At low dose, arginine vasotocin has vasopressor rather than vasodepressor effect in chickens.

It is widely accepted that both arginine vasopressin (AVP), and its avian analogue, arginine vasotocin (AVT) are vasopressor (VP) in mammals, but vasodepressor (VDP) in birds. The rat VP and chicken VDP assays were used to characterize neurohypophysial peptides and their analogues. However, these assays use pharmacological doses. Like AVP in mammals, AVT is secreted in birds during dehydration, when peripheral vasoconstriction rather than vasodilatation is needed. Mature White Leghorn cockerels with permanent cannulas in brachial artery and vein were put restrained in a sling. Mean arterial pr

Arginine vasotocin is the major adrenocorticotropic hormone-releasing factor in the bullfrog Rana catesbeiana.

In a previous study, we showed that corticotropin-releasing factor (CRF) is the major thyroid-stimulating hormone (TSH)-releasing factor in the bullfrog (Rana catesbeiana) hypothalamus. Our findings prompted us to ascertain whether CRF or arginine vasotocin (AVT), a known adrenocorticotropic hormone (ACTH) secretagogue in several vertebrates, is the main stimulator of the release of ACTH from the bullfrog pituitary. Both the frog CRF and AVT stimulated the release of immunoassayable ACTH from dispersed anterior pituitary cells in vitro in a concentration-dependent manner. AVT, however, exhibit

Brain levels of arginine-vasotocin and isotocin in dominant and subordinate males of a cichlid fish.

The nonapeptides arginine-vasotocin (AVT) and isotocin (IT), which are the teleost homologues of arginine-vasopressin and oxytocin in mammals, have well established peripheral effects on osmoregulation and stress response, and central effects on social behavior. However, all studies that have looked so far into the relationship between these nonapeptides and social behavior have used indirect measures of AVT/IT activity (i.e. immunohistochemistry of AVT/IT immunoreactive neurons, or AVT/IT or their receptors mRNA expression with in situ hybridization or qPCR) and therefore direct measures of p

Genetic pathways linking oxytocin-vasotocin hypothalamic subunit architecture with psychiatric and metabolic traits.

The neuropeptides oxytocin and vasotocin are predominantly produced in the supraoptic and paraventricular nuclei of the anterior-inferior, anterior-superior and tubular-superior hypothalamic subunits. Evidence suggests that oxytocin and vasotocin signaling play a role in both physiology and behavior, and that dysfunction of these signaling systems may contribute to the co-occurrence of metabolic and psychiatric conditions. The genetic pathways, however, that may underlie the connection between these physiological and behavioral traits are yet to be clearly delineated. We deployed bivariate mix

Arginine vasotocin mRNA revealed by in situ hybridization in bovine pineal gland cells.

Arginine vasopressin (AVP) is the main antidiuretic hormone in mammals and arginine vasotocin (AVT) in submammalian vertebrates. The possibility that the genetic material encoding AVT is maintained in mammals is controversial. In this study, we investigated by radioactive in situ hybridization the possible presence of the mRNA encoding AVP and AVT, and using immunocytochemistry the presence of structures immunoreactive for AVP and AVT in the bovine pineal gland. In situ hybridization was performed by use of 35S-labelled oligoprobes. Immunocytochemistry was performed using specific polyclonal r

Isolation and structure elucidation of bovine pineal arginine vasopressin: arginine vasotocin not identified.

A large number of reports have demonstrated the presence of neurohypophysial hormone-like peptides in mammalian pineal glands and an antigonadotropic function has been ascribed to pineal arginine vasotocin (AVT). We have undertaken large scale purification of bovine pineal neurohypophysial hormone-like substances which demonstrate mouse mammary milk-ejection activity (ME-activity) in vitro. Peptides with ME-activity were extracted from more than 5 kg of bovine pineal glands. ME-activity containing peptides were found in both high (Mr approximately 10,000-15,000) and low (Mr approximately 500-1

Distribution of the vasotocin type 4 receptor throughout the brain of the chicken, Gallus gallus.

The vasopressin 1a receptor (V1aR) has been shown to have a wide distribution throughout the mammalian brain and pituitary gland and mediates a number of physiological functions as well as social behavior following the binding of its agonist, vasopressin. The avian receptor homologous to the V1aR is the vasotocin 4 receptor (VT4R). Its mRNA distribution has been documented in brain regions of two species of songbird; however, its complete protein distribution in the brain has not been published to date for any avian species. The present work utilizes an antibody made to a sequence of the chick

Presence of relatively high concentrations of arginine vasotocin in the cerebrospinal fluid of newborns and infants.

Lumbar cerebrospinal fluid (CSF) was obtained from healthy newborns, infants, and children of both sexes between 23-777 days of age. Bioassay of the CSF revealed relatively high antidiuretic and hydroosmotic activities. Rat uterine activities could be detected only in the CSF of newborns, and these activities were increased in the presence of magnesium. The antidiuretic and hydroosmotic activities of CSF from newborns were significantly higher than the same CSF activities of infants (P less than 0.01) and children (P less than 0.001). No sex differences were observed in the above biological ac

Arginine Vasotocin and Neuropeptide Y Vary with Seasonal Life-History Transitions in Garter Snakes.

Transitions between life-history stages are often accompanied by dramatic behavioral switches that result from a shift in motivation to pursue one resource over another. While the neuroendocrine mechanisms that regulate such behavioral transitions are poorly understood, arginine vasotocin (AVT) and neuropeptide Y (NPY) are excellent candidates because they modulate reproductive and feeding behavior, respectively. We asked if seasonal changes in AVT and NPY are concomitant with the seasonal migration to and from the feeding grounds in red-sided garter snakes (Thamnophis sirtalis parietalis). Ma

Pineal modulation of the rat caudate-putamen spontaneous neuronal activity: roles of melatonin and vasotocin.

The effects of microiontophoretic application of melatonin and melatonin plus vasotocin on spontaneously active neurons of caudate-putamen in sham-operated and pinealectomized rats were studied. Extracellular unit recordings showed that in sham-pinealectomized rats, melatonin ejection primarily produced inhibition of the responsive neurons (74.1%), whereas only 24.9% of the neurons were excited. Iontophoretic ejection of vasotocin or melatonin+vasotocin produced, in both cases, an inhibition of 100% of the responsive neurons. In pinealectomized rats, iontophoretic melatonin ejection produced a

Vasotocin, melatonin and narcolepsy: possible involvement of the pineal gland in its patho-physiological mechanism.

Both the pineal nonapeptide hormone arginine vasotocin (AVT) (2.5 micrograms) administered intra-nasally and the pineal indole melatonin (50 mg) administered intravenously to three male narcoleptics (two with auxiliary symptoms and one with sleep attacks only), dramatically increased the amount of REM sleep and decreased REM sleep latency. The duration of the sleep onset REM periods in the two narcoleptics with auxiliary symptoms increased by more than 100 percent after AVT and melatonin administration. In the narcoleptic with sleep attacks only both AVT and melatonin induced REM periods at sl

The Neural Distribution of Vasotocin, Oxytocin, Dopamine, and Serotonin in Two Australian Skinks With Contrasting Social Lives.

The neurotransmitters vasotocin, oxytocin, dopamine, and serotonin are widely involved in vertebrate social behavior, and changes in their abundance and distribution in the brain have been linked to the evolution of complex sociality. Reptiles provide an excellent system in which to investigate the neural mechanisms of social living. Using immunohistochemistry, we compare distributions of these transmitters in two skinks differing primarily in social ecology: the family-living Liopholis whitii and the solitary Eulpamrus quoyii. We describe patterns of immunopositive signal for both cell bodies

Vasotocin and oxytocin modulation of the endocrine and behavioral response to an aggressive challenge in male Siamese fighting fish.

Aggressive behavior is an adaptive trait present across all taxa. However, the neuroendocrine mechanisms regulating it, particularly in fish, are not well understood. Oxytocin (OXT) and arginine vasotocin (VT) are known modulators of aggression, but their actions remain controversial. This study tested the possible modulation of endocrine and behavioral responses to an aggression challenge by these nonapeptides in Siamese fighting fish, Betta splendens, a species known for its intrinsic aggressiveness. Male B. splendens were injected with different dosages of either Manning compound or L-368,8

In ovo exposure to ethynylestradiol demasculinizes the vasotocin system in the medial preoptic nucleus in male Japanese quail.

Ethynylestradiol (EE2) is an estrogen that is used predominantly in oral contraceptives and contaminates the environment. In ovo exposure to EE2 impairs copulatory behavior in male Japanese quail, although the mechanisms responsible remain undetermined. The medial preoptic nucleus (POM) plays a pivotal role in male copulatory behaviors and is sexually dimorphic, with a higher number of vasotocin (VT) neuronal fibers in males. In this study, the effects of in ovo EE2 exposure on the VT neuronal fibers of the POM were examined to test the hypothesis that impaired copulatory behaviors caused by E

Regulation of gene expression of vasotocin and corticotropin-releasing hormone receptors in the avian anterior pituitary by corticosterone.

The effect of chronic stress (CS) on gene expression of the chicken arginine vasotocin (AVT) and corticotropin-releasing hormone (CRH) receptors [VT2R, VT4R, CRH-R1, and CRH-R2] was examined by measuring receptor mRNA levels in the anterior pituitary gland of the chicken after chronic immobilization stress compared to acute stress (AS). Radioimmunoassay results showed that blood circulating corticosterone (CORT) levels in the CS group were significantly decreased compared to that of birds in the AS group (P<0.05). The VT2R and CRH-R2 mRNA in CS birds were significantly decreased to that of con

African lungfish, Protopterus annectens, possess an arginine vasotocin receptor homologous to the tetrapod V2-type receptor.

In tetrapods, arginine vasopressin and its counterpart, arginine vasotocin (AVT), are involved in renal water conservation through vascular V1a-type and tubular V2-type receptors, and only the former has thus far been cloned in fish. We successfully cloned the V1a-type and V2-type AVT receptor from the kidney of the African lungfish, Protopterus annectens, and the deduced amino acid sequences exhibited high homology with amphibian V1a- and V2-type receptors, respectively. Functional analysis showed that AVT addition to CHO cells transfected with lungfish V1a-type receptor increased [Ca2+]i in

The arginine-vasotocin and serotonergic systems affect interspecific social behaviour of client fish in marine cleaning mutualism.

Many species engage in mutualistic relationships with other species. The physiological mechanisms that affect the course of such social interactions are little understood. In the cleaning mutualism, cleaner fish Labroides dimidiatus do not always act cooperatively by eating ectoparasites, but sometimes cheat by taking bites of mucus from so-called "client" reef fish. The physiological mechanisms in these interspecific interactions, however, are little studied. Here, we focussed on three neuromodulator systems known to play important roles in intraspecific social behaviour of vertebrates to exa

Arginine vasotocin activates phosphoinositide signal transduction system and potentiates N-acetyltransferase activity in the rat pineal gland.

The pineal gland is innervated by pinealopetal peptidergic fibers originating in the hypothalamic nuclei which release arginine vasopressin (AVP) and arginine vasotocin (AVT) from their endings. Since the mechanism of AVT action on the pineal signal transduction and melatonin synthesis has not been determined so far, we examined the effect of AVT on the phosphoinositide signalling system and the N-acetyltransferase (NAT) activity in the rat pineal gland. The effect of AVP 4-9 fragment and AVP analogue desmopressin was also tested. The phosphoinositide signalling system was studied by measuring

Arginine vasotocin neuronal phenotypes and their relationship to aggressive behavior in the territorial monogamous multiband butterflyfish, Chaetodon multicinctus.

Intra and interspecific comparisons of arginine vasotocin (AVT) and its mammalian homolog arginine vasopressin (AVP) demonstrate several relationships between these neuropeptides and aggression/dominance behaviors. Prior studies in coral reef butterflyfishes and other fishes indicate that features of AVT neurons in the gigantocellular preoptic area (gPOA) and axon varicosities within the ventral nucleus of the ventral telencephalon should have a positive relationship with aggressive behavior, whereas AVT-ir neuronal features in the parvocellular preoptic area (pPOA) should have a negative rela
