Function and transcription of stoS gene of Xanthomonas campestris pathovar campestris.

StoS is a hybrid histidine kinase that, to date, has been investigated only in Xanthomonas oryzae pathovar oryzae. In this study, we elucidated the function and transcriptional regulation of the stoS gene in X. campestris pathovar campestris (Xcc). An stoS deletion mutant and its complemented strain were constructed to assess phenotypic characteristics. Inactivation of StoS markedly decreased tolerance to heat, sodium dodecyl sulfate, and hydrogen peroxide, impaired membrane integrity, and reduced stationary-phase survival, whereas complementation restored these phenotypes. Transcriptional map…

Function and transcription of the lon gene in Xanthomonas campestris pv. campestris.

The Gram-negative bacterium Xanthomonas campestris pv. campestris is the etiological agent of black rot, a widespread and destructive disease affecting cruciferous plants. In this study, a lon mutant was obtained by EZ-Tn5 transposon mutagenesis of the X. campestris pv. campestris. The lon gene encodes an ATP-dependent protease implicated in protein quality control and stress adaptation across various bacterial species. Functional analysis revealed that lon disruption in X. campestris pv. campestris resulted in diminished extracellular protease activity, reduced virulence, and heightened sensi…

A Small Molecule Inhibits Xanthomonas oryzae pv. oryzae by Targeting MenG.

Bacterial leaf blight (BLB), caused by Xanthomonas oryzae pv. oryzae (Xoo), remains a major threat to global rice production. In this study, we identified the novel small-molecule compound PK150 with potent antibacterial activity against Xoo. PK150 exhibits a minimum inhibitory concentration (MIC) of 0.15 μg/mL in vitro, and achieved 78% protective efficacy at 200 μg/mL in planta. Mechanistic studies revealed that PK150 targets menaquinone biosynthesis by binding to demethylmenaquinone methyltransferase (MenG), which is a key enzyme in this pathway. Surface plasmon resonance (SPR) analysis con…

Antibacterial activity of monoacetylated alkyl gallates against Xanthomonas citri subsp. citri.

Asiatic citrus canker (ACC) is an incurable disease of citrus plants caused by the Gram-negative bacterium Xanthomonas citri subsp. citri (X. citri). It affects all the commercially important citrus varieties in the major orange producing areas around the world. Control of the pathogen requires recurrent sprays of copper formulations that accumulate in soil and water reservoirs. Here, we describe the improvement of the alkyl gallates, which are potent anti-X. citri compounds, intended to be used as alternatives to copper in the control of ACC. Acetylation of alkyl gallates increased their lipo…

Population Structure of Xanthomonas citri pv. citri in Iran: 1991 to 2022.

Citrus canker, caused by Xanthomonas citri pv. citri, is one of the most devastating bacterial diseases of citrus species. The global population of X. citri pv. citri includes three lineages designated as pathotypes A, A*, and Aw. Whereas pathotype A is the most prevalent lineage around the world, the citrus canker pathogen in Iran includes only pathotype A* strains. Previous work on the X. citri pv. citri strains collected before 2013 showed that two lineages of the pathogen were present in Iran (i.e., 4.1 and 4.4, with 4.4 not present anywhere else at that time). In this study, using a new s…

Efficient and Rapid Bioassay for Xanthomonas citri pv. punicae Blight on Pomegranate.

It is crucial that experiments dealing with plant-pathogen interactions are robust and reproducible. For evaluating the disease-resistance ability of a host plant against any pathogen, controlled conditions are a prerequisite that cannot be obtained with soil-based systems. Variation in environmental conditions during challenge inoculation of a pathogen, when performed in fields or a polyhouse, may lead to spurious results. This is of particular importance when dealing with disease resistance in a fruit tree like pomegranate, as determined after years of research efforts. Due to lack of unifor

Synergistic Effect of Essential Oils and Rhamnolipid on Xanthomonas citri Subsp. citri.

Citrus canker, caused by Xanthomonas citri subsp. citri, is a devastating disease that affects citrus production and trade worldwide. Traditional control methods, based on copper compounds, are effective but pose environmental and health risks due to their toxicity and potential for bioaccumulation. This study evaluates the synergistic potential of essential oils (EOs) and rhamnolipids as sustainable alternatives for disease management. Four EOS (citronella, palmarosa, geranium, and clove) were tested for their antibacterial activity. Citronella EO showed a 90% inhibitory concentration (IC 90)…

A Specific High Toxicity of Xinjunan (Dioctyldiethylenetriamine) to Xanthomonas by Affecting the Iron Metabolism.

Xanthomonas spp. encompass a wide range of phytopathogens that brings great economic losses to various crops. Rational use of pesticides is one of the effective means to control the diseases. Xinjunan (Dioctyldiethylenetriamine) is structurally unrelated to traditional bactericides, and is used to control fungal, bacterial, and viral diseases with their unknown mode of actions. Here, we found that Xinjunan had a specific high toxicity toward Xanthomonas spp., especially to the Xanthomonas oryzae pv. oryzae (Xoo), the causal agent of rice bacterial leaf blight. Transmission electron microscope …

Antibacterial effect of Cu2O/TiO2 visible-light photocatalytic composite on Xanthomonas campestris.

In this study, a Cu2O/TiO2 (CuTi) visible-light photocatalytic composite was employed for the treatment of Xanthomonas campestris and X. campestris-infected Brassica napus seedlings. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values against X. campestris were determined to be 8 and 32 μg ml-1, respectively. Transmission electron microscopy analysis demonstrated a direct correlation between the extent of bacterial cell damage and the concentration of CuTi. Noteworthily, a bactericidal rate of 100% was achieved at a concentration of 150 μg ml-1 over a…

3D Structure Modeling of Alpha-Amino Acid Ester Hydrolase from Xanthomonas rubrilineans.

Alpha-amino acid ester hydrolase (EC 3.1.1.43, AEH) is a promising biocatalyst for the production of semi-synthetic β-lactam antibiotics, penicillins and cephalosporins. The AEH gene from Xanthomonas rubrilineans (XrAEH) was recently cloned in this laboratory. The three-dimensional structure of XrAEH was simulated using the homology modeling method for rational design experiments. The analysis of the active site was performed, and its structure was specified. The key amino acid residues in the active site - the catalytic triad (Ser175, His341 and Asp308), oxyanion hole (Tyr83 and Tyr176), and …

Two-component signal transduction systems of Xanthomonas spp.: a lesson from genomics.

The two-component signal transduction systems (TCSTSs), consisting of a histidine kinase sensor (HK) and a response regulator (RR), are the dominant molecular mechanisms by which prokaryotes sense and respond to environmental stimuli. Genomes of Xanthomonas generally contain a large repertoire of TCSTS genes (approximately 92 to 121 for each genome), which encode diverse structural groups of HKs and RRs. Among them, although a core set of 70 TCSTS genes (about two-thirds in total) which accumulates point mutations with a slow rate are shared by these genomes, the other genes, especially hybrid…

Epidemiology, diversity, and management of bacterial spot of tomato caused by Xanthomonas perforans.

Tomato is an important crop grown worldwide. Various plant diseases cause massive losses in tomato plants due to diverse biotic agents. Bacterial spot of tomato (BST) is a worldwide disease that results in high losses in processed and fresh tomato. Xanthomonas perforans, an aerobic, single-flagellated, rod-shaped, Gram-negative plant pathogenic bacterium, is one of the leading causes of BST. Over the past three decades, X. perforans has increasingly been reported from tomato-growing regions and became a major bacterial disease. X. perforans thrives under high humidity and high temperature, wh…

Pan-genome and antibiotic resistance insights into Xanthomonas citri pv. punicae pathotypes.

BACKGROUND: Bacterial blight, caused by Xanthomonas citri pv. punicae (Xcp) is a significant threat to pomegranate cultivation in India. Although antibiotics such as streptomycin, streptocycline, and tetracycline are commonly used for its management, their effectiveness is declining due to the emergence of resistant Xcp strains carrying acquired resistance genes. Considering this growing concern, the present study aimed to investigate nine Xcp isolates obtained from pomegranate-growing regions of India by assessing their pathogenicity, followed by in vitro evaluation of their antibiotic sensit…

Xanthomonas encodes for a novel dodecameric ferritin-like protein with unique structural features.

Iron is considered an essential component of all life forms, but is maintained optimally by fine-tuning of iron homeostasis mechanisms. Iron storage proteins (ISPs), which are ferritin superfamily proteins, play a pivotal role in maintaining iron homeostasis by storing excess cellular iron, often serving as ROS scavengers for protecting cells from oxidative stress. Xanthomonas represents a large and ubiquitous group of phytopathogenic bacteria that causes severe crop loss worldwide. Xanthomonas survives host-imposed iron stress by governing iron uptake, transport, and metabolism in utmost coor…

Plant responses underlying nonhost resistance of Citrus limon against Xanthomonas campestris pv. campestris.

Citrus is an economically important fruit crop that is severely afflicted by citrus canker, a disease caused by Xanthomonas citri ssp. citri (X. citri); thus, new sustainable strategies to manage this disease are needed. Although all Citrus spp. are susceptible to this pathogen, they are resistant to other Xanthomonas species, exhibiting non-host resistance (NHR), for example, to the brassica pathogen X. campestris pv. campestris (Xcc) and a gene-for-gene host defence response (HDR) to the canker-causing X. fuscans ssp. aurantifolii (Xfa) strain C. Here, we examine the plant factors associated…

Non-host defense response in a novel Arabidopsis-Xanthomonas citri subsp. citri pathosystem.

Citrus canker, caused by Xanthomonas citri subsp. citri (Xcc), is one of the most destructive diseases of citrus. Progress of breeding citrus canker-resistant varieties is modest due to limited resistant germplasm resources and lack of candidate genes for genetic manipulation. The objective of this study is to establish a novel heterologous pathosystem between Xcc and the well-established model plant Arabidopsis thaliana for defense mechanism dissection and resistance gene identification. Our results indicate that Xcc bacteria neither grow nor decline in Arabidopsis, but induce multiple defens…

The Role of ClpV in the Physiology and Pathogenicity of Xanthomonas citri subsp. citri Strain zlm1908.

Xanthomonas citri subsp. citri (Xcc) is a Gram-negative bacterium responsible for citrus canker, a significant threat to citrus crops. ClpV is a critical protein in the type VI secretion system (T6SS) as an ATPase involved in bacterial motility, adhesion, and pathogenesis to the host for some pathogenic bacteria. In order to investigate the function of clpV gene in Xcc, the clpV-deletion strain ΔclpV was constructed, its biological properties were evaluated, and the differences in gene expression levels between the wild-type strain and ΔclpV were analyzed by transcriptomics. The results exhibi…
