Radiolabeling of amyloid-beta peptides.

Nowadays, a wide variety of protocols for labeling proteins is available. However, radiolabeling remains one of the most powerful, sensitive and accurate methods to trace and quantitate proteins. Additionally, radiolabeling techniques are steadily gaining importance for diagnosis and treatment in nuclear medicine. There is a considerable number of radioisotopes, but only some are commonly used for basic biomedical research. Among them, the iodine radioisotopes (gamma-emitters) have several advantages for the labeling of proteins. This chapter focuses on radioiodination protocols for amyloidoge…

Amyloid angiopathy-related cerebellar hemorrhage.

BACKGROUND: The posterior fossa localization of amyloid angiopathy-related hemorrhage is very unusual. Less than 10 cases have been previously reported. Surgical management of amyloid angiopathy-related hemorrhage is the subject of controversy. RESULTS: Typical aspects of amyloid angiopathy were found within the surgically removed biopsies of 71-year-old nonhypertensive, nondemented woman suffering cerebellar hemorrhage. CONCLUSIONS: Amyloid angiopathy-related hemorrhage could occur in the cerebellum, and diagnosis might be suspected when no past history of mental deterioration and/or arteri…

MRI evaluation of amyloid myopathy.

Amyloid myopathy is a rare complication of primary amyloidosis. The magnetic resonance imaging (MRI) features of two patients with amyloid myopathy were studied. Slight prolongation of muscle T1 and T2 relaxation times was evident but the striking abnormality was marked reticulation of the subcutaneous fat. The clinical findings of indurated extremities far exceeds the minimal signal intensity alteration seen in the muscles. The MR appearance of amyloid myopathy differs from that of other neuromuscular conditions in the minimal changes found in muscle, but the striking abnormality seen in subc…

Ubiquitin in cerebral amyloid angiopathy.

Immunohistological findings in cerebral blood vessels of 4 cases with cerebral amyloid angiopathy (CAA) were compared with those of 4 Alzheimer's (AD) cases. A panel of antibodies against 2 neurofilament subunits (BF10 and RT97), a microtubule-associated protein (TAU) and ubiquitin were used. CAA cases showed a strong immunoreactivity for ubiquitin in blood vessel wall. Senile plaques (SPs) in CAA cases showed strong ubiquitin positivity but the central amyloid core was negative. AD brains showed immunoreactivity with all antibodies in SPs and neurofibrillary tangles (NFTs); blood vessels were…

Aluminium, amyloid, and Alzheimer's disease.

Alzheimer's disease is a degenerative disease characterized by the presence of numerous senile plaques and neurofibrillary tangles, primarily in the cortical areas of the brain. There is no known cause of the disease, although exposure to aluminium has been implicated by epidemiological studies and the finding of aluminium in the cerebral plaques and tangles. There is a genetic predisposition for early-onset disease, and Down's syndrome patients are particularly vulnerable. Many of the plaques contain amyloid, and a gene for amyloid precursor protein has been identified on the long arm of chro…

Dialysis arthropathy: amyloid or iron?

The clinical, biochemical, radiological, and pathological features in five cases of dialysis arthropathy were analysed. All patients were re … ular cysts and erosions but most affected joints looked normal. All synovial tissue examined showed amyloid, which stained immunohistochemically for beta 2 microglobulin. Large amounts of iron were present in synovial tissue from affected joints. It is suggested that the deposits of iron, rather than amyloid, in synovial tissue may be the cause of the art …

Neuropathy, M components, and amyloid.

Of eighty-seven patients with nonhereditary systemic amyloidosis, ten had evidence of peripheral neuropathy. There was median-nerve neuropathy due to infiltration of the flexor retinaculum with amyloid in six patients. Two patients had diffuse sensorimotor neuropathy and two patients had sensory loss without evidence of motor impairment--three of these cases had amyloid deposition demonstrated by nerve biopsy. Neurological manifestations in all ten patients preceded other evidence of amyloidosis by 6 months-4 years (mean, 1 to 5 years). Neuropathy occurred in six of thirty-eight patients with …

Brain oligomeric β-amyloid but not total amyloid plaque burden correlates with neuronal loss and astrocyte inflammatory response in amyloid precursor protein/tau transgenic mice.

It has long been assumed that β-amyloid (Aβ) had to assemble into fibrillar amyloid plaques to exert its neurotoxic effects in Alzheimer disease. An alternative hypothesis is that soluble oligomers ofAβ play a much larger role in neuronal damage than the insoluble component. We have tested these competing hypotheses in vivo by studying the clinicopathologic correlates of oligomeric Aβ species and classic fibrillar amyloid plaques in the brains of double-transgenic APP-tau mice up to 17 months of age. Biochemical and immunohistochemical measures of brain oligomeric Aβ exponentially increased wi…

Amyloid PET Changes During Lecanemab Treatment in Patients With and Without Amyloid-related Imaging Abnormalities.

Lecanemab reduces cerebral amyloid-β burden in Alzheimer disease, but whether amyloid-related imaging abnormalities (ARIA) are associated with greater amyloid clearance remains unclear. We report 3 patients with comparable clinical backgrounds and similar baseline cognitive performance who underwent amyloid-β PET before and after lecanemab treatment. Quantitative analysis using Centiloid showed a greater reduction in amyloid burden in the patient who developed ARIA than in the 2 patients without ARIA. These observations suggest ARIA may be associated with more pronounced treatment-related amyl…

One Hundred Cerebral Microhemorrhages: Rethinking Cerebral Amyloid Angiopathy in the Era of Amyloid-Targeted Antibodies.

We report a case of probable cerebral amyloid angiopathy (CAA) presenting with almost 100 cerebral microhemorrhages in an apolipoprotein E 4 (APOE4) homozygous subject with mild cognitive impairment (MCI). CAA is highly prevalent in autopsy studies and may impact over 100 million individuals worldwide; however, it remains substantially underdiagnosed clinically. CAA raises the risk for intracerebral hemorrhages as well as amyloid-related imaging abnormalities (ARIA) with amyloid-targeted antibodies. CAA is especially a quandary in APOE4 homozygotes who are at greatest risk for both Alzheimer's…

Tau depletion diminishes vascular amyloid-related deficits in a mouse model of cerebral amyloid angiopathy.

INTRODUCTION: Tau is essential for amyloid beta (Aβ)-induced synaptic and cognitive deficits in Alzheimer's disease (AD), making its downregulation a therapeutic target. Cerebral amyloid angiopathy (CAA), a major vascular contributor to cognitive decline, affects over 90% of patients with AD. This study explores the impact of tau downregulation on CAA pathogenesis. METHODS: We crossed the Familial Danish Dementia mouse model (Tg-FDD), which develops vascular amyloid, with tau-null (mTau-/-) mice to generate a CAA model lacking endogenous tau (Tg-FDD/mTau-/-). Behavioral, electrophysiological,…

Salivary levels of amyloid beta reflect brain amyloid beta burden in cognitively-normal older adults.

BACKGROUND: Amyloid beta (Aβ) plaque burden, as measured by positron emission tomography (PET), is increasingly being used as a biomarker for Alzheimer's disease (AD) as well as a screening or monitoring tool for clinical trials with amyloid-lowering drugs. However, PET imaging is expensive, invasive and not widely available for all patients, necessitating alternative means to assess brain Aβ accumulation. OBJECTIVES: In this study, we measured levels of Aβ42, Aβ40 and Aβ38 in saliva samples from cognitively unimpaired older adults (n=93; 61.7 % female; mean age = 70.1 ± 6.6 years) using the …

Differentiating Cerebral Amyloid Angiopathy From Alzheimer's Disease Using Dual Amyloid and Tau Positron Emission Tomography.

BACKGROUND AND PURPOSE: Although amyloid positron emission tomography (PET) might provide a molecular diagnosis for cerebral amyloid angiopathy (CAA), it does not have sufficient specificity for this condition relative to incipient Alzheimer's disease (AD). To identify a regional amyloid uptake pattern specific to CAA, we attempted to reduce this overlap by selecting "pure CAA" (i.e., fulfilling the criteria for probable CAA but without tau PET AD signature) and "pure AD" (i.e., positive amyloid PET and presence of tau PET AD signature, but without lobar hemorrhagic lesions). We hypothesized t…

Progressive Spread of Beta-amyloid Pathology in an Olfactory-driven Amyloid Precursor Protein Mouse Model.

Years before Alzheimer's disease (AD) is diagnosed, patients experience an impaired sense of smell, and β-amyloid plaques accumulate within the olfactory mucosa and olfactory bulb (OB). The olfactory vector hypothesis proposes that external agents cause β-amyloid to aggregate and spread from the OB to connected downstream brain regions. To reproduce the slow accumulation of β-amyloid that occurs in human AD, we investigated the progressive accumulation of β-amyloid across the brain using a conditional mouse model that overexpresses a humanized mutant form of the amyloid precursor protein (hAPP…

Cerebral amyloid angiopathy related inflammation: An under recognized but treatable complication of cerebral amyloid angiopathy.

Cerebral amyloid angiopathy-related inflammation (CAA-ri) is a subset of cerebral amyloid angiopathy (CAA) causing a reversible encephalopathy characterized by seizures and focal neurological deficit. Previously, biopsy was required to make this diagnosis, distinct radiological features have allowed development for clinicoradiological criteria to assist in diagnosis. CAA-ri is an important condition to recognize as patients respond to high dose corticosteroids with significant resolution of symptoms. A 79-year-old woman presents with new onset seizures and delirium with prior history of mild c…

Phloroglucinol, a clinical-used antispasmodic, inhibits amyloid aggregation and degrades the pre-formed amyloid proteins.

Amyloid proteins, such as β-amyloid (Aβ) and α-synuclein (α-syn), could form neurotoxic aggregates during the progression of neurodegenerative disorders. Phloroglucinol, a clinical-used drug for treating spasmodic pain, was predicted to cross the blood brain-barrier and possesses neuroprotective potential. In this study, we have found, for the first time, that phloroglucinol inhibited the formation of amyloid aggregates, and degraded pre-formed amyloid aggregates with the similar efficacy as curcumin, a widely known amyloid aggregation inhibitor. Moreover, phloroglucinol decreased the seeding …

Olfactory Bulb Amyloid-β Correlates With Brain Thal Amyloid Phase and Severity of Cognitive Impairment.

The Alzheimer disease (AD) neuropathological hallmarks amyloid β (Aβ) and tau neurofibrillary (NF) pathology have been reported in the olfactory bulb (OB) in aging and in different neurodegenerative diseases, which coincides with frequently reported olfactory dysfunction in these conditions. To better understand when the OB is affected in relation to the hierarchical progression of Aβ throughout the brain and whether OB pathology might be an indicator of AD severity, we assessed the presence of OB Aβ and tau NF pathology in an autopsy cohort of 158 non demented control and 173 AD dementia case…

Combinatorial model of amyloid β and tau reveals synergy between amyloid deposits and tangle formation.

AIMS: To illuminate the pathological synergy between Aβ and tau leading to emergence of neurofibrillary tangles (NFT) in Alzheimer's disease … osition, neuroinflammation and synaptic abnormalities, suggesting that early tau tangles affect the amyloid cascade. In the post-deposit cohort, contemporaneous expression of S320F tau did not exacerbate amyloid pathology, showing a dichotomy in Aβ-tau synergy based on the nature of Aβ. P301L tau produced NFT-type inclusions in the post-deposit cohort, bu …
