Functional mutations in the thyroid-stimulating hormone receptor in natural stickleback populations at sites identical to human disease-causing mutations.

BACKGROUND: Thyroid hormones regulate multiple physiological functions, including metabolism, reproduction, and metamorphosis. Although there are variations in thyroid hormone signaling between populations and species, the causative mutations underlying these variations have rarely been identified. Here, we investigated whether information regarding the causative genes and mutations responsible for human thyroid diseases could assist with the identification of functional mutations in natural stickleback populations, which vary in thyroid hormone signaling between marine and stream-resident eco

Secretion of extracellular vesicles during ontogeny of the tapeworm Schistocephalus solidus.

We provide the first ultrastructural evidence of the secretion of extracellular vesicles (EVs) across all parasitic stages of the tapeworm Schistocephalus solidus (Müller, 1776) (Cestoda: Diphyllobothriidea) using a laboratory life cycle model. We confirmed the presence of EV-like bodies in all stages examined, including the hexacanth, procercoids in the copepod, Macrocyclops albidus (Jurine, 1820), plerocercoids from the body cavity of the three-spined stickleback, Gasterosteus aculeatus Linnaeus, and adults cultivated in artificial medium. In addition, we provide description of novel tegumen

Lipid and fatty acid composition of muscle, liver, ovary, and peritoneal fat in wild flathead grey mullet (Mugil cephalus) according to ovarian development.

Wild adult females of a low trophic omnivore teleost species, the flathead grey mullet (Mugil cephalus), caught in the western Mediterranean were sampled. The lipid and fatty acid composition of ovaries, liver, muscle, and peritoneal fat were analysed at previtellogenesis, early-vitellogenesis -first observed at mid-summer (early August)-, late-vitellogenesis, and the post-spawning period -from mid-September to mid-October-. During ovarian development, the lipid content of muscle was low and constant (3.85%-4.92%), indicating that the muscle was not used to store lipids for gonadal growth. Alt

Transcriptomic and epigenetic responses to androgen in throat skin tissue of the three-spined stickleback.

For sex-specific ornaments to be expressed only at a particular ontogenetic stage, genetic correlations between the sexes and between ontogenetic stages should be resolved. In vertebrates, sex-specific and ontogenetic stage-specific expression of sexual dimorphism is often mediated by sex steroid signalling. Here, we investigated the epigenetic mechanisms by which androgen induces sex-specific gene expression that may be involved in male-specific nuptial colouration in the three-spined stickleback (Gasterosteus aculeatus). First, we identified several androgen-responsive genes in skin tissues

Characteristic spermatogenesis process observed in Gasterosteidae: Considerations from histological observations of the testes and serum androgen levels of nine-spined stickleback, Pungitius sinensis.

To clarify the characteristics of spermatogenesis in Gasterosteidae, which are known to build nests for breeding, we conducted observations of gonadal tissues throughout the year and measured blood androgen (11-ketotestosterone; 11-KT) concentrations in the genus Pungitiu sinensis and compared the results with those of other genera. The presence of post-ovulatory follicles in the ovaries indicates that the breeding season for Pungitius is from April to August in Hokkaido. Spermatogenesis began in September, immediately after the breeding season, and is completed in October. Spermatozoa were th

Metabolic characterization of human sperm cells using the Seahorse metabolic flux analyzer.

BACKGROUND: The concerning trend on male infertility global prevalence, together with the unexplainable causes in half of those cases, highlights that there are still aspects of this disease to be understood and solved. To address this issue, one should not only be aware of the limitations of the implemented diagnostic tools, but also understand the sperm cell in depth, structurally, biochemically, molecularly in order to develop reliable and ready-to-be new/improved diagnostic tools. In this sense, the sperm cells metabolism, highly related to its functionality, seems to be a promising aspect

Elemental content of a host-parasite relationship in the threespine stickleback.

Parasite infections are ubiquitous and their effects on hosts could play a role in ecosystem processes. Ecological stoichiometry provides a framework to study linkages between consumers and their resource, such as parasites and their host, and ecosystem process; however, the stoichiometric traits of host-parasite associations are rarely quantified. Specifically, it is unclear whether parasites' elemental ratios closely resemble those of their host or if infection is related to host stoichiometry, especially in vertebrate hosts. To answer such questions, we measured the elemental content (%C, %

Co-occurrence and niche overlap among gill parasites of the white mullet (Mugil curema Valenciennes, 1836) (Osteichthyes: Mugilidae) from the western Atlantic, Brazil.

Different groups parasitize the gills of fishes, and the organisms can interact in positive, negative, or neutral ways, depending on parasitic abundance, environmental conditions, and the availability of resources. Here, we studied the distribution of ectoparasites on the gills of the mullet Mugil curema in the Cabedelo port region (Brazil), asking if (1) the gill parasites of M. curema fix on specific host gill arches and/or (2) co-occur in the same gill arches within a host individual; (3) if the abundance of one parasite taxon affects the abundance of another on the same gill arches; and (4

Seadragon genome analysis provides insights into its phenotype and sex determination locus.

The iconic phenotype of seadragons includes leaf-like appendages, a toothless tubular mouth, and male pregnancy involving incubation of fertilized eggs on an open "brood patch." We de novo-sequenced male and female genomes of the common seadragon (Phyllopteryx taeniolatus) and its closely related species, the alligator pipefish (Syngnathoides biaculeatus). Transcription profiles from an evolutionary novelty, the leaf-like appendages, show that a set of genes typically involved in fin development have been co-opted as well as an enrichment of transcripts for potential tissue repair and immune d

On the causes of geographically heterogeneous parallel evolution in sticklebacks.

The three-spined stickleback (Gasterosteus aculeatus) is an important model system for the study of parallel evolution in the wild, having repeatedly colonized and adapted to freshwater from the sea throughout the northern hemisphere. Previous studies identified numerous genomic regions showing consistent genetic differentiation between freshwater and marine ecotypes but these had typically limited geographic sampling and mostly focused on the Eastern Pacific region. We analysed population genomic data from global samples of the three-spined stickleback marine and freshwater ecotypes to detect

Moderate pathogenic effect of Ligophorus uruguayense (Monogenoidea, Ancyrocephalidae) in juvenile mullet Mugil liza (Actinopterygii, Mugilidae) from Brazil.

Monogenoidea pathogenic activity can elicit various histological responses in fish. Species of Ligophorus are specific parasites of mullets, and its relationship with host fish may result in a moderate pathogenic action. In order to ascertain this relationship, estuarine mullets (Mugil liza) were collected in an estuary, reared in laboratory, for three weeks, and forwarded for histological and parasitological analyses. Ligophorus uruguayense (Monogenoidea) infestation in the gills of the mullets was identified. The severe infestation by only one species of Monogenoidea may result from the spec

In situ experiments to assess effects of constraints linked to caging on ecotoxicity biomarkers of the three-spined stickleback (Gasterosteus aculeatus L.).

The aim of this study was to evaluate the effects of caging constraints on multiple fish biomarkers used during ecotoxicological studies (biometric data, immune and antioxidant systems, and energetic status). Two of these constraints were linked to caging: starvation and fish density in cages, and one in relation to the post-caging handling: a short transport. Three in situ experiments were conducted with three-spined sticklebacks (Gasterosteus aculeatus). The first experiment compared the effects of three densities (low, medium, and high). The second experiment compared effects of starvation

Comparative biochemical, transcriptome and metabolome investigation on reproductive maturation of wild and captive female Monopterus albus.

The main purpose of this study is to systematically elucidate mechanisms underlying ovarian development in wild and captive female Monopterus albus at the biochemical, transcriptomic, and metabolomic levels. Wild and captive female M. albus in Shanghai were continually collected over 1 year, and their ovarian development was analyzed statistically. We determined serum lipid metabolism and hormone contents of the fish during the study period, along with the largest differences in gonadosomatic indices (GSIs), and performed then transcriptome and metabolome detection analysis on their ovaries. Swamp eel aldehyde reductase is involved in response to nitrosative stress via regulating NO/GSH levels.

Aldehyde reductase (ALR) plays key roles in the detoxification of toxic aldehyde. In this study, the authors cloned the swamp eel ALR gene using rapid amplification of cDNA ends-PCR (RACE-PCR). The recombinant protein (rALR) was expressed in Escherichia coli and purified using a Ni2+ -NTA chelating column. The rALR protein exhibited efficient reductive activity towards several aldehydes, ketones and S-nitrosoglutathione (GSNO). A spot assay suggested that the recombinant E. coli strain expressing rALR showed better resistance to formaldehyde, sodium nitrite and GSNO stress, suggesting that swa

A CLUSTER OF CASES OF THYROID HYPERPLASIA IN AQUARIUM-HOUSED TROPICAL MARINE TELEOSTS FOLLOWING A CHANGE OF SALT MIX BRAND.

Follicular thyroid hyperplasia was diagnosed in nine out of 32 (28%) marine tropical teleosts housed in a public aquarium over a 9.5-mo period. These proliferative lesions were considered to be the cause of death in five of these fish. Iodine concentration was undetectable in nonozonized water (<0.005 mg/L), suggesting that an environmental iodine deficiency was the cause of these hyperplastic thyroid lesions. The only significant modification in the husbandry was a change, 18 mo before the first case, of the commercial salt mix brand used to make artificial seawater. The iodine content in thi

Gyrodactylus gasterostei a difficult meal to swallow for the three-spined sticklebacks, Gasterosteus aculeatus L.

This study examines the transmission strategies employed by parasites in order to colonize new hosts. Three-spined sticklebacks, Gasterosteus aculeatus L., were challenged with Gyrodactylus gasterostei Gläser, 1974 in a chamber and the colonization of new hosts was observed. Live-video recording indicates that gyrodactylid transmission possibly maximizes the chances of contact with an alternatively feeding response from dead parasitized hosts to predatory stickleback. It is proposed that host scavenge feeding may comprise an additional route for gyrodactylid transmission, although its relative

Markedly reduced prevalence of Schistocephalus solidus in pelagic three-spined sticklebacks (Gasterosteus aculeatus) from Lake Constance.

Host total length, body mass and prevalence of Schistocephalus solidus were assessed in pelagic three-spined sticklebacks (Gasterosteus aculeatus) from Upper Lake Constance, a prealpine lake in Central Europe, based on 987 individuals collected by pelagic trawl in late autumn 2024. In addition, 62 fish sampled using gillnets in late autumn 2025 were examined using the same biometric and parasitological procedures. The 2024 population was dominated by small-bodied individuals, with a mean total length of 5.26 ± 0.55 cm and a mean body mass of 1.56 ± 0.51 g. Infection was rare: 16 of 987 fish we
