resistance, and poor prognosis for patients with ca

Immunological signatures and predictive biomarkers for first-generation somatostatin receptor ligand resistance in Acromegaly.

PURPOSE: Predicting resistance to first-generation Somatostatin Receptor Ligands (fg-SRL) in Acromegaly patients remains an ongong challenge. Tumor-associated immune components participate in various pathological processes, including drug-resistance. We aimed to identify the immune components involved in resistance of fg-SRL, and to investigate biomarkers that can be targeted to treat those drug-resistant Acromegaly. METHODS: We conducted a retrospective study involving 35 Acromegaly patients with somatotropinomas treated postoperatively with fg-SRL. Gathering clinicopathological data, SSTR2

Targeting solid tumor antigens with chimeric receptors: cancer biology meets synthetic immunology.

Chimeric antigen receptor (CAR) T cell therapy is a medical breakthrough in the treatment of B cell malignancies. There is intensive focus on developing solid tumor-targeted CAR-T cell therapies. Although clinically approved CAR-T cell therapies target B cell lineage antigens, solid tumor targets include neoantigens and tumor-associated antigens (TAAs) with diverse roles in tumor biology. Multiple early-stage clinical trials now report encouraging signs of efficacy for CAR-T cell therapies that target solid tumors. We review the landscape of solid tumor target antigens from the perspective of

The implication of dendritic cells in lung diseases: Immunological role of toll-like receptor 4.

The immune responses play a profound role in the progression of lung lesions in both infectious and non-infectious diseases. Dendritic cells, as the "frontline" immune cells responsible for antigen presentation, set up a bridge between innate and adaptive immunity in the course of these diseases. Among the receptors equipped in dendritic cells, Toll-like receptors are a group of specialized receptors as one type of pattern recognition receptors, capable of sensing environmental signals including invading pathogens and self-antigens. Toll-like receptor 4, a pivotal member of the Toll-like recep

Binding, pharmacological and immunological profiles of the delta-selective opioid receptor antagonist HS 378.

HS 378 is a recently developed indolomorphinan with high selectivity and antagonist potency at the delta-opioid receptor. The present study was performed to characterize the opioid binding properties and pharmacological and immunological activity of HS 378 and to compare them with those of two well-known delta-opioid receptor antagonists, naltrindole (NTI) and naltriben (NTB). In vitro opioid receptor binding profiles were determined in rat brain homogenates. HS 378 showed 4.7- and 2.4-fold higher mu/delta selectivity compared to NTI and NTB, respectively. In the [35S]GTPgammaS functional assa

Immunological detection of muscarinic receptor subtype proteins (m1-m5) in rabbit peripheral tissues.

Employing subtype-specific antisera, we have measured the relative levels of five muscarinic receptor subtype proteins (m1-m5) in rabbit peripheral tissues. Immunoprecipitation assays demonstrated the presence of four distinct receptor proteins (m1-m4), which showed notable differences in their tissue distribution. Significant amounts of m5 receptors were not detected in any of the tissues examined. High levels of m1 receptors were found in sympathetic ganglia and submaxillary gland. The m1 receptor was the predominant subtype in vas deferens. Muscarinic receptors of the m2 type were present i

Optimizing the Hinge Length of Chimeric Antigen Receptor with a DdFP-Based Immunological Synapse Biosensor.

Chimeric antigen receptor (CAR)-T cell therapy has emerged as a transformative cancer immunotherapy, employing genetic engineering to express CARs in patient T cells. CARs are composed of a single-chain variable fragment (scFv), hinge, transmembrane, and intracellular signaling domains. While scFv directly recognizes tumor-associated antigens, the hinge domain connects the scFv to intracellular signaling domains, determining the formation of immunological synapse (IS). Because IS is critical for CAR activation, optimizing hinge length is crucial for successful CAR-T cell therapy. Here, we deve

Clinical and immunological responses after CD30-specific chimeric antigen receptor-redirected lymphocytes.

BACKGROUND: Targeting CD30 with monoclonal antibodies in Hodgkin lymphoma (HL) and anaplastic large cell lymphoma (ALCL) has had profound clinical success. However, adverse events, mainly mediated by the toxin component of the conjugated antibodies, cause treatment discontinuation in many patients. Targeting CD30 with T cells expressing a CD30-specific chimeric antigen receptor (CAR) may reduce the side effects and augment antitumor activity. METHODS: We conducted a phase I dose escalation study in which 9 patients with relapsed/refractory HL or ALCL were infused with autologous T cells that

The chemotactic receptor EBI2 regulates the homeostasis, localization and immunological function of splenic dendritic cells.

Spleen-resident dendritic cell (DC) populations occupy sentinel positions for the capture and presentation of blood-borne antigens. Here we found a difference in expression of the chemotactic receptor EBI2 (GPR183) on splenic DC subsets and that EBI2 regulated the positioning and homeostasis of DCs in the spleen. EBI2 and its main ligand, 7α,25-OHC, were required for the generation of the splenic CD4(+) DC subset and the localization of DCs in bridging channels. Absence of EBI2 from DCs resulted in defects in both the activation of CD4(+) T cells and the induction of antibody responses. Regula

Immunologic role of the cholinergic anti-inflammatory pathway and the nicotinic acetylcholine alpha 7 receptor.

Cytokines are small protein molecules that facilitate communication between cells of the immune system and other tissues. Their messenger function elicits responses in other cells, ranging from DNA binding and tissue remodeling to coordinating the local cellular response to inflammation. The magnitude of the cytokine response is regulated closely, because an over- or underabundance of cytokine activity can impair organ function and cause shock and tissue injury. Counterregulatory molecular and humoral mechanisms protect the host from cytokine excess, including the pituitary-adrenal-glucocortic

[Changes in the basic immunologic parameters interleukin-2 receptor and CD8-antigen in pre-eclampsia].

The serum level of interleukin-2 receptor (sIl-2R) and CD-8 antigen were determined as basic parameter in terms of the stimulation of the t-cell mediated immune reaction in patients suffering from pre-eclampsia (n = 21), HELLP syndrome (n = 12) and pre-existing hypertension (n = 10). The data were compared with those of healthy nonpregnant female volunteers (n = 10) and normotensive pregnant women in the course of pregnancy. Hypertension in pregnancy was associated with an increase of sIl-2R and sCD-8 levels depending on the severity of the hypertensive disorder. In the group of patients with

Immunological suppression by human CD8+ T cells is receptor dependent and HLA-DQ restricted.

Mechanisms of specific immunologic unresponsiveness or tolerance and their regulation by the major histocompatibility complex remain central issues in immunology. Recent findings that potentially reactive anti-self T cells are not completely clonally deleted in the thymus and that specific immunological unresponsiveness can be acquired in certain infectious diseases, such as leprosy, suggest that peripheral unresponsiveness can be developed and maintained in adults. Human antigen-specific T suppressor cells represent one mechanism of peripheral tolerance. Clones of CD8+ T suppressor cells have

Pan-cancer expression, methylation, and prognostic significance of α7 nicotinic acetylcholine receptor in tumor immunology.

The α7 nicotinic acetylcholine receptor (α7nAChR), functioning as a critical ligand-gated ion channel in the nervous system, has recently been implicated to play a role in various non-neuronal tissues. Nonetheless, a comprehensive understanding of its expression patterns, mechanisms of epigenetic control, and functions within the tumor microenvironment (TME) across various malignancies is still lacking. To address this gap, we performed an extensive multi-cancer investigation of CHRNA7 spanning 31 tumor types, leveraging datasets from The Cancer Genome Atlas and Genotype-Tissue Expression data

Immunologically programming the tumor microenvironment induces the pattern recognition receptor NLRC4-dependent antitumor immunity.

BACKGROUND: The efficacy of cancer immunotherapy can be limited by the poor immunogenicity of cancer and the immunosuppressive tumor microenvironment (TME). Immunologically programming the TME and creating an immune-inflamed tumor phenotype is critical for improving the immune-responsiveness of cancers. Here, we interrogate the immune modulator Flagrp170, engineered via incorporation of a pathogen-associated molecular pattern (ie, flagellin) into an immunostimulatory chaperone molecule, in transforming poorly immunogenic tumors and establishing a highly immunostimulatory milieu for immune augm

Immunological recognition of the prolactin receptor: identification of a single binding unit of molecular weight approximately 42,000.

Different polyclonal and monoclonal antibodies against the rabbit mammary prolactin (PRL) receptor were previously obtained that totally inhibited PRL binding in the rabbit mammary gland. Only polyclonal antibodies were shown to immunoprecipitate preformed PRL--receptor complexes in solubilized mammary membranes suggesting that they also recognized domains outside of the PRL binding site of the receptor. When partially purified PRL receptor preparations from both rabbit and pig mammary tissues were iodinated, immunoprecipitated and subsequently analyzed by SDS--PAGE, a single component of mole

Systemic testosterone induces structural, immunological, and sex steroid receptor changes in human XX skin.

Skin-resident cells express multiple sex steroid receptors, making the cutaneous microenvironment sensitive to hormonal fluctuations. Transgender individuals assigned female at birth receiving gender-affirming hormone treatment (GAHT) with systemic testosterone often undergo several surgical procedures, underscoring the importance of understanding how GAHT influences skin biology and wound-healing capacity. This study aimed to characterize how XX (genotype) skin responds to systemic testosterone exposure. Discarded skin samples from plastic surgeries were collected from XX patients with and wi
