Antimicrobial Peptides Against Antimicrobial-Resistant Bacteria: Focus on Machine Learning.

Antimicrobial resistance (AMR) represents a pressing public health threat of the 21st century, with an estimated ten million deaths annually from drug-resistant infections by 2050. Diminishing pipelines and the accelerating emergence of multidrug-resistant pathogens make the development of novel antibacterials more urgent than ever. Antimicrobial peptides (AMPs) are among the most promising alternatives to conventional drugs, exhibiting broad antimicrobial spectra, rapid kinetics, and mechanisms that are difficult for bacteria to circumvent. However, the problem of discovering and engineering

Application of curcuminoids in inflammatory, neurodegenerative and aging conditions - Pharmacological potential and bioengineering approaches to improve efficiency.

Curcumin, a natural compound found in turmeric, has shown promise in treating brain-related diseases and conditions associated with aging. Curcumin has shown multiple anti-inflammatory and brain-protective effects, but its clinical use is limited by challenges like poor absorption, specificity and delivery to the right tissues. A range of contemporary approaches at the intersection with bioengineering and systems biology are being explored to address these challenges. Data from preclinical and human studies highlight various neuroprotective actions of curcumin, including the inhibition of neur

Dextran is an effective alternative to egg yolk and glycerol in feline epididymal sperm cryopreservation.

The cryopreservation of epididymal spermatozoa is essential for conserving genetic material from endangered species of high-genetic value males suddenly die. The most effective extenders for cryopreservation contain glycerol and egg yolk as cryoprotective components, but variability in composition, contamination risks, and potential toxicity, make the need to find more effective and safe alternatives crucial. This study evaluated the effectiveness of dextran as a substitute for egg yolk and glycerol in feline epididymal sperm cryopreservation. Spermatozoa collected from 24 domestic tomcats aft

Genotyping and molecular characterization of antimicrobial resistance in thermophilic Campylobacter isolated from poultry breeders and their progeny in Eastern Spain.

Thermophilic Campylobacter spp. are recognized as a major cause of acute bacterial diarrhea in humans, with broiler meat being the most common source of human infection. Antibiotic therapy is usually necessary for severe or prolonged infections, especially in immunocompromised populations such as young or elderly individuals. However, different studies have demonstrated a close association between antibiotic use in animal production and antimicrobial resistance (AMR) in humans. In this sense, there is social pressure to reduce antibiotic administration and find adequate alternatives to control

Towards a bioengineered kidney: recellularization strategies for decellularized native kidney scaffolds.

Patients with end-stage renal disease often undergo dialysis as a partial substitute for kidney function while waiting for their only treatment option: a kidney transplant. Several research directions emerged for alternatives in support of the ever-growing numbers of patients. Recent years brought big steps forward in the field, with researchers questioning and improving the current dialysis devices as well as moving towards the design of a bioengineered kidney. Whole-organ engineering is also being explored as a possibility, making use of animal or human kidney scaffolds for engineering a tra

Flavonoid-Rich Cyperus esculentus Extracts Disrupt Cellular and Metabolic Functions in Staphylococcus aureus.

The escalating threat of antibiotic resistance, particularly from Staphylococcus aureus (S. aureus), has become a critical challenge in both public health and animal husbandry. The extensive use of conventional antibiotics in livestock production accelerates the emergence of resistant strains, heightening risks to food safety and human health. Although plant-derived bioactive compounds are increasingly recognized as promising alternatives to synthetic antimicrobials, the mechanisms underlying their efficacy-and the potential for synergistic action among different plant parts-remain poorly unde

Use of discarded corneo-scleral rims to create cornea-like tissue.

BACKGROUND: Corneal disease is a major cause of blindness. Transplantation of cadaver-derived corneas (keratoplasty) is still the current therapy of choice; however, the global shortage of donor corneas continues to drive a search for alternatives. To this end, biosynthetic corneal substitutes have recently begun to gain importance. Here, we present a novel method for the generation of a cornea-like tissue (CLT), using corneo-scleral rims discarded after keratoplasty. METHODS AND RESULTS: Type I collagen was polymerized within the corneo-scleral rim, which functioned as a 'host' mould, direct

Bacterial quorum sensing inhibitors: attractive alternatives for control of infectious pathogens showing multiple drug resistance.

Quorum sensing (QS) is a bacterial communication process that depends on the bacterial population density. It involves small diffusible signaling molecules which activate the expression of myriad genes that control diverse array of functions like bioluminescence, virulence, biofilm formation, sporulation, to name a few. Since QS is responsible for virulence in the clinically relevant bacteria, inhibition of QS appears to be a promising strategy to control these pathogenic bacteria. With indiscriminate use of antibiotics, there has been an alarming increase in the number of antibiotic resistant

Special article: general anesthetic gases and the global environment.

General anesthetics are administered to approximately 50 million patients each year in the United States. Anesthetic vapors and gases are also widely used in dentists' offices, veterinary clinics, and laboratories for animal research. All the volatile anesthetics that are currently used are halogenated compounds destructive to the ozone layer. These halogenated anesthetics could have potential significant impact on global warming. The widely used anesthetic gas nitrous oxide is a known greenhouse gas as well as an important ozone-depleting gas. These anesthetic gases and vapors are primarily e

Urethral replacement with vascularized tunica vaginalis: defining the optimal form of use.

PURPOSE: Residual urethral defects after failed hypospadias surgery present difficulties in reconstruction. In these situations, repair using vascularized tissue flaps remains ideal, yet challenging. In order to offer more surgical alternatives in these cases, the use of vascularized tunica vaginalis flaps as urethral replacements has recently been established in an animal model. We undertook the following studies to further define the role of tunica vaginalis in urethral reconstruction. MATERIALS AND METHODS: A vascularized flap of tunica vaginalis was used to replace an experimentally creat

Enzymatic approach to biodiesel production.

The need for alternative energy sources that combine environmental friendliness with biodegradability, low toxicity, renewability, and less dependence on petroleum products has never been greater. One such energy source is referred to as biodiesel. This can be produced from vegetable oils, animal fats, microalgal oils, waste products of vegetable oil refinery or animal rendering, and used frying oils. Chemically, they are known as monoalkyl esters of fatty acids. The conventional method for producing biodiesel involves acid and base catalysts to form fatty acid alkyl esters. Downstream process

Microfluidics-based skin irritation test using in vitro 3D angiogenesis platform.

A global ban on animal experiments has been proposed. Hence, it is imperative to develop alternative models. Artificial skin models should reflect the responses of subcutaneous blood vessels and the immune system to elucidate disease and identify cosmetics' base materials. Notably, in vivo skin-irritation cascades involve disruption of the epidermal barrier and the release of proinflammatory mediators in response to chemical stimuli. Such proinflammatory factors promote angiogenesis and blood vessel permeability, as observed in irritant contact dermatitis. As an alternative to animal models, w

Synthesis of 18F-Labeled FC-119S and Its Tosyl Precursor.

Animal models of Alzheimer's disease (AD) are essential for developing therapeutics and evaluating the efficacy of new drug candidates. Positron emission tomography (PET) is a useful method to monitor a major hallmark of the onset of AD, namely, the deposition of amyloid β peptide (Aβ) in the brain. [18F]FC-119S (1), a 2-pyridylbenzothiazole analog, has been applied as a radiotracer for PET visualization of Aβ plaques in an AD model, the 5xFAD mouse. Here, we present an alternative method for the automated synthesis of 18F-labeled 1 as a radiotracer for our animal PET studies. The first attemp

Small ruminant production system characteristics and their influence on use, alternatives of, and resistance to antimicrobials in Nigeria.

BACKGROUND: As the threat of antimicrobial resistance (AMR) escalates globally, the influence of indiscriminate antimicrobial use (AMU) in livestock cannot be overlooked. Antimicrobial use practices are continually explored in larger food-producing animals; however, small ruminants (sheep and goats) receive comparatively less research attention. Our study addresses this gap by investigating small ruminant production practices in Nigeria and exploring how they affect the use of antimicrobials and alternatives. METHODS: We adopted a mixed-methods study design. A semi-structured questionnaire wa

Opuntia spp. as Alternative Fodder for Sustainable Livestock Production.

During the past decades, livestock production has increased significantly, which has led to the degradation of rangelands due to overgrazing. The lack of water in several arid areas has led to a decline in crop and animal husbandry. As a consequence, the demand for drought-resistant crops has increased significantly so as to keep crop and animal husbandry systems viable and sustainable. Cactaceae have adaptive characteristics that ensure their development progress under drought conditions. The present review provides information on the nutritive value of Opuntia in animal fodder production, it

Baby Fae: a beastly business.

The Baby Fae experiment has highlighted the growing trend in medicine of using animal parts in the treatment of humans. This paper raises the question of the logical and moral justification for these current practices and their proposed expansion. We argue that the Cognitive Capacity Principle establishes morally justified necessary and sufficient conditions for the use of non-human animals in medical treatments and research. Some alternative sources for medical uses are explored as well as some possible programmes for their implementation.

[Infectivity of the entomopathogenic fungus Verticillium lecanii in mice and guinea pigs].

The resistance of plague insects to chemical insecticides as well as the importance of a healthy environment demands an alternative for agricultural plagues. Among others, biological control seems an alternate strategy with fungal entomopathogens playing a relevant role. The hyphomycete Verticillium lecanii is a natural bioregulator of aphids, scales and white-flies that attack different agricultural plantations. Its use in biological control programs must be assessed previously by safety procedures such as its innocuity in mammals and useful animals and plants. The aim of this study was point
