A family of pupal-specific cuticular protein genes in the mosquito Anopheles gambiae.

We have cloned and sequenced members of a cuticular protein multi-gene family from the mosquito Anopheles gambiae. Three genes (agcp2a-c), each approximately 1 kb in length, were found in a 17.4 kb genomic phage clone. Analysis of ten cDNAs revealed that at least four related genes are present. The open reading frame of the genes and cDNAs showed 95% sequence identity. Divergence was observed in the sequence of the 3' ends and the number of copies of two repeated coding sequences. In situ hybridizations with a probe prepared from one of these circular protein genes physically mapped to two loc…

Movement of Anopheles gambiae s.l. malaria vectors between villages in The Gambia.

Movement of mosquitoes belonging to the Anopheles gambiae complex (mixed wild populations of An.arabiensis, An.gambiae and An.melas) between three neighbouring rural villages in The Gambia was investigated by mark-release-recapture. A total of 12,872 mosquitoes were collected in bednets, marked with a magenta fluorescent powder and released over a 15-day period in one of the villages. A further 15,507 mosquitoes were collected in exit traps, marked with a yellow powder and released over the same period. Mosquitoes were captured daily in all three villages using pyrethrum spray catches, as well…

Pyrethroid-treated bednet effects on mosquitoes of the Anopheles gambiae complex in The Gambia.

The response of Anopheles gambiae complex mosquitoes to men sleeping under insecticide-impregnated or untreated bednets in six verandah trap huts was studied during the dry season in The Gambia. With this type of hut it was possible to collect live and dead indoor-resting mosquitoes and estimate the number of wild mosquitoes which entered, bloodfed on man, and exited each night. Bednets were treated with emulsions targetted to leave deposits of 25 mg/m2 lambda-cyhalothrin, or 5, 50 or 500 mg/m2 permethrin, diluted from emulsifiable concentrates (EC), or a blank formulation similar to the EC ex…

Genetic selection of a Plasmodium-refractory strain of the malaria vector Anopheles gambiae.

The anopheline mosquito is the target in most malaria control programs, primarily through the use of residual insecticides. A mosquito was studied that is refractory to most species of malaria through a genetically controlled mechanism. A strain of Anopheles gambiae, which was selected for complete refractoriness to the simian malaria parasite Plasmodium cynomolgi, also has varying degrees of refractoriness to most other malaria species examined, including the human parasites P. falciparum, P. ovale, and P. vivax for which this mosquito is the principal African vector. Furthermore, the refract…

Ovarian development and biting frequency in Anopheles culicifacies Giles in Sri Lanka.

In order to determine the duration of the gonotrophic cycle in Anopheles culicifacies Giles in Sri Lanka, ovarian development in freshly blood-fed wild-caught females was studied in the laboratory and the findings were compared with mark-and-release recoveries in the field. In specimens of unknown parity at first capture probable feeding intervals of 1, 2 and 3 days (mean: 1.7 - 2.1 days) were found. The release of freshly blood-fed nullipars bred from larvae produced two recaptures from which it could be concluded that the time required to become parous need not be more than three days. These…

[Insecticide tests for resistance on adults of Anopheles sinensis and Culex tritaeniorhynchus in Korea].

During the summer season(July-August) in 1967, the insecticide tests for resistance on adult A. sinensis and C. tritaeniorhynchus were carried out in Gobu-Myun Jeongup-Gun, Chollapuk-Do, where many Japanese encephalitis cases occur every year. 1)The mortality by 4.0% DDT in A. sinensis was 88.9% and by 4.0% dieldrin was 84.4%. Therefore the LC-50 of DDT for A. sinensis was 1.75%, while LC-50 of dieldrin was 1.1%. 2)The mortality by 4.0% DDT in C. tritaeniorhynchus was 36.7%, and by 4.0% dieldrin was 6.0% 3)The result indicates that A. sinensis in Korea is susceptible to DDT and resistant to di

Insecticide resistance status predetermines behavioural responses and oviposition fitness in Anopheles coluzzii.

Behavioural resistance is one of the major public health challenges facing planned vector control strategies involving insecticides. Understanding the behaviour of mosquitoes holistically when exposed to insecticides could be key to adding up to the fight against resistance. This study evaluated the behavioural responses and oviposition fitness of male and female Anopheles coluzzii exposed to selected pyrethroids, including lambda-cyhalothrin (0.05%), permethrin (0.75%), and DDT (4%). Male and female mosquitoes, aged three to five days, were exposed in eight replicates to selected insecticides…

Chromosomal inversions modulate 3D chromatin interactions and regulatory architecture in Anopheles gambiae.

Background Malaria parasite transmission by Anopheles mosquitoes remains a worldwide health burden, and understanding factors underlying natural vector susceptibility would inform rational design of vector control. The 2La chromosomal inversion segregates in the major vectors of human malaria, Anopheles coluzzii and Anopheles gambiae , and is associated with natural variation for malaria susceptibility, though underlying mechanisms are unknown. Here, we characterize alterations of chromatin conformation and gene expression induced by the two 2La inversion allelic forms, the ancestral 2La and t…

Biofilm-mediated antibiotic cross-protection: Acinetobacter baumannii-driven enhancement of Elizabethkingia anopheles.

UNLABELLED: Acinetobacter baumannii is a clinically prevalent multidrug-resistant bacterium, while Elizabethkingia, an emerging pathogen, has shown increasing infection and mortality rates in recent years. Although co-infections involving both pathogens have been reported, their interactions remain poorly understood. In this study, clinical data from 15 co-infected patients revealed that such infections were more frequent in elderly individuals with underlying diseases requiring intensive care and prolonged hospitalization. The two isolates exhibited similar antibiotic resistance profiles. In

Assessments of Anopheles arabiensis behaviour around bed nets using partial adhesive treatments.

Understanding the behaviour of malaria vectors could lead to improved designs of partially treated insecticidal nets and low-cost nets with less insecticide content. The behaviour of Anopheles arabiensis around cow baited bed nets with partial adhesive treatments were assessed in experimental huts. The study was conducted in Moshi Tanzania, using a Latin Square design with five arms: no bed-net, intact untreated bed-net, roof, sides and whole adhesive treated bed-nets. The data analyses were done using generalized linear mixed effects models with proportions of mosquitoes on the bed-net panels…

Virus-like particle-based vaccines targeting the Anopheles mosquito salivary protein, TRIO.

Malaria is a highly lethal infectious disease caused by Plasmodium parasites. These parasites are transmitted to vertebrate hosts when mosquitoes of the Anopheles genus probe for a blood meal. Sporozoites, the infectious stage of Plasmodium , transit to the liver within hours of injection into the dermis. Vaccine efforts are hindered by the complexity of the parasite's lifecycle and the speed at which the infection is established in the liver. In an effort to enhance immunity against Plasmodium , we produced a virus-like particle (VLP)-based vaccine displaying an epitope of TRIO, an Anopheles …

Molecular Tools for Early Detection of Invasive Malaria Vector Anopheles stephensi Mosquitoes.

Reports of the expansion of the Asia malaria vector Anopheles stephensi mosquito into new geographic areas are increasing, which poses a threat to the elimination of urban malaria. Efficient surveillance of this vector in affected areas and early detection in new geographic areas is key to containing and controlling this species. To overcome the practical difficulties associated with the morphological identification of immature stages and adults of An. stephensi mosquitoes, we developed a species-specific PCR and a real-time PCR targeting a unique segment of the second internal transcribed spa…

Swarming Behavior in Anopheles gambiae (sensu lato): Current Knowledge and Future Outlook.

Effective management of insect disease vectors requires a detailed understanding of their ecology and behavior. In Anopheles gambiae sensu lato (s.l.) (Diptera: Culicidae) mating occurs during swarming, but knowledge of their mating behavior under natural conditions is limited. Mosquitoes mate in flight over specific landmarks, known as swarm markers, at particular locations. Swarms consist of males; the females usually approach the swarm and depart following copulation. The number of mating pairs per swarm is closely associated with swarm size. The shape and height of swarm markers vary and m…

A mating-induced reproductive gene promotes Anopheles tolerance to Plasmodium falciparum infection.

Anopheles mosquitoes have transmitted Plasmodium parasites for millions of years, yet it remains unclear whether they suffer fitness costs to infection. Here we report that the fecundity of virgin and mated females of two important vectors-Anopheles gambiae and Anopheles stephensi-is not affected by infection with Plasmodium falciparum, demonstrating that these human malaria parasites do not inflict this reproductive cost on their natural mosquito hosts. Additionally, parasite development is not impacted by mating status. However, in field studies using different P. falciparum isolates in Anop…

Altered Gut Microbiota and Immunity Defines Plasmodium vivax Survival in Anopheles stephensi.

Blood-feeding enriched gut-microbiota boosts mosquitoes' anti-Plasmodium immunity. Here, we ask how Plasmodium vivax alters gut-microbiota, anti-Plasmodial immunity, and impacts tripartite Plasmodium-mosquito-microbiota interactions in the gut lumen. We used a metagenomics and RNAseq strategy to address these questions. In naïve mosquitoes, Elizabethkingia meningitis and Pseudomonas spp. are the dominant bacteria and blood-feeding leads to a heightened detection of Elizabethkingia, Pseudomonas and Serratia 16S rRNA. A parallel RNAseq analysis of blood-fed midguts also shows the presence of Eli

Versatile transgenic multistage effector-gene combinations for Plasmodium falciparum suppression in Anopheles.

The malaria parasite's complex journey through the Anopheles mosquito vector provides multiple opportunities for targeting Plasmodium with recombinant effectors at different developmental stages and different host tissues. We have designed and expressed transgenes that efficiently suppress Plasmodium infection by targeting the parasite with multiple independent endogenous and exogenous effectors at multiple infection stages to potentiate suppression and minimize the probability for development of resistance to develop. We have also addressed the fitness impact of transgene expression on the mo…

Pyrethroid exposure alters internal and cuticle surface bacterial communities in Anopheles albimanus.

A deeper understanding of the mechanisms underlying insecticide resistance is needed to mitigate its threat to malaria vector control. Following previously identified associations between mosquito microbiota and insecticide resistance, we demonstrate for the first time, the effects of pyrethroid exposure on the microbiota of F1 progeny of field-collected Anopheles albimanus. Larval and adult mosquitoes were exposed to the pyrethroids alphacypermethrin (only adults), permethrin, and deltamethrin. While there were no significant differences in bacterial composition between insecticide-resistant …

RDL mutations predict multiple insecticide resistance in Anopheles sinensis in Guangxi, China.

BACKGROUND: Anopheles sinensis is a major vector of malaria in China. The gamma-aminobutyric acid (GABA)-gated chloride channel, encoded by the RDL (Resistant to dieldrin) gene, is the important target for insecticides of widely varied structures. The use of various insecticides in agriculture and vector control has inevitably led to the development of insecticide resistance, which may reduce the control effectiveness. Therefore, it is important to investigate the presence and distribution frequency of the resistance related mutation(s) in An. sinensis RDL to predict resistance to both the wit…

Malaria vector Anopheles culicifacies sibling species differentiation using egg morphometry and morphology.

BACKGROUND: The malaria vector Anopheles culicifacies (sensu lato) is an important malaria vector in Southeast Asia which comprises of five sibling species namely A, B, C, D and E. However, only a few forms have been identified as malaria vectors in various endemic countries. Currently, for the first time egg morphometry and morphology has been used to differentiate the three known vector sibling species of Anopheles culicifacies collected from malaria endemic Madhya Pradesh state of central India. METHODS: The adult An. culicifacies (s.l.) was collected from five districts using standard mos…
