Overcoming Challenges in Small-Ring Transfer: Direct Decarboxylative Hydroalkylation of Alkenes via Iron-Thiol Catalysis.

Cyclopropanes, especially those substituted with trifluoromethyl (CF3) groups, are valuable scaffolds in medicinal chemistry. Their enhanced … hemically activated iron complexes. The resulting radicals were then engaged in radical addition to alkenes, followed by a timely hydrogen atom transfer (HAT) process mediated by a thiol donor. This efficient iron-thiol cooperative catalysis enables, for the first time, facile hydroalkylation of alkenes with highly reactive CF3-containing cyclopropyl and …

Photocatalyzed Oxycarboranylation of Alkenes with 1-Iodo-o-carboranes through a Halogen-Atom Transfer Strategy.

Carboranes are of growing importance to pharmaceutical chemistry as the key units of boron neutron capture therapy (BNCT) reagents and the t … yl radicals by a polarity-matched halogen-atom transfer (XAT) process and their transformation with alkenes to afford alcohols. The practicality of this protocol was emphasized by the late-stage functionalization of drugs and natural products. Mechanistic studies elucidated that fast and efficient generation of carbon-centered carboranyl radicals through …

Distal Heteroaryl Ipso-Migration of Unactivated Alkenes via Cobalt/Photoredox-Catalyzed Hydrogen/Deuterium Atom Transfer.

A structural isomerization of alkenyl alcohols via 1,4- or 1,5-heteroaryl migration initiated by metal-hydride hydrogen atom transfer (MHAT) under cobalt/photoredox dual catalysis is described. The remote migration method provided various heteroaryl-containing ketones inaccessible through conventional MHAT 1,2-migration pathways. The cobalt/photoredox catalysis was further applied to the synthesis of deuterium-labeled heteroaryl-containing ketones.

Photocatalytic Radical Azido/Fluorosulfonylation of Unactivated Alkenes: Accessing Hubs Bridging CuAAC and SuFEx Click Chemistry.

Herein, we describe the successful development of an azido-fluorosulfonylation reaction of alkenes under photoredox catalysis, which could allow the installation of the two "clickable" groups, -N3 and -SO2F, on a C-C double bond, with TMSN3 as the azide source. The utilization of the difunctionalization products is also demonstrated in the construction of a library of 1,2,3-triazolesulfonyl fluoride compounds as well as drug molecule ligation by merging copper-catalyzed azide-alkyne cycloaddition (CuAAC) and sulfur(VI) fluoride exchange (SuFEx), the two generations of click reactions. Mechanis…

Dual NHC/Organophotoredox-Catalyzed Radical Phosphinoylacylation of Alkenes with Acyl Fluorides and H-Phosphine Oxides.

A dual NHC/photoredox-catalyzed radical phosphinoylacylation of alkenes has been developed with organic dye Eosin Y as organophotocatalyst under green light irradiation. In this reaction, readily available acyl fluorides and secondary phosphine oxides are used as radical precursors to simultaneously construct C-C and C-P bonds. A wide range of β-phosphinoyl ketones are smoothly accessed in moderate to good yields under these mild reaction conditions. The scale-up reaction and synthesis of new valuable organophosphorus compounds, such as phosphine-olefin ligands, highlight the great potential a…

Palladium-Catalyzed Cross-Coupling Reactions of Carboranes with Alkenes via Selective B-H Bond Activation.

A palladium-catalyzed Heck-type cross-coupling reaction of carboranes with alkenes in 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP) was realized. This reaction shows good B(9) selectivity for m-carboranes and is also suitable for o- and p-carborane. Meanwhile, a series of mono-, di-, and trisubstituted alkenes were compatible substrates to afford the alkenylated products in 16-89% yields. The 1,2-bis(carboranyl)ethylene was first synthesized by the reaction of vinyltrimethylsilane and m-carborane. Further transformations of the C═C bond in the product were examined by hydroboration oxidation, oxida…

Engaging Alkenes in Metallaphotoredox: A Triple Catalytic, Radical Sorting Approach to Olefin-Alcohol Cross-Coupling.

Metallaphotoredox cross-coupling is a well-established strategy for generating clinically privileged aliphatic scaffolds via single-electron reactivity. Correspondingly, expanding metallaphotoredox to encompass new C(sp3)-coupling partners could provide entry to a novel, medicinally relevant chemical space. In particular, alkenes are abundant, bench-stable, and capable of versatile C(sp3)-radical reactivity via metal-hydride hydrogen atom transfer (MHAT), although metallaphotoredox methodologies invoking this strategy remain underdeveloped. Importantly, merging MHAT activation with metallaphot…

Palladium-catalyzed regioselective synthesis of mono and bis(arylthiol) alkenes from propargyl carbonate and thiophenol.

A palladium catalyzed regioselective reaction of propargylic carbonate with thiophenols and benzene selenol is described. Atom-economic addition of thiols to propargylic carbonates provides an excellent opportunity for effective processes. The reaction proceeds via hydrothiolation to produce mono(arylthiol) alkenes and hydrothiolation followed by Tsuji-Trost type substitution to form bis(arylthiol) alkenes through single and double sequential attack by soft thio nucleophiles by control of the equivalence of thiophenols. This coupling reaction with good tolerance towards functional groups in bo…

EnT-Mediated Amino-Sulfonylation of Alkenes with Bifunctional Sulfonamides: Access to β-Amino Sulfone Derivatives.

β-Amino sulfones are commonly found structural motifs in biologically active compounds. Herein, we report a direct photocatalyzed amino-sulfonylation reaction of alkenes for the efficicient production of important compounds by simple hydrolysis without the need for additional oxidants and reductants. In this transformation, the sulfonamides worked as bifunctional reagents, simultaneously generating sulfonyl radicals and N-centered radicals which were added to alkene in a highly atom-economical fashion with high regioselectivity and diastereoselectivity. This approach showed high functional gro…

Asymmetric Photoaerobic Lactonization and Aza-Wacker Cyclization of Alkenes Enabled by Ternary Selenium-Sulfur Multicatalysis.

An adaptable, sulfur-accelerated photoaerobic selenium-π-acid ternary catalyst system for the enantioselective allylic redox functionalization of simple, nondirecting alkenes is reported. In contrast to related photoredox catalytic methods, which largely depend on olefinic substrates with heteroatomic directing groups to unfold high degrees of stereoinduction, the current protocol relies on chiral, spirocyclic selenium-π-acids that covalently bind to the alkene moiety. The performance of this ternary catalytic method is demonstrated in the asymmetric, photoaerobic lactonization and cycloaminat…

Visible-Light Photocatalytic Aerobic C═C Bond Cleavage of Alkenes to Carbonyls by CsPbBr3 Nanocrystals.

Herein, we reported a facile and readily accessible visible-light-driven photocatalytic protocol to induce oxidative cleavage of C═C bonds to corresponding carbonyls using CsPbBr3 nanocrystals as photocatalysts. This catalytic system was applicable to a wide range of terminal and internal alkenes. Detailed mechanism studies indicated that a single-electron transfer (SET) process was involved in this transformation, wherein the superoxide radical (O2•-) and photogenerated holes played crucial roles. Additionally, DFT calculations revealed that the reaction was initiated by the addition of O2•- …

Intermolecular Metal-Free Cyclopropanation and Aziridination of Alkenes with XH2 (X=N, C) by Thianthrenation.

Three-membered cyclic structures are widely existing in natural products and serve as enabling intermediates in organic synthesis. However, the efficient and straightforward access to such structures with diversity remains a formidable challenge. Herein, a general and practical protocol to aziridines and cyclopropanes synthesis using free XH2 (X=C or N) with alkenes by thianthrenation is presented. This metal-free protocol features the direct aziridination and cyclopropanation with unprotected XH2 . Free sulfonamides, amides, carbamates, amines, and methylene with acidic protons, are good prec…

Photocatalytic Addition Reactions of Ketene Silyl Acetals with Alkenes through Formation of α-Carbonyl Radicals.

Addition reactions of ketene silyl acetals with alkenes that do not have an electron-withdrawing group are generally difficult because the nucleophilicity of ketene silyl acetals and the electrophilicity of alkenes are not sufficient. Herein, we report photocatalytic addition reactions of ketene silyl acetals with alkenes that proceed through formation of α-carbonyl radicals. In the presence of an appropriate protic additive, the reactions proceeded smoothly under blue-light irradiation to afford the desired products in moderate to high yields.

Photocatalytic Oxidative Cleavage of Alkenes by Molecular Oxygen: Reaction Scope, Mechanistic Insights, and Flow Application.

The oxidative cleavage of C═C bonds with molecular oxygen was promoted effectively by a catalytic amount of a decatungstate photocatalyst using black light irradiation (365 nm). Not only aromatic ketones but also aliphatic ketones were obtained by the photocatalytic protocol. The continuous flow reaction of α-methylstyrene using a high-power ultraviolet light-emitting diode (365 nm) dramatically decreased the reaction time.

Photoredox-catalyzed hydrosulfonylation reaction of electron-deficient alkenes with substituted Hantzsch esters and sulfur dioxide.

A sulfonylation reaction of 4-substituted Hantzsch esters, DABCO·(SO2)2, and electron-deficient alkenes at room temperature in the presence of photoredox catalysis under visible light irradiation is described. Not only (E)-chalcones but also (vinylsulfonyl)benzene and 2-vinylpyridine are all suitable substrates in the transformation. This sulfonylation reaction under mild conditions shows a broad substrate scope with a good functional group compatibility. A plausible mechanism is proposed, which shows that the 4-substituted Hantzsch esters would be radical reservoirs under photoredox catalysis…
