Tetracyclines have a broad range of clinical usefulness because of their broad antimicrobial spectrum of activity. For most routine gram positive and gram negative infections, alternative agents are available, but for Chlamydiae, Rickettsiae, Brucella, and Borrelia they still remain agents of choice. To some extent, gastrointestinal intolerance and inability to use these agents in patients with renal dysfunction have been overcome by the availability of doxycycline. Phototoxicity is a problem with this agent, however. Tetracycline is still useful as a sclerogenic agent for malignant effusions,…

Tetracyclines are well established antibiotics but show phototoxicity as a side effect. Antimicrobial photodynamic inactivation uses nontoxic dyes combined with harmless light to destroy microbial cells by reactive oxygen species. Tetracyclines (demeclocycline and doxycycline) can act as light-activated antibiotics by binding to bacterial cells and killing them only upon illumination. The remaining tetracyclines can prevent bacterial regrowth after illumination has ceased. Antimicrobial photodynamic inactivation can be potentiated by potassium iodide. Azide quenched the formation of iodine, bu…

Tetracyclines are a class of antibiotics which could act as neuroprotective molecules in several neurological disorders, such as Huntington disease, Parkinson disease, stroke and multiple sclerosis. The main biological effects of tetracyclines are the inhibition of microglial activation, the attenuation of apoptosis and the suppression of reactive oxygen species production. The anti-apoptotic effect of tetracyclines involves the mitochondrion, and the major target for neuroprotective effects of tetracyclines lies within the complex network that links mitochondria, oxidative stress and apoptosi…

In the last decade information has accumulated on the potential anti-prion activity of polycyclic compounds. Initially we showed that the antitumoral idodoxorubicin reduced the infectivity in experimental scrapie. On the basis of the chemical homology with anthracyclines, we rapidly moved to tetracyclines, compounds that are safer and widely used as antibiotics in clinical practice. The tetracyclines, essentially doxycycline and minocycline, were characterized as a therapeutical tool in transmissible spongiform encephalopathies (TSE) through the cell-free condition, in cellular and animal mode…

Tetracycline is one of the most widely used antibiotics. It may share some properties with conventional immunosuppressive drugs and act as an anti-inflammatory agent in the treatment of inflammatory disease. This article reviews cutaneous diseases that have been treated with tetracyclines and their antiinflammatory and immunosuppressive properties.

Macrolides, lincosamides, tetracyclines and chloramphenicol are structurally unrelated antibiotics which share protein synthesis inhibition as their common mechanism of action. Despite their individual differences, they can all be considered broad spectrum antibiotics with practical use for a wide variety of infections. Due to their similarities in function, however, concurrent or sequential administration of these agents must be undertaken with caution in order to prevent antagonism and induction of bacterial resistance. Full understanding of their function and potential interactions are, the…

Tetracyclines, whether natural or semisynthetic, act by inhibiting bacterial protein synthesis. These antibiotics are bacteriostatic and have a broad spectrum of activity. In the past, tetracyclines were discredited because of the high prevalence of isolates with acquired resistance, but they are now regaining status because of the incorporation of new, more active components. Doxycycline is currently the most commonly used tetracycline and is considered an essential drug by the World Health Organization. Tigecycline, a third-generation tetracycline, has a broader spectrum of activity and is a…

Tetracyclines are a family of antibiotics very common in clinical practice that have been used in not infectious affections. One of their most studied actions is their ability to inhibit matrix metalloproteinases (MMPs), a group of proteinases that have been implicated in pathological processes as oncogenesis and inflammation. Tetracyclines have been shown to play an important role in malignant angiogenesis and cancer invasion, which is related with tumor aggressiveness and metastatic potential. They also show anti-inflammatory activity in neurological, respiratory, bone and heart diseases, and…

Tetracycline and its derivatives, such as chlortetracycline, oxytetracycline, minocycline, doxycycline, methacycline and lymecycline, are naturally occurring or semi-synthetic polyketide compounds that exhibit a well known broad-spectrum antibacterial activity that interferes with prokaryotic protein synthesis at the ribosome level. In addition to this well known antibacterial activity these compounds also exhibit a variety of additional, less well known properties. Among them are separate and distinct anti-inflammatory properties. Tetracycline and related compounds have been shown to be effec

The tetracyclines are a family of related natural products that were originally discovered by virtue of their antibacterial activities. As one of the earliest antibiotics to be marketed after penicillin and streptomycin, and because of their convenient oral dosing, tetracyclines have achieved wide clinical usage. However, this widespread clinical use, in addition to their use in animal feed, and even as an antibiotic spray for fruit and other crops, has produced widespread resistance that ultimately has limited the clinical utility of the entire family of tetracycline antibiotics. More recentl…
