A rapid ELISA was developed for the detection of specific IgG against Micropolyspora faeni and Thermoactinomyces vulgaris and compared with counterimmunoelectrophoresis (CIE) in twenty-seven patients with suspected farmer's lung disease (FLD). Seventeen patients had precipitins to M. faeni or T. vulgaris or both, and ten had no precipitins. The optimum conditions for each step in the ELISA were determined: pre-equilibration of reagents at 37 C and vigorous agitation during incubation enabled the total test time required for the procedure to be reduced to 4 hr. A serum dilution of 10-fold produ…

Interactions of mouse alveolar macrophages from three different inbred strains of mice and Thermoactinomyces vulgaris, a microbe associated with Farmer's lung disease, were studied. Alveolar macrophages were found to abolish the mitogenic activity of T. vulgaris. A prostaglandin synthesis inhibitor indomethacin, could not restore the activity. Alveolar macrophage supernatants generated by T. vulgaris treatment exerted strong suppression in secondary concanavalin A-induced lymphocyte transformation. Indomethacin partly relieved the suppression but a histamine 2 receptor blocker, cimetidine, had…

The objectives of the study were to determine whether there was a significant risk of members of the work force of raw sugar mills developing bagassosis. Airborne Thermoactinomyces sacchari spores were measured to determine whether they were sufficient to cause acute bagassosis, and whether there was any evidence of previous exposure to sufficient airborne T. sacchari spores to cause the development of chronic bagassosis in any of the work force. Monitoring of total airborne bacteria spore concentrations was undertaken in and around two cane sugar mills before, during, and after the 1992 cane …

The aim of this study was to determine which precipitins against four antigens in 2,440 farmers are associated with the occurrence of farmer's lung (FL). The antigen panel consisted of mycelial antigens of Micropolyspora faeni, Thermoactinomyces vulgaris, Aspergillus fumigatus and Aspergillus umbrosus. As reference groups we used healthy farmers and those with chronic bronchitis. For the occurrence of precipitins against the four antigens there was a statistically significant difference between farmers with FL and healthy farmers but not between farmers with chronic bronchitis and healthy farm…

The extracellular growth products of Micropolyspora faeni, Thermoactinomyces vulgaris and Thermoactinomyces candidus were prepared by three methods; (1) double dialysis; (2) growth in tryptone soya broth (TSB) and (3) synthetic broth (AOAC). It was found that double dialysis was superior to growth in TSB or AOAC for the production of M. faeni antigens, as determined by immunoblot. Double dialysis, TSB and AOAC were similar for antigens of T. vulgaris and T. candidus. Both organisms showed bands of the same mobility on immunoblot and also produced lines of identity on double diffusion.

This study was based on a sample of 3,065 farmers from a larger survey population of 12,056 Finnish farmers. Data were gathered in a postal survey conducted by the Social Insurance Institution of Finland. Serum precipitins were determined by a microplate method of immune diffusion. The antigen panel consisted of mycelial antigens of Micropolyspora faeni, Thermoactinomyces vulgaris, Aspergillus fumigatus, and Aspergillus umbrosus. Geographical variation in the prevalence of precipitins was statistically significant, evidently due to climatic differences. The more intensive the cattle raising, t…

Antibodies produced in rabbits against Micropolyspora faeni, Thermoactinomyces vulgaris, T. sacchari, Thermoactinomyces candidus, and Saccharomonospora viridis were tested against antigens derived from many strains of thermophilic actinomycetes for precipitating antibodies by immunodiffusion test. It was found that immune sera reacted strongly against antigens from strains belonging to the same species and weakly against antigens from different species of thermophilic actinomycetes. However, sera from farmer's lung patients showed cross-reactivity against antigens from different species. This …

Sera from individuals exposed to moldy hay, mushroom compost and unexposed individuals, were studied for antibodies against thermophilic actinomycetes and Aspergillus fumigatus. Antigens extracted from Thermoactinomyces vulgaris strains isolated from China were used in the study along with antigens from standard strains to screen the sera of individuals exposed to farms and unexposed controls from both China and the United States. It was found that very little antibody response to A. fumigatus and F. rectivirgula was demonstrated in the sera of both individuals exposed to farm environments and…

This article reports on an epidemiological survey of the presence of farmer's lung among 1054 farmers who grind moldy hay. The prevalence was 8.3-11.4%. The main pathogen was Thermoactinomyces vulgaris. The factors which influenced farmer's lung were the moldy state of the hay and the dust concentration. The epidemiological features of farmer's lung are analyzed and discussed and certain preventive measures are suggested.

The prevalence of bagassosis was investigated in a Louisiana paper mill, which in the past had considerable numbers of workers with the disease. Based on negative clinical histories and the low number of positive serological reactions of the workers' serum with Thermoactinomyces sacchari antigen, it was concluded that bagassosis was no longer present. This was thought to be due to a different method of storage of bagasse, which retards microbial decay and reduces airborne organic dust, and to the increased awareness of plant management resulting in greater safety measures.

The actinomycetes Micropolyspora faeni and Thermoactinomyces vulgaris are important sources of antigens in extrinsic allergic alveolitis, the deuteromycete Aspergillus fumigatus is it to a lesser extent. These microorganisms have been sought in hay samples obtained from farmers who on the one hand show farmer's lung symptoms and on the other antibodies to these microorganisms. A relatively good correlation has been found between the immunological and microbiological findings. The conditions necessary for the occurrence of antigen sources in farmer's lung disease are discussed, and the need for…

Sera from few Finnish patients with clinical farmers' lung react in precipitin tests with extracts of the thermophilic actinomycetes that commonly cause the disease elsewhere. Hays associated with the disease in Finland showed less evidence of spontaneous heating and contained fewer actinomycete spores than British hays. Only Thermoactinomyces vulgaris was sometimes abundant. Some species of mesophilic fungi were more abundant than in Britain and one, Aspergillus umbrosus, reacted with most sera from farmers' lung patients in precipitin tests. A panel of antigens, including thermophilic actino…

Recent studies suggest that besides the long-known farmer's lung antigen sources Saccharopolyspora rectivirgula (Micropolyspora faeni), Thermoactinomyces vulgaris, and Aspergillus fumigatus, additionally the mold Absidia (Lichtheimia) corymbifera as well as the bacteria Erwinia herbicola (Pantoea agglomerans) and Streptomyces albus may cause farmer's lung in Germany. In this study the sera of 64 farmers with a suspicion of farmer's lung were examined for the following further antigens: Wallemia sebi, Cladosporium herbarum, Aspergillus versicolor, and Eurotium amstelodami. Our results indicate …

Farmer's lung, caused by the inhalation of microspores--particularly of the genus, Thermoactinomyces--has been recognised for the past 30-40 years as a condition affecting adult farm workers, especially up to the mid-sixties when undried crops were still threshed indoors. Although it used to be extremely rare in children, between 1985 and 1989 occurred five of the seven cases reported hitherto. The possibility is suggested that with the increasingly large scale use of grain drying plant and stubble burn-off on Danish farms, we shall see an increase in the incidence of the disease, even in infa…

The prevalence of serum-precipitating antibodies to Micropolyspora faeni and Thermoactinomyces vulgaris was studied by immunoelectrophoresis in 442 dairy farmers living in Hokkaido, the northernmost district of Japan. The prevalence rates of antibodies to M. faeni and T. vulgaris were 24.2 and 11.6%, respectively. The rate of antibodies to M. faeni was higher among females than males. This difference may be due to the large number of nonsmokers among females. Prevalence of antibodies to M. faeni or M. faeni and T. vulgaris was associated with a higher hay acreage and longer working hours per d…

In this study we have investigated a number of technical aspects of a sensitive solid-phase radioimmunoassay (microtitre-plate RIA) for the quantitative determination of antigen-specific IgG antibodies in the sera of patients suffering from farmer's lung. Microtitre-plates were coated with antigens of Aspergillus fumigatus, Micropolyspora faeni, Aureobasidium pullulans, Alternaria tenuis, Thermoactinomyces vulgaris and hay. Optimal antigen concentration and serum dilution were determined, the temperature-dependence of the assay and the kinetics of the binding of antibodies and protein A were s…
