Airborne concentrations of peanut protein.

Food allergy to peanut is a significant health problem, and there are reported allergic reactions to peanuts despite not eating or having physical contact with peanuts. It is presumed that an allergic reaction may have occurred from inhalation of airborne peanut allergens. The purpose of this study was to detect the possible concentrations of airborne peanut proteins for various preparations and during specific activities. Separate Ara h 1 and Ara h 2 monoclonal enzyme-linked immunosorbent assays and a polyclonal sandwich enzyme immunoassay for peanuts were used to detect the amount of airborn

Autoimmune amelogenesis imperfecta in patients with APS-1 and coeliac disease.

Ameloblasts are specialized epithelial cells in the jaw that have an indispensable role in tooth enamel formation-amelogenesis1. Amelogenesis depends on multiple ameloblast-derived proteins that function as a scaffold for hydroxyapatite crystals. The loss of function of ameloblast-derived proteins results in a group of rare congenital disorders called amelogenesis imperfecta2. Defects in enamel formation are also found in patients with autoimmune polyglandular syndrome type-1 (APS-1), caused by AIRE deficiency3,4, and in patients diagnosed with coeliac disease5-7. However, the underlying mecha

A rare complication of the duodenal diverticulum.

UNLABELLED: Introduction: An 80-year-old woman presented to the emergency department with severe right-sided abdominal pain that had started after her last meal. Physical examination revealed fever (38.6°C) and rebound tenderness in the right upper quadrant of the abdomen. The laboratory studies showed a leucocyte count of 11.3×109/L (normal, 3.7-8.0×109/L) and a C-reactive protein level of 2.34 mg/dL (normal, <0.03 mg/dL). There were no other significant findings. A CT scan of the abdomen with contrast revealed retroperitoneal air around a duodenal diverticulum (figures 1 and 2).Figure 1Compu

Pulmonary surfactant.

The mammalian lung is stabilized by a specialized material, the pulmonary surfactant, which acts by reversibly reducing the surface tension at the air-liquid interface of the lung during breathing. Pulmonary surfactant contains approximately 90% lipid and 10% proteins. Dipalmitoyl phosphatidylcholine, the major lipid component, appears to be primarily responsible for the ability to reduce surface tension to near 0 dyn/cm (1 dyn = 10 microN). The other components of pulmonary surfactant promote the adsorption and spreading of this disaturated lecithin at the air-liquid interface. Surfactant act

Umbrella-like Helical Structure of α-Synuclein at the Air-Water Interface Observed with Experimental and Theoretical Sum Frequency Generation Spectroscopy.

The misfolding of α-synuclein (αS) into amyloid aggregates is catalyzed by hydrophobic surfaces and associated with severe brain disorders, such as Parkinson's disease. Despite the important role of interfaces, the three-dimensional structure of αS at the interfaces is still not clear. We report interface-specific sum frequency generation (SFG) experiments of monomeric αS binding to the air-water interface, a model system for the important hydrophobic surfaces. We combine the SFG spectra with calculations of theoretical spectra based on molecular dynamics simulations to show that αS, which is

Air pollution exposure increases ABCB1 and ASCT1 transporter levels in mouse cortex.

Membrane transporters are important for maintaining brain homeostasis by regulating the passage of solutes into, out of, and within the brain. Growing evidence suggests neurotoxic effects of air pollution exposure and its contribution to neurodegenerative disorders, including Alzheimer's disease (AD), yet limited knowledge is available on the exact cellular impacts of exposure. This study investigates how exposure to ubiquitous solid components of air pollution, ultrafine particles (UFPs), influence brain homeostasis by affecting protein levels of membrane transporters. Membrane transporters w

[Effect of the temperature-humidity regimen on broiler resistance and on the efficacy of coccidiostats in experimental coccidial infection].

Experiments are carried out to elucidate the effect of the temperature and Moisture Regime on the resistance of broiler chickens to an experimental infection with coccidia and how far the prophylactic, anti-coccidial activity of coccidiostatics may be raised or reduced on this basis. During the experiment, prior to and after the infection of the chicken, investigations have been carried out on the morphological and biochemical composition of the blood, determining the contents of hemoglobin, total protein, vitamin A and vitamin E. It is established that the hematological and biochemical indice

C/EBPalpha is required for lung maturation at birth.

Epithelial cells lining the peripheral lung synthesize pulmonary surfactant that reduces surface tension at the air-liquid interface. Lack of surfactant lipids and proteins in the lungs causes respiratory distress syndrome, a common cause of morbidity and mortality in preterm infants. We show that C/EBPalpha plays a crucial role in the maturation of the respiratory epithelium in late gestation, being required for the production of surfactant lipids and proteins necessary for lung function. Deletion of the Cebpa gene in respiratory epithelial cells in fetal mice caused respiratory failure at bi

Toxic in vitro effects of flavoured-air inhalers containing essential oils in differentiated human nasal epithelial cells at air-liquid interface.

Flavoured-air inhalers containing essential oils are marketed as safe, natural alternatives to smoking and vaping, yet their potential respiratory toxicity remains effectively unexamined. Manufacturers claim safety based on "100% natural" ingredients, despite evidence that essential oil inhalation can cause adverse respiratory effects. Our objective was to assess the in vitro toxicity of six commercially available flavoured-air products on human nasal airway epithelial cells. Differentiated air-liquid interface cultures of primary nasal airway epithelial cells from eight healthy adults were ex

Development of size-selective sampling of Bacillus anthracis surrogate spores from simulated building air intake mixtures for analysis via laser-induced breakdown spectroscopy.

Size-selective sampling of Bacillus anthracis surrogate spores from realistic, common aerosol mixtures was developed for analysis by laser-induced breakdown spectroscopy (LIBS). A two-stage impactor was found to be the preferential sampling technique for LIBS analysis because it was able to concentrate the spores in the mixtures while decreasing the collection of potentially interfering aerosols. Three common spore/aerosol scenarios were evaluated, diesel truck exhaust (to simulate a truck running outside of a building air intake), urban outdoor aerosol (to simulate common building air), and f

A comparison of air pouch, sponge and pleurisy models of acute carrageenan inflammation in the rat.

A comparative assessment of the time course of changes in three models of acute carrageenan-induced inflammation, the six-day air pouch, polyester sponge and pleurisy models, was obtained by measuring exudate volume, leucocyte numbers, PGE2 concentrations and lactate dehydrogenase activity at 2, 6 and 24 h. The greatest increases in exudate volume, leucocyte numbers and PGE2 concentration and the smallest rise in protein occurred in the air pouch model. Increases in lactate dehydrogenase were greatest in the sponge and least in the pleural exudate, indicating that the least cell damage occurre

Cold temperature enhances innate eosinophilic airway inflammation via transient receptor potential ankyrin1.

BACKGROUND AND OBJECTIVE: Asthma exacerbations due to cold air exposure are well recognized; however, the underlying mechanisms remain unclear. We investigate the role of the transient receptor potential ankyrin1 (TRPA1) channel in cold air-induced aggravation of innate airway inflammation using a murine model of papain stimulation combined with cold air exposure. METHODS: Wild-type (WT) and Trpa1 knockout (KO) mice were treated intranasally with papain under different temperature conditions. Bronchoalveolar lavage fluid (BALF) and lung tissues were analyzed. The effects of the TRPA1 antagoni

Autoimmune Retinopathy: An Immunologic Cellular-Driven Disorder.

Autoimmune retinopathy (AIR) was often mistaken for retinitis pigmentosa (RP), due to an overlap of clinical findings, but increasingly has been recognized as a unique entity in the last decade. AIR has distinctive features: sudden onset of photopsias and scotomata in patients with no family history of RP, followed by visual field and central vision loss. Initially, retina exams are normal with no sign of pigment deposits or retinal degeneration. A family history of autoimmune diseases (all types) occurs in 60% of patients. One hallmark of AIR has been the presence of anti-retinal autoimmune a

Air Pollution from Road Traffic and Systemic Inflammation in Adults: A Cross-Sectional Analysis in the European ESCAPE Project.

BACKGROUND: Exposure to particulate matter air pollution (PM) has been associated with cardiovascular diseases. OBJECTIVES: In this study we evaluated whether annual exposure to ambient air pollution is associated with systemic inflammation, which is hypothesized to be an intermediate step to cardiovascular disease. METHODS: Six cohorts of adults from Central and Northern Europe were used in this cross-sectional study as part of the larger ESCAPE project (European Study of Cohorts for Air Pollution Effects). Data on levels of blood markers for systemic inflammation-high-sensitivity C-reactiv

Storage conditions differentially alter the human tooth enamel proteome.

INTRODUCTION: Exfoliated or extracted human teeth are increasingly used as accessible records of health history, biomarkers for predicting health risks, or analyzed for organic components to uncover the mechanisms of atypical development or disease. During formation, dental enamel incorporates and preserves biological information through mineralization, thereby creating a fossil-like record within the tooth structure as it reaches a mineral content of over 95% by weight. The key to unlocking this record depends not only on using appropriate analytical methodologies, but also on how storage con

The expressions of protooncogenes and CYP1A in lungs of rats exposed to sulfur dioxide and benzo(a)pyrene.

Sulfur dioxide (SO2) is a ubiquitous air pollutant, present in low concentrations in the urban air, and in higher concentrations in the working environment. Benzo(a)pyrene (B(a)P), a polycyclic aromatic hydrocarbon, is a ubiquitous environmental contaminant with diverse toxicological effects. To investigate the interactions between SO2 and B(a)P, male Wistar rats were exposed to intratracheally instilled with benzo(a)pyrene (B(a)P; 3 mg) or SO2 (20 ppm) inhalation alone or together. The mRNA of CYP1A1 and 1A2, c-fos, and c-jun and protein levels of c-fos and c-jun were analyzed in lungs using

Effects on airways of short-term exposure to two kinds of wood smoke in a chamber study of healthy humans.

INTRODUCTION: Air pollution causes respiratory symptoms and pulmonary disease. Airway inflammation may be involved in the mechanism also for cardiovascular disease. Wood smoke is a significant contributor to air pollution, with complex and varying composition. We examined airway effects of two kinds of wood smoke in a chamber study. MATERIALS AND METHODS: Thirteen subjects were exposed to filtered air and to wood smoke from the start-up phase and the burn-out phase of the wood-burning cycle. Levels of PM(2.5) were 295 µg/m(3) and 146 µg/m(3), number concentrations 140 000/cm(3) and 100 000/cm

Elevated atmospheric CO2-induced reprogramming leads to decreased seed protein and nutritional quality in forest trees.

While the global increases in atmospheric CO2 levels have had a beneficial effect on plant growth, the negative impacts of this CO2 fertilization effect on seed quality are often overlooked. Using data from acorns produced by mature oak (Quercus robur) trees in the eighth year of eCO2, we present evidence of negative consequences for seed quality. The acorns produced by the near-200-yr-old oak trees under eCO2 at the free air carbon dioxide enrichment facility at the Birmingham Institute for Forest Research (BIFoR) had higher phytate levels but decreased protein content. Quantitative label-fre

Oral malodorous compound induces osteoclast differentiation without receptor activator of nuclear factor κB ligand.

BACKGROUND: Hydrogen sulfide (H(2)S), the main component of halitosis, is one of the etiologic factors for periodontitis. We recently reported that H(2)S may induce pathologic changes in rat alveolar bone. The objective of this study is to determine the effect of H(2)S on osteoclast differentiation. METHODS: Murine macrophage cells RAW264 were cultured in medium lacking nuclear factor κB ligand (receptor activator of nuclear factor κB ligand) in 5% CO(2) with air at 37°C for 24 hours; then 0.05, 0.5, or 5 ng/ml H(2)S was added to the CO(2)-air mix for 4 days.
