The technique of intravascular autoradiography of 3H-thymidine labeled nuclei of microvessels endothelia was developed. The density difference of labeled cells in microvascular bed of mesentery was estimated. The maximal density of labelled nuclei was found in pre- and postcapillary vessels, and minimal--in capillaries and large arteries and veins of mesenteric bed. The number of labeled nuclei per vessel was found to be relatively constant. The density of labeled cells in 4-months old rats is less than that in 3 weeks-old ones.

Autoradiography revealed very low density of muscarinic receptors in the rat pineal gland. Yet, the magnitude of phosphoinositide hydrolysis elicited with 0.1 mM carbachol was similar to that seen with 1 mM norepinephrine. The cholinergic and adrenergic phosphoinositide responses were fully additive. The cholinergic signal was insensitive to pertussis toxin both in vivo and in vitro and persisted in pineals cultured for 5 days. The data expand our previous finding on functional muscarinic acetylcholine receptors in the rat pineal gland.

Circulating vitamin A is transported by a carrier termed retinol binding protein (RBP). Autoradiography and immunofluorescence studies showed that, in normal human skin, RBP is confirmed exclusively to the dermis. Following oral vitamin A loading, a large increase in dermal RBP but only a very small rise of serum RBP was found. Serum RBP was slightly decreased in eczema patients and markedly decreased in patients with porphyria cutanea tarda. Values in other dermatoses, including psoriasis, were normal.

Using in vitro autoradiography, the distribution of 2-[125I]iodomelatonin (IMEL) binding sites in the brain of the pied flycatcher and the zebra finch was examined. The results show IMEL binding in retinofugal, tectofugal and thalamofugal brain areas of the visual system. Additionally, IMEL binding sites were detected in the ectostriatum, the thalamus, the mesencephalon and the limbic system. No IMEL binding could be demonstrated in the pineal gland, the hippocampus, the nucleus suprachiasmaticus, the visual wulst or the pituitary.

The distribution of sigma (sigma) receptors in the rat brain was studied with autoradiography using [3H]1,3-di-ortho-tolyl-guanidine ([3H]DTG) as a ligand. The highest concentration of sigma receptors was seen in the pineal gland, an area which has not been previously studied. This result is of interest as both sigma receptors and the pineal gland have recently been shown to play a role not only in the nervous system but also in the immune and endocrine systems.

The localization of gamma-aminobutyric acid (GABA) high-affinity binding sites was investigated in the exocrine and endocrine pancreas of neonatal rats by means of 3H-GABA autoradiography. GABA-binding was identified on Schwann cells and on the cells of the intralobular excretory ducts. In the endocrine part of the pancreas, no labelling was observed except in peripheral islet cells which, on the basis of their scarcity and distribution, could be somatostatin cells. Furthermore, peri-insular innervation showed considerable labelling.

The rate of glycoprotein synthesis by the columnar cells of the human endocervix was measured during one cycle in 12 women by autoradiography. An increase in the rate of synthesis was associated with an increase in the plasma oestradiol concentration. A change in progesterone concentration in the blood did not seem to influence cervical glycoprotein synthesis. The ratio between the different cell types in the epithelium, basal cells, ciliated cells and mucus cells did not show cyclic variation in the cycles studied.

In view of recent evidence for a role for the B subtype of cholecystokinin (CCKB) receptor in panic and anxiety, the distribution of CCKB receptors in the forebrain of a Rhesus macaca monkey was examined by receptor autoradiography employing [125I]D-Tyr25(Nleu28,31)-CCK25-33S. CCKB receptors were widely and topographically distributed in cortex. Other structures with notable labelling included the basal ganglia, presubiculum, amygdala, mamillary bodies, cerebellar cortex and pineal gland. The distribution of CCKB receptors further supports roles for this peptide in behavioural processes.

Binding of 2-[125I]melatonin to the anteroventral and anterodorsal nuclei of the rat limbic thalamus was analyzed with in vitro autoradiography. In both nuclei the binding was of high affinity (Kd values 24-41 pM) and was competed with by nanomolar concentrations of 6-hydroxymelatonin. Kd values were in the same range as those reported for other high-affinity binding sites in the brain. Bmax values were 0.68-2.99 fmol/mg protein. The results suggest that some effects of melatonin are mediated via the limbic thalamus.
