Recent Developments in Recombinant Proteins for Diagnosis of Human Fascioliasis.

Fascioliasis is an important neglected tropical disease that causes severe injury to the bile ducts and liver. Therefore, a rapid and accurate method for detection of Fasciola hepatica infection plays a vital role in early treatment. Currently, the diagnosis of fascioliasis is mainly conducted via serological tests using the excretory/secretory (E/S) products, which might cross-react with antigens from other helminth parasitic diseases. Hence, the development of serodiagnosis test using recombinant antigens may contribute to differentiate fascioliasis from other helminth infections. In the pas

ELISA for brucellosis detection based on three Brucella recombinant proteins.

Control of brucellosis among farm animals, wildlife and humans require reliable diagnosis. Rose Bengal serological test (RBT) is based on lipopolysaccharide antigen of Brucella, which may cross react with other gram-negative bacteria and produce false positive result. Immunoreactive proteins, such as outer-membrane protein BP26, ribosome recycling factor protein CP24 and Brucella lumazine synthase (BLS), previously reported to be recognized by infected sheep sera, were selected for production of recombinant proteins for use in an ELISA in order to investigate immune response among goats and co…

Glycosylation control strategies for recombinant therapeutic proteins in CHO cells.

Chinese hamster ovary (CHO) cells are widely employed as host cells for the production of recombinant therapeutic proteins (RTPs). Glycosylation, a crucial post-translational modification, significantly affects the function and quality of RTPs expressed in CHO cells. This modification not only influences protein folding, stability, and immunogenicity, but also dictates their biological activity and half-life. This review summarizes the biological role of glycosylation within CHO cells, analyzes both intrinsic and extrinsic factors affecting recombinant protein glycosylation, and explores recen

Recombinant multiepitope proteins for hepatitis C virus diagnosis: Systematic review.

Hepatitis C virus (HCV) infection remains as major global health concern given the more than 50 million chronic cases recorded and the significant barriers to its early diagnosis, mainly in low-resource settings. Conventional serological assays rely on recombinant or synthetic antigens accounting for higher production costs and for limiting diagnostic accessibility. Recombinant multiepitope proteins have emerged as promising alternative for incorporating conserved antigenic regions from multiple HCV proteins, such as Core, NS3, NS4 and NS5, into a single construct capable of broadly detecting

Engineering Recombinant Chimeric Proteins That Deliver Therapies to the Brain.

To engineer biotherapeutics that can specifically target glioma, a highly lethal and recurrent type of brain tumor, we created a class of recombinant chimeric proteins containing multiple functional modules, such as a blood-brain barrier (BBB)-penetrating domain and an immune checkpoint blockade (ICB) agent, PD-L1 nanobody, through fusion protein engineering. The recombinant proteins that could prolong circulation time, cross the BBB, target brain tumors, penetrate cell membranes, and release PD-L1 nanobody were demonstrated. Further, by covalently linking doxorubicin (DOX), an inducer of immu…

Recombinant chimeric proteins offer new hope to control of leptospirosis.

Leptospirosis is an emerging zoonotic disease that causes over a million human cases and numerous fatalities annually, imposing substantial public health, economic, and veterinary burdens worldwide. Despite its impact, control strategies remain limited, as commercial bacterin vaccines offer only short-term, serovar-specific protection. The extensive genetic and antigenic diversity of Leptospira species presents a major challenge to the development of broadly protective vaccines. In response, several alternative strategies have been explored, including DNA vaccines, viral-vectored platforms and

Regeneration of the skeleton by recombinant human bone morphogenetic proteins.

Recombinant human bone morphogenetic proteins (rhBMPs) have past a long journey in human orthopaedic surgery during the last 15 years. From the first reports of the use of rhBMPs in hostile environments such as critically-sized bone defects, avascular femoral head necrosis, unstable thoracolumbar vertebral fractures, instability between the atlas and axis due to rheumatoid arthritis; over the use for nonunions of long bones and the scaphoid, reconstructive and revision surgeries of the hip, acute fractures, allograft nonunions, congenital pseudarthrosis, and various approaches of lumbar and ce

Recombinant human bone morphogenetic proteins in the treatment of fractures.

Recently, several prospective randomized clinical trials have resulted in the publication of critical studies on the efficacy of recombinant human morphogenetic proteins BMP-2 (rhBMP-2) and BMP-7 (OP-1) in spinal fusion and fracture healing. The clinical use of BMPs is now in its infancy and understanding the mechanism and the appropriate application of these proteins is necessary for all practicing orthopedic surgeons. This article will revisit some of the early studies using rhBMPs and review the current literature on their role in fracture healing.

Sensitive and specific serodiagnosis of onchocerciasis with recombinant hybrid proteins.

Onchocerciasis remains a major health hazard in many tropical countries. However, the existing tools for diagnosis of the disease have limitations, particularly regarding the detection of low level or early infections. To design an optimized reagent, we exploited the high antibody reactivity of patient sera against the Onchocerca volvulus proteins Ov20 and Ov33, which have been described as highly sensitive and specific immunodiagnostic reagents for producing hybrid proteins. The construct OvH2 was composed of Ov20 fused to Ov33, while OvH3 consisted of the C-terminus of Ov20 linked to Ov33. W

Recognition of Mycobacterium leprae recombinant 18-kDa proteins in leprosy.

Three different, purified, Escherichia coli-derived, recombinant preparations of the Mycobacterium leprae 18K protein were compared for their immunological recognition in leprosy. The preparations tested were 18K fusion proteins containing 70% (amino acids 38-148) of the full 18K protein fused to either a short leader sequence containing six asparagine residues or to beta-galactosidase, and the full length 18K protein. All three recombinant antigens were recognized by IgG antibodies which were restricted mostly to lepromatous leprosy patients. The 18K antigen with the asparagine leader sequenc

Characterizations of recombinant human tartrate-resistant acid phosphatase from osteosarcoma: comparison study between recombinant and placental proteins.

We cloned the human tartrate-resistant acid phosphatase (TRAP) gene from human osteosarcoma cells (Saos-2), and produced recombinant human TRAP (rhTRAP) using a baculovirus vector expression system. RhTRAP from Sf9 culture medium was purified by cation exchange chromatography, gel filtration and affinity chromatography. The molecular mass and amino acid composition of the rhTRAP were consistent with the deduced amino acid composition from the TRAP gene. The N-terminal amino acid sequence of rhTRAP was identical to that of TRAP purified from osteoclastoma and hairy cell leukemia spleen. The mon

Cell Engineering for Increasing Production of Recombinant Proteins in Mammalian Cells.

Mammalian cell lines are the preferred host cells for the biopharmaceutical industry. Chinese hamster ovary (CHO) and human embryonic kidney 293 (HEK293) cells are frequently utilized in the production of recombinant therapeutic proteins (RTPs) owing to their capacity to facilitate appropriate protein folding and perform accurate post-translational modifications (PTMs). However, there are still some bottlenecks in the biopharmaceutical process using mammalian cells, including lower productivity, higher production cost and increased risk of contamination compared with bacterial or yeast express

Somatic Embryogenesis: A Biotechnological Route in the Production of Recombinant Proteins.

Somatic embryogenesis (SE) is a morphogenetic pathway widely employed in the commercial micropropagation of plants. This route enables the generation of somatic embryos from somatic tissues, which give rise to complete (bipolar) plants that develop like zygotic embryos. SE can proceed via direct or indirect pathways, and both approaches have been adapted not only for large-scale clonal propagation but also for the regeneration of genetically modified plants. In this context, SE can be harnessed as a versatile platform for recombinant protein production, including vaccine antigens and therapeut

Anti-stroke biologics: from recombinant proteins to stem cells and organoids.

The use of biologics in various diseases has dramatically increased in recent years. Stroke, a cerebrovascular disease, is the second most common cause of death, and the leading cause of disability with high morbidity worldwide. For biologics applied in the treatment of acute ischaemic stroke, alteplase is the only thrombolytic agent. Meanwhile, current clinical trials show that two recombinant proteins, tenecteplase and non-immunogenic staphylokinase, are most promising as new thrombolytic agents for acute ischaemic stroke therapy. In addition, stem cell-based therapy, which uses stem cells o…

Potential of recombinant opa proteins as vaccine candidates against hyperinvasive meningococci.

Neisseria meningitidis causes half a million cases of septicemia and meningitis globally each year. The opacity (Opa) integral outer membrane proteins from N. meningitidis are polymorphic and highly immunogenic. Particular combinations of Opa proteins are associated with the hyperinvasive meningococcal lineages that have caused the majority of serogroup B and C meningococcal disease in industrialized countries over the last 60 years. For the first time, this genetic structuring of a diverse outer membrane protein family has been used to select a novel combination of representative antigens for
