A direct enzyme immunoassay for 18-hydroxycortisol in urine: a new tool for screening primary aldosteronism.

A microplate enzyme immunoassay has been developed for the measurement of 18-hydroxycortisol in urine. An antiserum was produced by immunization of rabbits with a 3-O-(carboxymethyl)oximino-18-hydroxycortisol-bovine serum albumin conjugate. IgG was isolated from the antiserum and was biotinylated. Newly synthesized p-nitrophenyl ester of the oxime was used for the preparation of steroid-horseradish peroxidase conjugate. After an incubation of diluted urine samples (or standards) and the steroid-enzyme conjugate with the biotinylated antibody, the resulting antigen-antibody complex was separate

Detection of numerical chromosomal abnormalities in neoplastic hematopoietic cells by in situ hybridization with a chromosome-specific probe.

The feasibility of using molecular hybridization techniques for the detection of malignant clones that contain numerical chromosomal abnormalities was tested in clinical specimens from patients who had hematologic malignancies. A biotinylated DNA probe specific for chromosome 9 was used for in situ hybridization to interphase and terminally differentiated cells, and fluoresceinated avidin or avidin followed by biotinylated alkaline phosphatase was used for probe detection. In a blinded analysis of ten clinical samples from patients with hematologic malignancies and cytogenetically documented m

[Biocytin13]dynorphin A 1-13 amide: a potential probe for the kappa-opioid receptor.

A dynorphin A 1-13 amide (DYN) derivative biotinylated in position lysine 13 (B-DYN) has been prepared by automated solid-phase peptide synthesis. The derivative retained its ability to bind to avidin, and B-DYN-avidin complex showed a dissociated half-life of 10 hr at 37 degrees C. Opioid receptor binding was measured in membrane preparations of rat brain (mu), NG-108-15 neuroblastoma-glioma hybrid cells (delta), and guinea pig cerebellum (kappa). Biotinyl substitution of DYN either did not effect receptor binding (delta) or slightly reduced binding affinity (mu and kappa). Binding of B-DYN t

Multifunctional Peptide-Based Biohybrid for Targeted Reduction of Metastatic Breast Carcinoma-Associated Osteolysis.

Metastatic breast carcinoma (BC) cells are prone to spreading in the bone microenvironment, leading to a vicious cycle between local osteoclast-mediated osteolysis and tumor progression. Therefore, the targeted pharmacological down-modulation of BC cell proliferation as well as osteoclast differentiation and hyperactivity might represent a promising treatment option. We developed a multifunctional peptide nanocarrier combining bioactive EPI-X4 peptides and the Rho-inhibiting C3bot enzyme from Clostridium botulinum. C3bot is preferentially internalized into the cytosol of monocytic cells, inclu

Ultrasensitive detection of trace protein by Western blot based on POLY-quantum dot probes.

In this study, we describe an ultrasensitive quantum dots (QDs)-based Western blot. With the high affinity of avidin-functionalized POLY-QDs and simplification of the detection process, this enabled the quantitative analysis of protein by Western blotting. To prepare the POLY-QDs, CdTe quantum dots were first coated with biotinylated denatured bovine serum albumin and then, via the effect of the biotin-avidin system, the biotinylated denatured bovine serum albumin-coated QDs, which had strong fluorescence, were linked together. With this series of modifications, the fluorescence intensity of C

Generation of a human IgG3-streptavidin fusion protein. Implications for the inhibition and elimination of auto-reactive B cells.

Antibodies against self-molecules play a significant role in the development and progression of Systemic Lupus Erythematosus, as well as a number of other autoimmune disorders. Immunosuppressive drugs have been used to control this process. However, they are normally not specific to the offending cell, and can actually suppress beneficial immune responses to pathogens. In this paper a genetically engineered targeting molecule is described, which has the capacity to target antigen-specific B cells for inhibition or elimination. The targeting molecule is a fusion of streptavidin subunit to the c

Rapid and easy detection of Helicobacter pylori by in situ hybridization.

Various in situ hybridization (ISH) methods have been used to identify Helicobacter pylori, a causative organism responsible for chronic gastritis and peptic ulcer disease, but they were hard to perform and time consuming. To detect H. pylori in a rapid and easily reproducible way, we developed synthetic biotinylated oligonucleotide probes which complement rRNA of H. pylori. Formalin-fixed and paraffin-embedded tissues from 50 gastric biopsy specimens were examined. Using a serologic test and histochemical stain (Warthin-Starry silver stain and/or Giemsa stain) as a standard, 40 of them were c

A Novel Toxoplasma Inner Membrane Complex Suture-Associated Protein Regulates Suture Protein Targeting and Colocalizes with Membrane Trafficking Machinery.

The cytoskeleton of Toxoplasma gondii is composed of the inner membrane complex (IMC) and an array of underlying microtubules that provide support at the periphery of the parasite. Specific subregions of the IMC carry out distinct roles in replication, motility, and host cell invasion. Building on our previous in vivo biotinylation (BioID) experiments of the IMC, we identified here a novel protein that localizes to discrete puncta that are embedded in the parasite's cytoskeleton along the IMC sutures. Gene knockout analysis showed that loss of the protein results in defects in cytoskeletal sut…

A model study of the use of monoclonal antibodies in capture enzyme immunoassays for antigen quantification exploiting the epitope map of tick-borne encephalitis virus.

On the basis of an epitope model, capture enzyme immunoassay systems using monoclonal antibodies have been devised for the detection and quantification of Tick-borne encephalitis virus and compared with a reference system employing polyclonal sera. Monoclonal antibodies were used both as capture and detector antibodies, their suitability depending primarily on their avidity and intrinsic background activity. A considerable increase in sensitivity was achieved by combining antibodies to different non-overlapping epitopes. Biotinylation of the detector antibodies allowed the construction of mult…

Tracking and Testing Transfused Versus Endogenous Platelets with Biotin or Human Leukocyte Antigen.

Platelet transfusions are a critical and life-saving intervention utilized in bleeding patients and those at risk of bleeding. The laboratory assessment of fresh or stored platelets, both before and after transfusion, is of interest in licensing and basic science research. Platelets can be given either allogeneic (i.e., from a genetically different donor) or autologous (i.e., donor and recipient are the same, or genetically identical). In this protocol, we demonstrate how to distinguish circulating endogenous platelets from transfused platelets using biotinylation (useful for autologous and al…

ELISA-based assay for scatchard analysis of ligand-receptor interactions.

A simple, nonradioactive method is presented that can be used for performing large numbers of binding assays of cell membrane receptors with their ligands. The method adopts the simple membrane preparation and biotin-based quantitation methods of the semi-intact cell endocytosis assays. After binding of the biotinylated ligand to its receptors on the semi-intact cell membranes, a rapid centrifugation step separates the membranes from unbound ligand. Bound ligand is subsequently released by detergent, captured by a specific antibody coated on teh surface of microwells, and quantitated with pero

Biotin Ligase-Based Reporter for Detecting Protease Activity.

Protease cleavage is a crucial process, associated with cellular signaling and viral infection. Here, we present a novel sensing motif based on a proximity-dependent biotin ligase, TurboID. The sensor, Biotin Annotating Genetically Encoded Ligase-based Sensor (BAGELS), was created by engineering TurboID such that it is only active following protease activity, linking cleavage events to the biotinylation of proximal proteins. BAGELS produces a signal-to-background ratio of approximately 48 when tested with and without protease. This reporter represents the first protease activity sensor with bi…

Early diagnosis of rickettsioses by electrochemiluminscence.

The diagnosis of acute rickettsioses during the acute phase of the disease is challenging. We present preliminary evidence that antigen-capture using streptavidin-coated magnetic beads with biotinylated anti-rickettsia typhi rabbit polyclonal antibodies followed by electrochemiluminescent detection with ruthenylated antibodies of the same specificity could be used for the diagnosis of rickettsial diseases in the acute phase.

Preparation of chromosome-specific paints and complete assignment of chromosomes in the pig flow karyotype.

Sorted chromosomes from each of the 20 clusters of the male porcine bivariate flow karyotype were amplified and biotinylated using DOP-PCR. The chromosomes comprising each cluster were identified by fluorescence in situ hybridization (FISH) of the 20 probes to R-banded male pig metaphase spreads. A standard flow karyotype for the pig is presented.

A proximity ligation screen identifies SNAT2 as a novel target of the MARCH1 E3 ubiquitin ligase.

E3 ubiquitin ligases include hundreds of members that can regulate the half-life of other proteins but also modulate their cellular localization and functions. While MARCH1 appears to target principally immune cell components, such as MHC class II molecules and the co-stimulatory molecule CD86, the repertory of its targets remains to be fully documented. Here, we adapted a proximity-dependent biotin identification (BioID)-based screening approach in live HEK293 cells. We transfected a fusion protein consisting of mouse MARCH1 linked to YFP at its N-terminus and to the biotin ligase of Aquifex

Rolling Circle Amplification-Enabled Ultrasensitive Point-of-Care Test Method for Aflatoxin B1 in the Environment and Food.

Aflatoxin B1 (AFB1) contamination poses a fatal risk to human beings and urgently needs highly sensitive detection for environmental monitoring and food safety. However, the existing challenges are the unsatisfied sensitivity of the immunoassay methods and the complex matrix effect. Rolling circle amplification (RCA) is a promising method for nucleic acid isothermal amplification due to its high specificity and sensitivity. Herein, we constructed a general RCA-based point-of-care test method (RCA-POCT). With biotinylated antibodies, streptavidin, and biotinylated RCA primers, we realized the s

Real-time monitoring of voltage-responsive biomolecular binding onto electro-switchable surfaces.

Voltage-responsive biosensors capable of monitoring real-time adsorption behavior of biological analytes onto electroactive surfaces offer attractive strategies for disease detection, separations, and other adsorption-dependent analytical techniques. Adsorption of biological analytes onto electrically switchable surfaces can be modelled using neutravidin and biotin. Here, we report self-assembled monolayers formed from voltage-switchable biotinylated molecules on gold surfaces with tunable sensitivity to neutravidin in response to applied voltages. By using electrochemical quartz crystal micro

Temporal and compositional characteristics of salivary protein adsorption to hydroxyapatite.

Salivary proteins bind to enamel surfaces and hydroxyapatite in a highly selective manner. Numerous studies have identified these proteins as primarily proline-rich proteins, cystatins, statherin, and histatins. Previously, the hydroxyapatite-binding potential of these proteins had been characterized in systems consisting of singly purified protein and adsorbent. The purpose of this study was to investigate the adsorption of each protein in the presence of complete salivary secretion. Proteins, shown to adsorb to hydroxyapatite, were purified, biotinylated, and added back to the remaining prot
