BACKGROUND CONTEXT: Targeting chemokines or chemokine receptors is a promising treatment strategy for diseases with chronic inflammation such as rheumatoid arthritis and discogenic pain. Identifying specific molecules and determining their effectiveness in animal models are the first steps in developing these treatments. Macrophage markers have been detected in the intervertebral disc tissues of patients with disc degenerative disease and discogenic pain and in different animal models. Macrophage recruitment into the disc may play a role in initiation of inflammation and if unresolved may lead

Single allelic mutations in the FOXG1 gene lead to FOXG1 syndrome (FS). To understand the pathophysiology of FS, which vary depending on FOXG1 mutation types, patient-specific animal models are critical. Here, we report a patient-specific Q84Pfs heterozygous (Q84Pfs-Het) mouse model, which recapitulates various FS phenotypes across cellular, brain structural, and behavioral levels. Q84Pfs-Het cortex shows dysregulations of genes controlling cell proliferation, neuronal projection and migration, synaptic assembly, and synaptic vesicle transport. The Q84Pfs allele produces the N-terminal fragmen

Collective cell migration has a key role in tissue morphogenesis, wound healing, tissue regeneration, and cancer invasion. In recent years, different animal models have been established to analyze how chemical and mechanical stimuli shape the behavior of single cells into tissues and organs. At present, there are still only a few model systems that allow to genetically dissect underlying molecular mechanisms driving cell motility during tissue morphogenesis at high resolution in real time. Here, we provide a detailed protocol and toolbox for ex vivo culturing of Drosophila testes for 4D live i

During the gastrulation stage in animal embryogenesis, the cells leading the axial mesoderm migrate toward the anterior side of the embryo, vigorously extending cell protrusions such as lamellipodia. It is thought that the leading cells sense gradients of chemoattractants emanating from the ectodermal cells and translate them to initiate and maintain the cell movements necessary for gastrulation. However, it is unclear how the extracellular information is converted to the intracellular chemical reactions that lead to motion. Here we demonstrated that intracellular Ca2+ levels in the protrusion

Tumor metastasis is a complex process in which cells detach from the primary tumor and colonize a distant organ. Metastasis is also the main process responsible for cancer-related death. Despite the enormous efforts made to unravel the metastatic process, there is no effective therapy, and patients with metastatic tumors have poor prognosis. In this regard, there is an urgent need for new therapeutic tools for the treatment of this disease. Small molecules with the capacity to reduce cell migration could be used to treat metastasis. Migrastatin-core analogs are naturally inspired macrocycles t

Proliferative and migratory abilities of fibroblasts are essential for wound healing at the skin surface. Cytoplasmic linker-associated protein-2 (CLASP2) was originally found to interact with cytoplasmic linker protein (CLIP)-170. CLASP2 plays an important role in microtubule stabilization and the microtubule-stabilizing activity of CLASP2 depends on its interactions with end binding (EB)-1 and CLIP-170. Although the microtubule-stabilizing role of CLASP2 is well established, the effects of CLASP2 on the migration and proliferation of fibroblasts remain unclear in the context of wound healing

BACKGROUND: Excessive proliferation and migration of pulmonary artery smooth muscle cells (PASMCs) play an important role in the occurrence and development of pulmonary arterial hypertension (PAH). miR-143/145 was reported to be up-regulated in the animal models and PAH patients, and deletion of miR-143/145 cluster prevented the development of hypoxia-induced pulmonary hypertension, but its underlying mechanism has not been elucidated.
METHODS: qRT-PCR and Western blot were performed to detect the expressions of miR-143/145 and ATP-binding cassette transporter A1 (ABCA1) in PAH patients and P

The protozoan parasite Toxoplasma gondii (T. gondii) causes one of the most common human zoonotic diseases and infects approximately one-third of the global population. T. gondii infects nearly every cell type and causes severe symptoms in susceptible populations. In previous laboratory animal studies, T. gondii movement and transmission were not analyzed in real time. In a three-dimensional (3D) microfluidic assay, we successfully supported the complex lytic cycle of T. gondii in situ by generating a stable microvasculature. The physiology of the T. gondii-infected microvasculature was monito

During the third millennium BC, Mesopotamia (the land between the Tigris and Euphrates Rivers, in modern Iraq-Syria), was dominated by the world's earliest cities and states, which were ruled by powerful elites. Ur, in present-day southern Iraq, was one of the largest and most important of these cities, and irrigation-based agriculture and large herds of domesticated animals were the twin mainstays of the economy and diet. Texts suggest that the societies of the Mesopotamian city-states were extremely hierarchical and underpinned by institutionalised and heavily-managed farming systems. Prevai

Migratory birds are a source of natural beauty and help in maintaining ecosystems, pest control and pollination. However, as with other pathogens, gastrointestinal (GI) parasites residing within migratory birds consequently contaminate the environment through their droppings. The study was aimed at identifying and estimating the prevalence and spatial distribution of GI parasites in migratory birds. To do this, 265 fecal samples were collected, (Bangladesh Livestock Research Institute (BLRI) Lake, Dhaka: 203), (Tanguar Haor, Sunamganj: 62), from October 2022 to March 2023. Samples were process

It has been become increasingly clear that swimming performance is crucial for the migratory success of wild Atlantic salmon (Salmo salar) smolts, yet the associated traits remain largely unexplored. In this study, smolts were caught during their downstream migration to the sea and were categorized as strong or poor swimmers based on their time to fatigue in a swimming test. Behavioral profiles were characterized through a confinement and novelty test, and relationships to cardiac morphology and glucocorticoid profiles were investigated. Strong swimmers increased activity over time in the conf

Radar technology has become a powerful tool for studying animal aeroecology in the lower atmosphere, particularly bird migration across large spatio-temporal scales. Quantifying bird density from radar data requires radar cross-section (RCS) estimates. Although RCS data exist for small bird species, large birds, especially soaring and flocking species, remain poorly characterized, partly due to technical challenges in measuring and modelling their complex morphology. This study introduces a practical and accessible modelling framework for estimating species-specific RCS in large birds, using t

Recent evidence suggests that bats can detect the geomagnetic field, but the way in which this is used by them for navigation to a home roost remains unresolved. The geomagnetic field may be used by animals both to indicate direction and to locate position. In birds, directional information appears to be derived from an interaction of the magnetic field with either the sun or the stars, with some evidence suggesting that sunset/sunrise provides the primary directional reference by which a magnetic compass is calibrated daily. We demonstrate that homing greater mouse-eared bats (Myotis myotis)

BACKGROUND: Tyrosine kinase inhibitors (TKIs) are widely used to treat non-small cell lung cancer (NSCLC) harboring epidermal growth factor receptor (EGFR) mutations. However, long-term use frequently leads to drug resistance. These resistant cells often exhibit distinct phenotypes from their parental counterparts and may influence cancer progression, including proliferation, migration, and invasion.
METHODS AND RESULTS: We investigated the effects of afatinib resistance, a second-generation EGFR-targeting TKI, on cell growth, mesenchymal marker expression, and TGF-β1-induced migration and in

BACKGROUND: Murine bone marrow-derived myofibroblasts (BMFs) have previously been shown to promote gastric cancer growth. However, whether BMFs promote gastric cancer cell metastasis remains largely unknown.
METHODS: Wound healing assay, Transwell invasion and migration assay and 3D organotypic co-culture systems were conducted to study the effects of BMFs on invasion and migration of gastric cancer cells and the invasion and migration ability of gastric cancer stem cell-like cells (CSC-LCs) induced by BMFs. We employed two animal model to study the role of BMFs on the in vivo metastasis of g

Hunting governance in Europe blends territorial and licensing traditions, producing hybrid regulatory regimes. We compared Poland's centralised hunting act with Germany's federal state-level regulations to examine how governance architecture was associated with season design, legal game status, and reported harvest for shared migratory game birds. We combined comparative legal analysis with national bag statistics from 12 hunting seasons (2012/2013-2023/2024) and available abundance estimates for wood pigeon, Eurasian woodcock, and three Anser geese taxa. Germany had higher indicative harvest-

Seasonal migratory animals can evolve residency through adaptation. The threshold model of quantitative genetics predicts that residency evolves from migrants when genetic variation for lower liability, or a latent state underlying migratory propensity, is positively selected. However, it is unknown what cellular and molecular regulations underlie this process. The hypothalamus, which regulates physiology and behaviour of vertebrates by integrating internal and environmental conditions, is a strong candidate structure where migratory propensity may be regulated. To understand the hypothalamic

This study presents an Agent-Based Model (ABM) simulation to assess the impact of varying migration routes on sea lice (Caligus clemensi) infestation levels in juvenile wild sockeye salmon (Oncorhynchus nerka) in the Discovery Islands, British Columbia, Canada. This research highlights the importance of migratory routes in determining the extent of exposure to sea lice originating from nearby salmon farms. Three northward out-migration routes were modelled, each exposing the fish to different levels of infestation pressure based on proximity to salmon farms. The ABM incorporates spatially expl

Food deprivation is a naturally occurring stressor that is thought to influence the ultimate life-history strategy of individuals. Little is known about how food deprivation interacts with other stressors to influence migration success. European populations of brown trout (Salmo trutta) exhibit partial migration, whereby a portion of the population smoltifies and migrates to the ocean, and the rest remain in their natal stream. This distinct, natural dichotomy of life-history strategies provides an excellent opportunity to explore the roles of energetic state (as affected by food deprivation)
