Could immune cells be associated with nephropathy in Fabry disease patients?

BACKGROUND: In Fabry Disease (FD), although the primary factor initiating kidney damage is glycosphingolipid accumulation, secondary conditions such as increased inflammation and fibrosis may cause this damage to progress. These processes may be induced by immune cells. Therefore, we aimed to investigate the peripheral lymphocyte subgroup analysis of the patients with FD and compare these results with healthy individuals. In addition, we performed T, B, NK, and plasma cell analyses in kidney biopsy materials and compared these kidney biopsy results with the biopsy results of patients whose kid

Immune profiling of mpox survivors reveals divergent durability of antibody and T cell responses.

Despite the global spread of mpox virus (MPXV), the durability and breadth of infection-induced immunity remain incompletely defined. Here, we comprehensively characterize MPXV-specific antibody and T cell responses up to 18 months after natural infection in male individuals. Neutralizing antibodies exhibit typical acute viral kinetics, with titers peaking early and declining over time, from a mean of 328 at 12 months to 180 at 18 months post-infection. Neutralization analyses against MPXV clade Ib, clade IIb and VACV WR strains demonstrate pronounced cross-neutralization among orthopoxviruses

The Immunogenicity of an Artificial Tick-Borne Encephalitis Virus Polyepitope Immunogen May Depend on Spacers Flanking Epitope.

The development of artificial polyepitope immunogens based on the conserved CD4+ and CD8+ epitopes from different virus proteins combined into a single molecule is a promising approach in the development of vaccines for activating a specific T-cell response. Selection of spacer sequences flanking the epitopes within the construct and providing the optimal processing of such immunogens within the cell is an important step in the design of such vaccines. We report the results of designing polyepitope T-cell immunogens of tick-borne encephalitis virus that differ in the presence or absence of opt

Cellular-mediated immune responses in the liver tissue of patients with severe Plasmodium falciparum malaria.

The immune responses against Plasmodiumfalciparum malaria infections are complex and poorly understood. No published studies have yet reported the lymphocyte subsets involved in the human liver tissue of P. falciparum malaria patients. To understand the cellular-mediated immune responses in the liver during malaria infection, we determined the numbers of the various lymphocyte subsets in tissue samples obtained at autopsy from patients who died with P. falciparum malaria infection. All the liver tissue specimens had been stored at the Department of Tropical Pathology, Faculty of Tropical Medic

Defective integration of activating signals derived from the T cell receptor (TCR) and costimulatory molecules in both CD4+ and CD8+ T lymphocytes of common variable immunodeficiency (CVID) patients.

CVID is characterized by hypogammaglobulinaemia and impaired antibody production. Previous studies demonstrated defects at the T cell level. In the present study the response of purified CD4+ and CD8+ T lymphocytes to stimulation with anti-TCR monoclonal antibody (the first signal) in combination with anti-CD4 or anti-CD8, anti-CD2 and anti-CD28 MoAbs (the costimulatory signals) was investigated. Both CD4+ and CD8+ T cells from the patients showed significantly reduced IL-2 release following stimulation via TCR and costimulation via CD4 or CD8 and CD2, respectively. However, normal IL-2 produc

The evolving biology of T cell memory: A dynamic continuum of function and persistence.

Immunological memory underpins long-term protection and vaccination. Although early work established the durability and recall capacity of memory T cells, understanding of CD8 T cell memory generation, maintenance, and plasticity has advanced substantially over recent decades. Initial models viewed memory as an outcome of peak effector responses and were defined mainly by persistence. Successive waves of technological innovation, from MHC tetramers and genetic lineage-tracing approaches to single-cell and epigenomic profiling and modern high-throughput gene-perturbation screens, have reshaped

Origins of CD8 tissue resident memory.

The concept that immune cells, specifically T cells, become established and reside in peripheral tissues, especially at barrier sites, has been around for several decades. Recent studies have revealed substantial heterogeneity at the cellular level that depends on the type of immune response and tissue environment, among many other factors. Nevertheless, some features are common to most resident memory T cells, especially CD8+ cells. These include certain surface receptors and transcription factors. It is important to understand how tissue-resident memory (TRM) T cells develop and function, as

CD8+ T cell exhaustion in anti-tumour immunity: The new insights for cancer immunotherapy.

CD8+ T cells play a crucial role in anti-tumour immunity, but they often undergo exhaustion, which affects the anti-tumour activity of CD8+ T cells. The effect and mechanism of exhausted CD8+ T cells have become the focus of anti-tumour immunity research. Recently, a large number of studies have confirmed that long-term antigen exposure can induce exhaustion. Cytokines previously have identified their effects (such as IL-2 and IL-10) may play a dual role in the exhaustion process of CD8+ T cells, suggesting a new mechanism of inducing exhaustion. This review just focuses our current understand

Instructing Memory in CD8+ Thymocytes.

T cells can sometimes acquire properties of a memory cell without encountering foreign antigen. Miller et al. now show that development of such 'memory phenotype' (MP) CD8+ T cells starts with recognition of self-ligands in the thymus, and finishes in peripheral tissues. These cells respond rapidly during infection, and infiltrate tumors, where they express high amounts of PD-1.

Antiviral CD4 and CD8 T-cell memory: differences in the size of the response and activation requirements.

Following acute lymphocytic choriomeningitis virus (LCMV) infection, there is a potent antiviral CD8 T-cell response that eliminates the infection. This initial CD8 T-cell response is followed by a period of memory during which elevated numbers of virus-specific CD8 T cells remain in the mouse. CD4 T cells are also activated after LCMV infection, but relatively less is known about the magnitude and duration of the CD4 response. In this study, we used intracellular staining for interferon-gamma to measure both CD4 and CD8 responses in the same mice at the single cell level. After LCMV infection

Social stressors associated with age-related T lymphocyte percentages in older US adults: Evidence from the US Health and Retirement Study.

Exposure to stress is a risk factor for poor health and accelerated aging. Immune aging, including declines in naïve and increases in terminally differentiated T cells, plays a role in immune health and tissue specific aging, and may contribute to elevated risk for poor health among those who experience high psychosocial stress. Past data have been limited in estimating the contribution of life stress to the development of accelerated immune aging and investigating mediators such as lifestyle and cytomegalovirus (CMV) infection. This study utilizes a national sample of 5,744 US adults over age

Rheumatoid papules in a patient with acquired immune deficiency syndrome and symmetric polyarthritis.

We describe a patient with acquired immunodeficiency syndrome (AIDS) and an unusual rheumatoid factor-positive symmetric inflammatory polyarthritis who met all 7 American College of Rheumatology criteria for RA, but who also had many features suggestive of seronegative inflammatory arthritis. Although cutaneous vasculitis has also been described in human immunodeficiency virus infection, this is the first report of cutaneous extravascular necrobiotic granuloma (rheumatoid papule) in AIDS. This case highlights the immunopathogenesis of inflammatory arthritis in AIDS and illustrates the difficul

T cell dysregulation reflects disease stage in hepatitis virus and alcohol-related liver disease.

Liver cirrhosis is characterized by both immunodeficiency and an exaggerated immune response leading to systemic inflammation. In this study, we investigated the role of T cells in different stages of liver disease and therefore analyzed 72 patients stratified into those with and without cirrhosis and compensated and decompensated cirrhosis. Flow cytometry revealed that CD8+ T cells, but not CD4+ T cells were diminished in patients with decompensated liver cirrhosis and this further resulted in an increased CD4/CD8 T cell ratio in those patients. In addition, the phenotype of CD4+ and CD8+ T c

Immunodominant tuberculosis CD8 antigens preferentially restricted by HLA-B.

CD8(+) T cells are essential for host defense to intracellular bacterial pathogens such as Mycobacterium tuberculosis (Mtb), Salmonella species, and Listeria monocytogenes, yet the repertoire and dominance pattern of human CD8 antigens for these pathogens remains poorly characterized. Tuberculosis (TB), the disease caused by Mtb infection, remains one of the leading causes of infectious morbidity and mortality worldwide and is the most frequent opportunistic infection in individuals with HIV/AIDS. Therefore, we undertook this study to define immunodominant CD8 Mtb antigens. First, using IFN-ga

T lymphocyte activation in acute severe asthma.

T lymphocyte subsets and lymphocyte activation markers in the peripheral blood were assessed in patients admitted to hospital with acute severe asthma (status asthmaticus). Measurements were made on admission, day 3, and day 7 (or on discharge from hospital if this was sooner). The results were compared with 3 control groups (mild asthma, chronic obstructive airways disease, and normal individuals). The percentages of CD4-positive and CD8-positive T lymphocytes, and the CD4/CD8 ratios, were similar in the patients with acute severe asthma and the control groups, and were within the normal rang

T lymphocytes (CD3) may participate in the recurrence of cervical intraepithelial neoplasia grade III.

OBJECTIVE: Data from the literature demonstrate that the local and systemic immune responses seem to play an important role in the progression of cervical intraepithelial neoplasia (CIN). Our aim was to investigate whether recurrences among CIN III patients might be related to the presence of local lymphocytes, macrophage and enzyme iNOS. METHODS: We analyzed 35 patients with CIN III who underwent conization and followed up for a minimum of 4 years. Using immunohistochemistry, the presence of T lymphocytes (CD3, CD8 and CD45RO), B lymphocytes (CD20), macrophages (CD68) and the expression of t
