Abnormal membrane protein of red blood cells in hereditary spherocytosis.

We present evidence that the hereditable hemolytic disease, hereditary spherocytosis (HS), involves an abnormality in protein of the red cell membrane. Unlike that from normal red cells, lipid-free proteins extracted from HS red cell membranes fail to increase in sedimentation rate when treated with cations; such treatment of normal membrane proteins has been shown by others to cause the formation of microfilaments. That microfilament formation might be defective in HS red cell membranes is supported by observations with vinblastine. This compound, a potent precipitant of filamentous, structur

Nucleated red blood cell in cord blood as a marker of perinatal asphyxia.

BACKGROUND: Perinatal asphyxia is a major cause of neurological morbidity and mortality in India. The purpose of this study was to investigate variations in nucleated red blood cell (NRBC) in blood associated with perinatal asphyxia and its relationship to both the severity and short term prognosis of asphyxia. METHODS: A prospective (case-control) study was undertaken at Gandhi Medical College and Associated Hospitals. A total of 100 neonates were included in the study. Levels of NRBC/100 white blood cells (WBC) and absolute NRBC counts in cord blood were compared for 50 asphyxiated (case gr…

Opposite effects of red blood cell aggregation on resistance to blood flow.

We summarized the results of two series of independent experiments recently published by our group which studied the consequences of changes in red blood cell (RBC) aggregation on resistance to blood flow in vivo. The first series compared in vivo apparent viscosity in isolated hearts perfused by either normo-aggregating or non-aggregating RBC suspension. We showed that a moderate degree of RBC aggregation induces a significant decrease in apparent relative viscosity of a suspension of RBC when compared to a non-aggregating RBC suspension. The second series studied the effects of normo vs. hyp

Two distinct pathways mediate the formation of intermediate density cells and hyperdense cells from normal density sickle red blood cells.

In sickle cell anemia (SS), some red blood cells dehydrate, forming a hyperdense (HD) cell fraction (>1.114 g/mL; mean corpuscular hemoglobin concentration [MCHC], >46 g/dL) that contains many irreversibly sickled cells (ISCs), whereas other SS red blood cells dehydrate to an intermediate density (ID; 1.090 to 1.114 g/mL; MCHC, 36 to 46 g/dL). This study asks if the potassium-chloride cotransporter (K:Cl) and the calcium-dependent potassium channel [K(Ca2+)] are participants in the formation of one or both types of dense SS red blood cells. We induced sickling by exposing normal density (ND; 1…

Red Blood Cell Piezo1 Activation Drives Faster Coagulation and Structural Alterations in Blood Clots through Red Blood Cell Deformability Impairment.

OBJECTIVE: Red blood cells (RBCs) must be highly deformable to pass through capillaries narrower than their own diameter for delivering oxygen to the tissues. Ion channels are central regulators of RBC deformability and volume. Gain-of-function mutations in Piezo1, as seen in hereditary xerocytosis, cause RBC dehydration and have been associated with an increased risk of thromboembolism. Here we investigated how pharmacological RBC-Piezo1 activation affects RBC membrane potential, RBC ion content, RBC deformability, coagulation dynamics, and clot structure. STUDY DESIGN: Blood samples were co…

Evidence for immunologic immaturity of cord blood T cells. Cord blood T cells are susceptible to tolerance induction to in vitro stimulation with a superantigen.

T cell blasts resulting from stimulating human cord blood (CB) mononuclear cells (MC) and adult peripheral blood (APB) MC with a bacterial superantigen, toxic shock syndrome toxin-1 (TSST-1), and then with human rIL-2 were investigated for their reactions to restimulation with TSST-1. Expression of TCR V beta 2, which determines the potential reactivity to TSST-1 and of CD45RO, was increased in both TSST-1-induced CB and APB T cell blasts. Most preparations of the TSST-1-induced CB T cell blasts exhibited low or no production of IL-2 and IL-4 in response to restimulation with TSST-1, while the

Sleep-dependent clearance of brain lipids by peripheral blood cells.

Sleep is viewed typically through a brain-centric lens, with little known about the role of the periphery1,2. Here we identify a sleep funct … n high oxidation and reduced NAD+, is accompanied by impaired memory and lifespan. Thus, peripheral blood cells, which we suggest are precursors of mammalian microglia, perform a daily function of sleep to maintain brain function and fitness.

Red blood cell differentiation using canine-induced pluripotent stem cells.

BACKGROUND: Red blood cell (RBC) transfusions are essential for treating various medical conditions, but global demand is difficult to meet due to a dwindling donor pool and compatibility issues. Pluripotent stem cells (PSCs) offer a promising alternative of blood dependent on volunteer donors for RBC production, and dogs serve as an excellent model for translational research due to their physiological and genetic similarities to humans. METHODS: Canine induced pluripotent stem cells (ciPSCs) were differentiated toward hematopoietic and erythroid lineages. Differentiated cells were evaluated

Flow Cytometric Examination of Galectin Binding to Red Blood Cells.

Lectins have the capacity to interact with cell surface carbohydrates, where they can influence various cellular processes. While early studies utilized lectin-induced cell agglutination to assess general binding characteristics, flow cytometry offers a more precise and quantitative method for measuring lectin interactions with specific cell populations. This technique is particularly valuable in studying galectins, a family of lectins that exhibit distinct binding preferences to red blood cells (RBCs) based on their blood type. This approach enables the determination of each lectin's binding …

Stereospecificity Membrane Impact of Two Catechins on Red Blood Cells.

Catechins are characterized by a basic structure consisting of two benzene rings and a hydropyran heterocyclic ring. In (-)-epicatechin (ECT), the substituents in C2 and C3 of the dihydropyran ring are in cis conformation, whereas in (+)-catechin (CT), they are in trans conformation. Catechins tend to interact with membrane proteins, affecting their activity and/or function and metabolic processes. In this study, the impact of CT and ECT on erythrocyte membrane and cell functions was analyzed. Surprisingly, although the two compounds have a very similar structure that differs only in the orien

Glycine receptors in circulating white blood cells regulated by neuroinflammation.

INTRODUCTION: Neuroinflammation is involved in a wide range of neurological disorders, yet the lack of minimally invasive biomarkers hampers … use models, we found that the expression of GlyRα1, α2, and α3 in macrophages and circulating white blood cells (WBCs) was not directly mediated by inflammatory cytokine signaling in the brain or WBCs. Neuroinflammation upregulates GlyRα1 and α3 expression in the brain, spleen, bone marrow, and circulating WBCs. Immunostaining revealed GlyRα3 to the membrane …

The long-term effects of chemotherapy on normal blood cells.

Several chemotherapeutic agents act by increasing DNA damage in cancer cells, triggering cell death. However, there is limited understanding of the extent and long-term consequences of collateral DNA damage in normal tissues. To investigate the impact of chemotherapy on mutation burdens and the cell population structure of normal tissue, we sequenced blood cell genomes from 23 individuals aged 3-80 years who were treated with a range of chemotherapy regimens. Substantial additional somatic mutation loads with characteristic mutational signatures were imposed by some chemotherapeutic agents, bu

On-Chip Impedance Spectroscopy of Malaria-Infected Red Blood Cells.

Malaria is a disease that affects millions of people worldwide, particularly in developing countries. The development of accurate and efficient methods for the detection of malaria-infected cells is crucial for effective disease management and control. This paper presents the electrical impedance spectroscopy (EIS) of normal and malaria-infected red blood cells. An EIS microfluidic device, comprising a microchannel and a pair of coplanar electrodes, was fabricated for single-cell measurements in a continuous manner. Based on the EIS results, the aim of this work is to discriminate Plasmodium f…
