Pigeon paramyxovirus type 1 (PPMV-1) is considered an antigenic and variant of avian paramyxovirus type 1 (APMV-1) that has adapted to pigeons as hosts. However, how this host-specific adaption of PPMV-1 is related to its biological characteristics is unknown. In this study, seven unique amino acids in PPMV-1 that are not present in other APMV-1 strains (n = 39 versus n = 106) were identified. R36 of the M protein was found to be not only a unique amino acid but also a positive-selection site. To investigate the role of R36 in host adaptation, a recombinant PPMV-1 with R36Q mutation was constr

Bluetongue (BT) viruses (BTV) have been propagated in laboratory culture systems for more than 50 years, especially for the production of vaccines. An important outcome of the adaptation of these viruses to laboratory culture is attenuation of their virulence. As a consequence of this modification, it has been possible to produce vaccines that have reduced the clinical impact of BT in a number of countries. Unfortunately, the adaptation of these viruses has also introduced undesirable properties. In particular, modified live BT vaccine viruses have a high capacity to cross the ovine placenta a

MOTIVATION: Intratumoral heterogeneity drives cancer recurrence and metastasis, yet single-cell drug response prediction faces severe "cross-domain" challenges, such as applying in vitro models to in vivo tissues or inferring metastatic resistance from primary tumors. These scenarios trigger distribution shifts arising from heterogeneous sequencing platforms, distinct tissue microenvironments, and metastatic evolution-problems rarely addressed by existing methods.
RESULTS: We introduce CSGDA, a cell state-guided graph domain adaptation framework designed to predict drug responses across these

Domestic species such as cattle (Bos taurus taurus and B. t. indicus) represent attractive biological models to characterize the genetic basis of short-term evolutionary response to climate pressure induced by their post-domestication history. Here, using newly generated dense SNP genotyping data, we assessed the structuring of genetic diversity of 21 autochtonous cattle breeds from the whole Mediterranean basin and performed genome-wide association analyses with covariables discriminating the different Mediterranean climate subtypes. This provided insights into both the demographic and adapti

UNLABELLED: Nα- and Nε-acetylation represent a pivotal post-translational modification used by both eukaryotes and prokaryotes to modulate diverse biological processes. Acinetobacter baumannii has been described as an important nosocomial pathogen for the past 30 years, frequently involved in ventilator-associated pneumonia, bloodstream and urinary tract infections. Many aspects of the biology of A. baumannii remain elusive, in particular the extent and function of N-acetylation. We investigated here N-acetylation in A. baumannii strain ATCC 17978 by proteomic analysis, and we showed the usefu

Borrelia burgdorferi, the vector-borne bacterium that causes Lyme disease, was first identified in 1982. It is known that much of the pathology associated with Lyme borreliosis is due to the spirochete's ability to infect, colonize, disseminate, and survive within the vertebrate host. Early studies aimed at defining the biological contributions of individual genes during infection and transmission were hindered by the lack of adequate tools and techniques for molecular genetic analysis of the spirochete. The development of genetic manipulation techniques, paired with elucidation and annotation

Treatment of AAV follows the principle of a combined remission induction and maintenance strategy and is adapted in a stage and activity-adapted fashion. So far the combination therapy of glucocorticoids and conventional immunosuppressive drugs has mainly been used to control disease. This approach has led to a significant improvement in outcome in spite of persistently high early mortality rates of nearly 11% within the first year. Besides conventional treatment, biologics have emerged as a new treatment option. The paper summarizes the current evidence for the use of conventional therapy and

BACKGROUND: The soil-dwelling Rhabdocoela flatworm, Luticola nematophagus, recently identified as a specialized predator of plant-parasitic nematodes such as Meloidogyne incognita, demonstrates considerable potential as a biocontrol agent for agricultural nematode diseases. This study presents the first chromosome-level genome assembly of L. nematophagus, achieved by integrating PacBio HiFi long-read sequencing with Hi-C chromatin conformation capture. RESULTS: The assembled genome is approximately 899.92 Mb with a scaffold N50 of 231.83 Mb, comprising 50.08% repetitive sequences and 45,225 pr

How much arboreality characterized the Australopithecus locomotor repertoire and how this differed from early Homo are vigorously debated. Most previous studies rely on traits of uncertain functional significance or are constrained by sampling concerns. Bone shaft strength is plastic and reflects in vivo mechanical loading. Because interlimb bone strength proportions among living apes correlate with arboreality, they are ideal contributors to this debate. We present the most comprehensive analysis yet of relative limb shaft strengths in Australopithecus and early Homo by adding two individuals

The integument serves as the interface between an organism and its environment. It primarily comprises ectoderm-derived epithelium and mesenchyme derived from various embryonic sources. These integumentary organs serve as a barrier defining the physiological boundary between the internal and exterior environments and fulfill diverse functions. How does the integument generate such a large diversity? Here, we attempt to decipher the organizational principles. We focus on amniotes and use appendage follicles as the primary examples. The integument begins as a simple planar sheet of coupled epith

The mammalian circadian clock governs physiological, endocrine, and metabolic responses coordinated in a 24-h rhythmic pattern by the suprachiasmatic nucleus (SCN) of the anterior hypothalamus. The SCN also dictates circadian rhythms in peripheral tissues like the kidney. The kidney has several important physiological functions, including removing waste and filtering the blood and regulating fluid volume, blood osmolarity, blood pressure, and Ca2+ metabolism, all of which are under tight control of the molecular/circadian clock. Normal aging has a profound influence on renal function, central

Bioluminescent dinoflagellates grow at one third the rate of their competitors of equivalent size, such as diatoms [1]. Despite this disadvantage, dinoflagellates successfully persist within phytoplankton communities and even form large blooms during favourable conditions. One explanation for this paradox is that bioluminescence acts as a defence that reduces losses to zooplankton grazers, such as copepods [2,3]. Lindström et al.[4] found that the dinoflagellate Lingulodinium polyedra (F.Stein) J.D.Dodge increase their bioluminescence in response to copepodamides [5], polar lipids exuded by co

Adult onset Still's disease (AOSD) is a rare inflammatory disorder that remains poorly understood. Its pathophysiology is yet to be completely elucidated, but is known to consist mainly on a cytokine cascade, responsible for the systemic manifestations. AOSD diagnosis is usually difficult and delayed, with physicians having to rule out several other conditions, including cancer or infectious diseases. Prognosis is heterogeneous and difficult to establish, ranging from benign outcome to chronic destructive polyarthritis and/or life-threatening events. In addition, treatment remains to be codifi

This review outlines the contribution of evolutionary science to experimental and clinical psychotraumatology. From an evolutionary perspective, traumatic and psychosocial stressors are conceived of as events or circumstances that thwart the achievement of biological goals. The more important is the adaptive value of the goal, the more painful is the emotional impact of the life event that endangers goal achievement. Life history theory and sexual selection theory help to explain why goal priorities differ between the sexes and across age groups. Cultural values and social learning interact wi

Endemic species flocks inhabiting ancient lakes, oceanic islands and other long-lived isolated habitats are often interpreted as adaptive radiations. Yet molecular evidence for directional selection during species flocks radiation is scarce. Using partial transcriptomes of 64 species of Lake Baikal (Siberia, Russia) endemic amphipods and two nonendemic outgroups, we report a revised phylogeny of this species flock and analyse evidence for positive selection within the endemic lineages. We confirm two independent invasions of amphipods into Baikal and demonstrate that several morphological feat

Systemic acquired resistance (SAR) is a highly desirable form of resistance that protects against a broad-spectrum of related or unrelated pathogens. SAR involves the generation of multiple signals at the site of primary infection, which arms distal portions against subsequent secondary infections. The last decade has witnessed considerable progress, and a number of chemical signals contributing to SAR have been isolated and characterized. The diverse chemical nature of these chemicals had led to the growing belief that SAR might involve interplay of multiple diverse and independent signals. H

The cerebellum is well developed in cartilaginous fishes, with the same cell types (barring basket cells) and organizational features found in other vertebrate groups, including mammals. In particular, the lattice-like organization of cerebellar cortex (with a molecular layer of parallel fibers, interneurons, spiny Purkinje cell dendrites, and climbing fibers) is a defining characteristic. In addition to the cerebellum, cartilaginous fishes have cerebellum-like structures in the dorsolateral wall of the hindbrain. These structures are adjacent to and, in part, contiguous with the cerebellum. T

Migration between populations can be a major evolutionary force. However, some disagreement exists as to precisely how migration affects population adaptation. Some theories emphasize the inhibitory effects of gene flow between locally adapted populations, whereas others propose that migration can enhance adaptation. Migration has also been theorized to rescue sink populations from extinction. In our experiments, we serially passaged bacteriophage Φ6 host range mutants under sink conditions on a novel host while manipulating the source and number of migrants into these experimental populations

This review of human tooth wear describes the fundamental mechanisms underlying this process. Using the tribological approach they can be systematised and this in turn aids our understanding of them. In past populations wear was ubiquitous, intense, abrasive and physiological as it was related to their food and their technologies. In these populations, it affected the proximal surfaces, and the occlusal surfaces which modified the occlusal plane profoundly. To categorise this wear many different classification systems are used, from which we can determine diet, cultural changes and the age at

Mature tissues can often adapt to changes in their chemical, mechanical, or thermal environment. For example, in response to sustained increases or decreases in mechanical loads, some tissues grow and remodel so as to restore the stress or strain to its homeostatic state. Whereas most previous work addresses gross descriptors of tissue growth, this paper introduces a possible cell-mediated mechanism by which remodeling may occur in a soft connective tissue--that the kinetics of collagen deposition and degradation is similar regardless of the configuration of the body at which it occurs.
