Depression in Dementia or Dementia in Depression? Systematic Review of Studies and Hypotheses.

The majority of research works to date suggest that Major Depressive Disorder (MDD) is a risk factor for dementia and may predispose to cognitive decline in both early and late onset variants. The presence of depression may not, however, reflect the cause, rather, an effect: it may be a response to cognitive impairment or alters the threshold at which cognitive impairment might manifest or be detected. An alternative hypothesis is that depression may be part of a prodrome to Alzheimer's Disease (AD), suggesting a neurobiological association rather than one of psychological response alone. Gene

Couple serostatus patterns in sub-Saharan Africa illuminate the relative roles of transmission rates and sexual network characteristics in HIV epidemiology.

HIV prevalence has surpassed 30% in some African countries while peaking at less than 1% in others. The extent to which this variation is driven by biological factors influencing the HIV transmission rate or by variation in sexual network characteristics remains widely debated. Here, we leverage couple serostatus patterns to address this question. HIV prevalence is strongly correlated with couple serostatus patterns across the continent; in particular, high prevalence countries tend to have a lower ratio of serodiscordancy to concordant positivity. To investigate the drivers of this continenta

Blood cell composition reveals distinct biological interpretation of DNA methylation age and age acceleration.

BACKGROUND: Epigenetic clocks are widely applied biomarkers of biological age, but their biological underpinnings remain unclear. We previously showed that epigenetic clocks are affected by naïve and memory T cell proportions, suggesting blood cell composition as a potential driver. However, analysis of cell composition is complicated by collinearity between cell fractions. METHODS: We first develop a principal component analysis (PCA) method that is robust to collinearity of blood cell counts and show that this approach provides biologically meaningful insights. Next, we quantify the contrib

Single-Cell RNA Sequencing Reveals that the Switching of the Transcriptional Profiles of Cysteine-Related Genes Alters the Virulence of Entamoeba histolytica.

Entamoeba histolytica is an intestinal protozoan that causes human amoebic colitis and extraintestinal abscesses. Virulence variation is observed in the pathogenicity of E. histolytica trophozoites, but the detailed mechanism remains unclear. Here, a single trophozoite was cultured alone, and the progeny of the trophozoites of each generation were subjected to single-cell RNA sequencing (scRNA-seq) to study the transcriptional profiles of trophozoites. The scRNA-seq analysis indicated the importance of sulfur metabolism and the proteasome pathway in pathogenicity, whereas the isobaric tags for

Biological predictors of chemotherapy-induced peripheral neuropathy (CIPN): MASCC neurological complications working group overview.

Chemotherapy-induced peripheral neuropathy (CIPN) is a common and debilitating condition associated with a number of chemotherapeutic agents. Drugs commonly implicated in the development of CIPN include platinum agents, taxanes, vinca alkaloids, bortezomib, and thalidomide analogues. As a drug response can vary between individuals, it is hypothesized that an individual's specific genetic variants could impact the regulation of genes involved in drug pharmacokinetics, ion channel functioning, neurotoxicity, and DNA repair, which in turn affect CIPN development and severity. Variations of other

The contribution of genetics to addiction therapy approaches.

Addictions, including alcohol dependence, which is the focus of this article, are complex genetic diseases. Recently, several individual genes that contribute to the risk for alcohol dependence have been identified, and more are expected to be in the near future. Among these are genes encoding alcohol and aldehyde dehydrogenases and GABA(A) receptor subunits. These reveal pathways of vulnerability and provide targets for rational drug design. It is likely that response to particular therapies is also a complex trait influenced by genetics, but studies to explore this are just beginning. We dis

The new biologic drugs: Which children with asthma should get what?

Novel biologics (targeted antibody therapies) have revolutionized the management of severe childhood asthma. However, it is important that the right biologic is selected for the right patient, and understanding the evidence base for each biologic is crucial. Currently, four biologics (all monoclonal antibodies) are licensed in the UK for the treatment of children with severe asthma - omalizumab (Xolair), mepolizumab (Nucala), and dupilumab (Dupixent) in children aged 6 years and over; and tezepelumab (Tezspire), only in children aged 12 years and over. Tezepelumab is the only licensed biologic

Normal Audiogram Does Not Confirm Intact Cochlear Amplification: Compressive Nonlinearity Reveals Incipient Cochlear Changes.

The cochlear amplifier, mediated by outer hair cell-driven nonlinear mechanics, enables the remarkable sensitivity and dynamic range of human hearing. Cochlear compression is a defining feature of this active process and is known to deteriorate with hearing loss. Among individuals with normal audiograms, cochlear compression is generally presumed to be invariant, an assumption that has not been systematically examined. If incipient cochlear changes precede measurable threshold shifts, variability in compressive nonlinearity may represent an early peripheral correlate of emerging dysfunction an

Biologic therapies in stress fractures: Current concepts.

Stress fractures, a common overuse injury in physically active individuals, present a significant challenge for athletes and military personnel. Patients who sustain stress fractures have demanding training regimes where periods of rest and immobilisation have unacceptable negative consequences on sports goals and finances. Aside from being an overuse injury, there are various contributing risk factors that put certain individuals at risk of a stress fracture. The main two being nutritional deficiencies and hormonal variations, which have significant effects on bone metabolism and turnover. Hi

Molecular Pathways within Autism Spectrum Disorder Endophenotypes.

Autism spectrum disorder (ASD) is a condition that includes a number of neurodevelopmental mental disorders. Recent genetic/genomic investigations have reported an increased prevalence of copy number variations (CNVs) in individuals with autism. Despite the extensive evidence of a genetic component, the genes involved are not known and the background is heterogeneous among subjects. As such, it is highly likely that multiple events (molecular cascades) are implicated in the development of autism. The aim of this work was to shed some light on the biological background behind this condition. We

Exploratory Immunophenotyping of Programmed cell death-1 (PD-1) Expression and Regulatory T-Cell (Treg) Profiles in Systemic Lupus Erythematosus and Healthy Controls: A Cohort Study with Illustrative Clinical Cases.

BACKGROUND: Immune checkpoint signaling and regulatory T cells (Tregs) play essential roles in immune homeostasis and immune dysregulation in autoimmune diseases. Programmed cell death-1 (PD-1) signaling and Treg dysfunction have been implicated in systemic lupus erythematosus (SLE); however, temporal variation in immune regulatory profiles across disease states remains incompletely characterized. This study aimed to characterize PD-1 expression and Treg-associated immune profiles in patients with SLE compared with healthy controls while descriptively exploring selected illustrative clinical i

The Impact of Age and Genetics on Naltrexone Biotransformation.

Naltrexone, an opioid antagonist primarily metabolized by aldo-keto reductase 1C4 (AKR1C4), treats pediatric conditions involving compulsiveness (e.g., autism spectrum, Prader-Willi, eating disorders, non-suicidal self-injury). Pharmacokinetic variability is apparent in adults, yet no data are available for children. This study aimed to examine the impact of age and genetic variation on naltrexone biotransformation. Human liver cytosol (HLC) samples (n = 158) isolated from children and adult organ donors were incubated with therapeutically relevant concentrations of naltrexone (0.1, 1 µM). Nal

Autoreceptor control of serotonin dynamics.

BACKGROUND: Serotonin is a neurotransmitter that has been linked to a wide variety of behaviors including feeding and body-weight regulation, social hierarchies, aggression and suicidality, obsessive compulsive disorder, alcoholism, anxiety, and affective disorders. Full understanding involves genomics, neurochemistry, electrophysiology, and behavior. The scientific issues are daunting but important for human health because of the use of selective serotonin reuptake inhibitors and other pharmacological agents to treat disorders. This paper presents a new deterministic model of serotonin metabo

Spatial transcriptomics on an expanded dataset at the brain-electrode interface: exploration of variability and identification of novel biomarkers.

The foreign body reaction to implanted electrodes in the brain has long been recognized as a major challenge impacting the performance and reliability of indwelling neurotechnologies. Spatially resolved transcriptomic approaches have enabled high-resolution mapping of cellular and molecular dynamics at the device-tissue interface, yielding novel insight into both acute and chronic tissue responses. Recent whole-transcriptome profiling methods generate exceptionally dense gene expression datasets from individual samples, offering unprecedented resolution and analytical power. Yet, limited studi

Mallard super-shedders of avian influenza exhibit distinct cloacal microbial abundance profiles.

In many infectious disease systems, a small fraction of hosts accounts for a disproportionate share of transmission, but the biological basis of this heterogeneity remains poorly resolved. In avian influenza, mallards show substantial variation in viral shedding, raising the possibility that only a subset of individuals contributes strongly to environmental contamination and onward spread. To examine whether host microbiome structure is associated with this variation, we experimentally infected wild, captive mallards with low-pathogenic avian influenza virus. We characterized cloacal microbiom

Methylome changes associated with functional movement/conversion disorder: Influence of biological sex and childhood abuse exposure.

Epigenetic changes, such as DNA methylation (DNAm), may represent an important mechanism implicated in the etiopathogenesis of functional movement/conversion disorder (FMD). Here, we aimed to identify methylomic variations in a case-control cohort of FMD and to uncover specific epigenetic signatures associated with female sex and childhood abuse, two key risk factors for FMD and other functional neurological disorders. Genome-wide DNAm analysis was performed from peripheral blood in 57 patients with FMD and 47 healthy controls with and without childhood abuse. Using principal component analysi

Biochemical and brain heterogeneity characterizes psychiatric and non-psychiatric illness.

Biological heterogeneity presents a formidable challenge in understanding psychiatric illness, yet it remains underexplored at scale or in a pan-disease setting. We characterized biochemical and brain heterogeneity in 445,374 individuals, considering 9 psychiatric disorders and 30 non-psychiatric medical conditions. Psychiatric and non-psychiatric illnesses were characterized by greater biological heterogeneity than predicted by healthy variation. Notably, heterogeneity in psychiatric disorders was comparable to non-psychiatric illnesses and often diverged from diagnostic classification. Circu

Whole-body patterns of the range of joint motion in young adults: masculine type and feminine type.

BACKGROUND: Understanding the whole-body patterns of joint flexibility and their related biological and physical factors contributes not only to clinical assessments but also to the fields of human factors and ergonomics. In this study, ranges of motion (ROMs) at limb and trunk joints of young adults were analysed to understand covariation patterns of different joint motions and to identify factors associated with the variation in ROM. METHODS: Seventy-eight healthy volunteers (42 males and 36 females) living on Okinawa Island, Japan, were recruited. Passive ROM was measured at multiple joint

The frequency of granulocytes with spontaneous somatic mutations: a wide distribution in a normal human population.

Germ-line mutation rate has been regarded classically as a fundamental biological parameter, as it affects the prevalence of genetic disorders and the rate of evolution. Somatic mutation rate is also an important biological parameter, as it may influence the development and/or the course of acquired diseases, particularly of cancer. Estimates of this parameter have been previously obtained in few instances from dermal fibroblasts and lymphoblastoid cells. However, the methodology required has been laborious and did not lend itself to the analysis of large numbers of samples. We have previously

Measurement of Glomerular Filtration Rate.

Glomerular filtration rate (GFR) can be estimated from the plasma concentration of endogenous substances such as creatinine, and it is simple and cost-effective to do so, but the error on these estimates on an individual patient basis is high. Thus, GFR measurement is indicated in any clinical situation in which greater accuracy is required especially if the result will impact treatment decisions. Outside of the United States the radiopharmaceutical that has been most commonly used for measurement of GFR is 51Cr-EDTA. However, due to a recent reduction in supply, there has been a shift to 99mT
