Spermatogenesis is a multistep process that supports the production of millions of sperm daily. Understanding of the molecular mechanisms that regulate spermatogenesis has been a major focus for decades. Yet, the regulators involved in different cellular processes of spermatogenesis remain largely unknown. Human diseases that result in defective spermatogenesis have provided hints on the molecular mechanisms regulating this process. In this review, we have summarized recent findings on the function and signaling mechanisms of several genes that are known to be associated with disease or pathol

Male infertility affects one man in twenty and a genetic basis seems likely in at least 30% of those men. Genetic regulation of fertility involves the inter-related processes of testicular development, spermatogenesis (involving germ cell mitosis, meiosis and spermatid maturation), and their endocrine and paracrine regulation. In regard to spermatogenesis, particular attention has been given to the Yq11 region, where some spermatogenesis genes ('azoospermia factors') appear to be located. Several candidate genes have been identified but have not been shown to have a defined or essential role i

The results of a study of seminal fluid, chromosomal formula and testicular tissue performed on 32 patients with Klinefelter's syndrome are presented. The significance of the microscopic study of seminal fluid in this syndrome is well emphasized. The possible relationship of the histological pattern to spermatogenic function and chromosomal constitution is discussed. Our conclusions are that most patients were azoospermic and the the volume of the ejaculate was low or absent. The decreased density of seminal fluid was the result of the absence of spermatozoa. The seminal pH remained normal dur

Objective. To explore the effect of scrotal reconstruction following avulsion on spermatogenesis. Methods. Three patients were followed-up after having their scrotums reconstructed. Two scrotums were reconstructed by skin flap. Another patient's testicles were buried in thigh pockets. The follow-up continued from 3 months to 4 years after operation, and we examined the sperm quality, sexual function, sexual hormone, and testis biopsy. Results. The follow-up data showed that the spermatogenesis was not altered in the early stage (up to three months), but was substantially abnormal in the late s

SPATS1 (spermatogenesis-associated, serine-rich 1) is an evolutionarily conserved, testis-specific protein that is differentially expressed during rat male meiotic prophase. Some reports have suggested a link between SPATS1 underexpression/mutation and human pathologies such as male infertility and testicular cancer. Given the absence of functional studies, we generated a Spats1 loss-of-function mouse model using CRISPR/Cas9 technology. The phenotypic analysis showed no overt phenotype in Spats1-/- mice, with both males and females being fertile. Flow cytometry and histological analyses did no

Adult XYSxra mice exhibit varying degrees of spermatogenic deficiency but are usually fertile, while XOSxra mice have severe spermatogenic failure and are always sterile. The present quantitative spermatogenic analysis documents when these anomalies first appear during puberty. The results demonstrate that in XYSxra mice there was increased degeneration of pachytene spermatocytes and, to a lesser extent, meiotic metaphase stages. On average, there were only one-half the number of spermatids compared with the XY controls. The defect in XOSxra mice appeared a little later, with an almost complet

MM-1α is a c-Myc-binding protein and acts as a transcriptional co-repressor in the nucleus. MM-1α is also PDF5, a subunit of prefoldin that is chaperon comprised of six subunits and prevents misfolding of newly synthesized nascent polypeptides. Prefoldin also plays a role in quality control against protein aggregation. It has been reported that mice harboring the missense mutation L110R of MM-1α/PFD5 exhibit neurodegeneration in the cerebellum and also male infertility, but the phenotype of infertility has not been fully characterized. In this study, we first analyzed morphology of the testis

Measuring mRNA abundance at the level of the whole transcriptome has become an indispensable tool for the investigation of fundamental biological processes in higher eukaryotes. However, traditional transcriptomic methods are destructive and ignore the three-dimensional organization of cells within tissues and organs. Spatial transcriptomics is a cutting-edge technology that begins to address this limitation and facilitates transcriptomic analyses within the normal spatial context of two-dimensional tissue sections. This method permits a comprehensive appreciation of cellular heterogeneity acc

Adaptors are component proteins of junctional complexes in all epithelia, including the seminiferous epithelium of the mammalian testis. They recruit other regulatory and structural proteins to the site of both anchoring junctions (such as cell-cell actin-based adherens junctions [AJs], e.g., ectoplasmic specialization [ES] and tubulobulbar complex, which are both testis-specific cell-cell actin-based AJ types, and cell-cell intermediate filament-based desmosome-like junctions) and tight junctions (TJ). Furthermore, adaptors per se can be substrates and/or activators of kinases or phosphatases

Spermatogenesis is composed of three consecutive stages, mitosis, meiosis and spermiogenesis, during which spermatogenic cells undergo continuous molecular and cellular transformation. Regulation of gene expression has been underlying major molecular mechanisms that govern the progressive cell fate determination during spermatogenesis. Besides epigenetic and transcriptional controls, the un-coupled transcription and translation is a common phenomenon conserved across phyla during spermatogenesis. Translational regulation determines the dynamic proteomic landscapes in spermatogenic cells. Aberr

Spermatogenesis is a highly sophisticated process involved in the transmission of genetic heritage. It includes halving ploidy, repackaging of the chromatin for transport, and the equipment of developing spermatids and eventually spermatozoa with the advanced apparatus (e.g., tightly packed mitochondrial sheat in the mid piece, elongating of the tail, reduction of cytoplasmic volume) to elicit motility once they reach the epididymis. Mammalian spermatogenesis is divided into three phases. In the first the primitive germ cells or spermatogonia undergo a series of mitotic divisions. In the secon

The male gamete, the spermatozoon, is the carrier of paternal inheritance. Besides paternal genome, the spermatozoon contributes essential factors for successful fertilization, embryonic development, and continuation of species. A dynamic and extraordinarily complex process called spermatogenesis produces spermatozoa capable of maturing and acquiring full fertilizing capacity. Spermatogenesis proceeds through a series of events that involve cell division (proliferative phase assuring continuous restocking of spermatogenic stem cells), meiosis (recombination and haploidization of paternal genom

Spermatid production is a complex regulatory process in which coordination between hormonal control and apoptosis plays a pivotal role in maintaining a balanced number of sperm cells. Apoptosis in spermatogenesis is controlled by pro-apoptotic and anti-apoptotic molecules. Hormones involved in the apoptotic process during spermatogenesis include gonadotrophins, sex hormones, and glucocorticoid (GC). GC acts broadly as an apoptosis inducer by binding to its receptor (glucocorticoid receptor: GR) during organ development processes, such as spermatogenesis. However, the downstream pathway induced

The male reproductive system consists of testes, a series of ducts connecting the testes to the external urethral orifice, accessory sex glands, and the penis. Spermatogonial stem cells differentiate and mature in testes and epididymides, and spermatozoa are ejaculated with exocrine fluids secreted by accessory sex glands. Many studies have clarified the detailed structure and function of the male reproductive system, and have shown that various biologic controls, including genomics, epigenetics, and the neuroendocrine-immune system regulate proliferation, differentiation, and maturation of ge

Sperm production starts from puberty in the seminiferous tubules providing testosterone production by the Leydig cells that takes place in the interstice of the testicles. Normal spermatogenesis depends on specific signalling from the hypothalamic-pituitary-gonadal axis. GnRH, FSH and LH are the main hormones involved in the production and maturation of spermatozoa. Exogenous administration of androgens influences the hypothalamic- pituitary-gonadal axis with negative feedback that may lead to a partial or complete cessation of spermatogenesis by decreasing FSH and LH. Despite the fact that ma
